Cockayne syndrome group A (CS-A) is a rare recessive progeroid disorder characterized by sun sensitivity and neurodevelopmental abnormalities. Cells derived from CS-A patients present as pathological hallmarks excessive oxidative stress, mitochondrial fragmentation and apoptosis associated with hyperactivation of the mitochondrial fission dynamin related protein 1 (DRP1). In this study, by using human cell models we further investigated the interplay between DRP1 and CSA and we determined whether pharmacological or genetic inhibition of DRP1 affects disease progression. Both reactive oxygen and nitrogen species are in excess in CS-A cells and when the mitochondrial translocation of DRP1 is inhibited a reduction of these species is observed together with a recovery of mitochondrial integrity and a significant decrease of apoptosis. This study indicates that the CSA-driven modulation of DRP1 pathway is key to control mitochondrial homeostasis and apoptosis and suggests DRP1 as a potential target in the treatment of CS patients. 相似文献

Foods based on sweet lupin proteins are gaining attention from industry and consumers because of their possible role in the prevention of cardiovascular disease. When promoting lupin-based foods for inclusion in a daily diet, the thermal damage suffered during processing is of relevance to the bioactive and nutritional quality of the food product. N-(2-furoylmethyl)-L-lysine (furosine) quantification demonstrates that currently available sweet lupin protein isolates have a thermal damage comparable to or lower than other traditional food ingredients, and are a good source of lysine in non-dairy products. In lupin-based foods claiming to have cholesterol-lowering potential, shotgun proteomics offers itself as a fast and effective screening method for assessing the biological availability of active peptides. Since its starting in the 19th century, modern food packaging has made great advances as results of global trends and consumer preferences. These advances are oriented to obtain improved food quality and safety. Moreover, with the move toward globalization, food packaging requires also longer shelf life, along with the monitoring of safety and quality based upon international standards. Nanotechnology can address all these requirements and extend and implement the principal packaging functions - containment, protection and preservation, marketing and communications. Applications of polymer nanotechnology in fact can provide new food packaging materials with improved mechanical, barrier and antimicrobial properties, together with nano-sensors for tracing and monitoring the condition of food during transport and storage.The latest innovations in food packaging, using improved, active and smart nanotechnology will be analyzed. We investigated the efficacy of tigecycline and FS8, alone or combined, in preventing prosthesis biofilm in a rat model of staphylococcal vascular graft infection. Graft infections were established in the back subcutaneous tissue of adult male Wistar rats by implantation of Dacron prostheses followed by topical inoculation with 2 × 107 colony-forming units of Staphylococcus aureus, strain Smith diffuse. The study included a control group, a contaminated group that did not receive any antibiotic prophylaxis, and three contaminated groups that received: (i) intraperitoneal tigecycline, (ii) FS8-soaked graft, and (iii) tigecycline plus FS8-soaked graft, respectively. Each group included 15 animals. The infection burden was evaluated by using sonication and quantitative agar culture. Moreover, an in vitro binding-study was performed to quantify the how much FS8 was coated to the surface of the prosthesis. Tigecycline, combined with FS8, against the adherent bacteria showed MICs (2.00 mg/L) and MBCs (4.00 mg/L) four-fold lower with respect to tigecycline alone in in vitro studies. The rat groups treated with tigecycline showed the lowest bacterial numbers (4.4 × 104 ± 1.2 × 104 CFU/mL). The FS8-treated group showed a good activity and significant differences compared to control group with bacterial numbers of 6.8 × 104 ± 2.0 × 104 CFU/mL. A stronger inhibition of bacterial growth was observed in rats treated with a combined FS8 and tigecycline therapy than in those that were singly treated with bacterial numbers of 101 CFU/mL graft. In conclusion, the ability to affect biofilm formation as well, its property to be an antibiotic enhancer suggests FS8 as alternative or additional agent to use in conjunction with conventional antimicrobial for prevention of staphylococcal biofilm related infection. 相似文献

The ethanol steam reforming reaction carried out in a Pd-based tubular membrane reactor has been modelled via a finite element code. The model considers the membrane tube divided into finite volume elements where the mass balances for both lumen and shell sides are carried out accordingly to the reaction and permeation kinetics. Especially, a simplified “power law” has been applied for the reaction kinetics: the comparison with experimental data obtained by using three different kinds of catalyst (Ru, Pt and Ni based) permitted defining the coefficients of the kinetics expression as well as to validate the model. Pseudostatic approaches for the seismic analysis of pile foundations are attractive for practicing engineers because they are simple when compared to difficult and more complex dynamic analyses. To evaluate the internal response of piles subjected to earthquake loading, a simplified approach based on the “p-y” subgrade reaction method has been developed. The method involves two main steps: first, a site response analysis is carried out to obtain the free-field ground displacements along the pile. Next, a static load analysis is carried out for the pile, subjected to the computed free-field ground displacements and the static loading at the pile head. A pseudostatic push over analysis is adopted to simulate the behavior of piles subjected to both lateral soil movements and static loadings at the pile head. The single pile or the pile group interact with the surrounding soil by means of hyperbolic p-y curves. The solution derived first for the single pile, was extended to the case of a pile group by empirical multipliers, which account for reduced resistance and stiffness due to pile-soil-pile interaction. Numerical results obtained by the proposed simplified approach were compared with experimental and numerical results reported in literature. It has been shown that this procedure can be used successfully for determining the response of a pile foundation to “inertial” loading caused by the lateral forces imposed on the superstructure and “kinematic” loading caused by the ground movements developed during an earthquake. 相似文献

Traumatic brain injury (TBI) frequently affects both the basic and the superordinate components of attention; deficits vary according to patient age. This study evaluated the efficacy of a specific remediation intervention for attention. Sixty-five TBI patients (aged 6?18 years) with attention deficit were assessed at baseline and at 1-year follow-up: 40 patients received attention-specific neuropsychological training for 6 months, and the control group comprised 25 patients. Cognitive assessment included a Wechsler Intelligence Scale (e.g., A. Orsini, 1993) and the Continuous Performance Test II (CPT II; C. K. Conners, 2000). The Vineland Adaptive Behavior Scales (VABS; S. Sparrow, D. Balla & D. V. Cicchetti, 1984) was administered to assess the treatment's ecological validity. At baseline, all patients presented with a mild intellectual disability and pathological scores on the CPT II. At follow-up, significant differences were found between the 2 groups on the CPT II and VABS: The clinical group improved more than the control group. Specific remediation training for attention, including a combination of a process-specific approach and metacognitive strategies, significantly improved attention performance. Radon (222Rn) concentration was evaluated in shops of the Savona Province, Italy, between summer 2002 and winter 2002-2003. The main characteristics of each shops were recorded through a questionnaire investigating the ventilation rate and factors related to 222Rn precursors in the soil and the construction materials. The main variables that were related to radon concentration were the following: age of the building, level of the shop above ground, season of the year, wind exposure, active windows, and type of heating system. Shops equipped with individual air heating/conditioning systems exhibited radon concentrations that were three times higher than those of shops heated by centralized furnaces. Our data indicate that the level of pollution in the shops was of medium level, with an expected low impact on the salespersons' health. Only in wintertime, the action level of 200 Bq m(-3) for the confined environment was reached in 10 shops equipped with individual air heating/conditioning systems. 相似文献
