Ideomotor apraxia, disordered movement execution to command, commonly follows left-hemisphere damage, implying left-hemisphere dominance for certain kinds of movements. To delineate this dominance we used different command modalities to elicit meaningful movements and tested imitation of nonsense movements. Twenty-seven patients with unilateral hemispheric stroke and 10 age-matched controls were evaluated. Patients with left-hemisphere damage performed both meaningful and nonsense movements poorer than the other study groups; thus, the meaningfulness of the movements is irrelevant for the left-hemisphere motor dominance. The performance varied, however, with the command modality and movement type. Based on this and earlier studies we posit that the left-hemisphere motor dominance is determined by the artificiality of the test situation (it concerns movements performed to command and out of the natural context) and increased spatial and temporal complexity of the demanded movements. No association between the lesion locus within the left hemisphere and the severity of the ideomotor apraxia was found.  相似文献

The fibrous sheath is a unique cytoskeletal structure in the sperm flagellum believed to modulate sperm motility. FSC1 is the major structural protein of the fibrous sheath. The yeast two-hybrid system was used to identify other proteins that contribute to the structure of the fibrous sheath or participate in sperm motility. When FSC1 was used as the bait to screen a mouse testis cDNA library, two clones were isolated encoding the type Ialpha regulatory subunit (RIalpha) of cAMP-dependent protein kinase. Deletion analysis using the yeast two-hybrid system and in vitro binding assays with glutathione S-transferase-FSC1 fusion proteins identified two RIalpha tethering domains on FSC1. A domain located at residues 219-232 (termed domain A) corresponds to the reported tethering domain for a type II regulatory subunit (RII) of cAMP-dependent protein kinase, indicating that this binding domain has dual specificity to RI and RII. Another RIalpha tethering site (termed domain B) at residues 335-344 shows specific binding of RIalpha and had no significant sequence homology with known RII tethering domains. However, helical wheel projection analysis indicates that domain B is likely to form an amphipathic helix, the secondary structure of RII tethering domains of protein kinase A anchoring proteins. This was supported by the finding that site-directed mutagenesis to disrupt the amphipathic helix eliminated RIalpha binding. This is apparently the first report of an RIalpha-specific protein kinase A anchoring protein tethering domain.  相似文献

Outlines a psychological procedure aimed at improving the working relationship between a newly hired executive and the executive's reporting official. The "Executive Fit Rehearsal" (EFR) attempts to systematically engage the executive and reporting official in a dialogue designed to anticipate and overcome problems resulting from individual differences in personality, expectations, work styles, and communication patterns. The procedure supports consensus and mutual understanding. To assess the effectiveness of the procedure, evaluation forms were mailed to participants of 15 consecutive Executive Fit sessions. Ss' reactions to the EFR indicate that the process facilitated adjustment to the work setting and improved communications. By 2? years of age, human infants appear to understand how others are connected to the external world through the mental state of attention and also appear to understand the specific role that the eyes play in deploying this attention. Previous research with chimpanzees suggests that, although they track the gaze of others, they may simultaneously be unaware of the underlying state of attention behind gaze. In a series of 3 experiments, the investigators systematically explored how the presence of eyes, direct eye contact, and head orientation and movement affected young chimpanzees' choice of 2 experimenters from whom to request food. The results indicate that young chimpanzees may be selectively attracted to other organisms making direct eye contact with them or engaged in postures or movements that indicate attention, even though they may not appreciate the underlying mentalistic significance of these behaviors. Nanowire TiO2 thin films were successfully prepared on Ti metal substrates by hydrothermal treatment of calcined Ti foils in 10 M NaOH. The nanowire TiO2 thin films exhibited much larger surface area and higher photoelectrochemical performance than the TiO2 thin films prepared on Ti metal substrates by the calcination of Ti foil. These nanowire films were shown to act as an efficient photoanodes for the photoelectrochemical water splitting reaction.  相似文献

Corrosion experiments were performed on two metallic substrates, a ferritic–pearlitic steel (P285NH) and a ferritic one (Armco), in silicate environment during 30 days at 120°C. Corrosion products were characterized in terms of morphology (scanning electron microscope and transmission electron microscopy), composition (energydispersive X-ray spectroscopy) and structure (µ-Raman, selected area electron diffraction, X-ray absorption near edge structure). Results show a nanometric inner layer made of compact and adherent nanocrystallized magnetite with, locally a thickness up to several micrometers due to the metal microstructure. An outer layer of Fe-rich phyllosilicate, smectite and serpentine, more porous than the inner one and poorly adherent is observed around both the samples.  相似文献

YoungCinchona ledgeriana plants contain two types of alkaloid: indole alkaloids in the leaves and quinoline alkaloids in the root. FromCinchona leaves, a crude alkaloid extract was made, containing the cinchophylline type of indole alkaloids and a small amount of 5-methoxytryptamine. The leaf alkaloid extract exerted a strong detrimental effect on the growth of larvae of the polyphagous beet armyworm,Spodoptera exigua (Lepidoptera). Feeding of larvae on an artificial diet containing the leaf alkaloids at the same concentrations as those found in the plant resulted in significant growth reduction, retardation in development, and mortality of the larvae. Cinchophyllines are composed of 5-methoxytryptamine coupled to a corynantheal unit. When incorporated into the artificial diet, 5-methoxytryptamine alone had no effect on the 5.exigua larvae. Corynantheal, however, had a strong detrimental effect on growth of the larvae, its effect being comparable to that of the leaf alkaloid extract. In contrast to the indole-type leaf alkaloids, the quinolinetypeCinchona root alkaloids did not affect growth and development of the larvae. These results suggest that the indole-type alkaloids, which inCinchona plants are present at the highest concentrations in the young, vulnerable leaflets, are involved in the chemical defense of the plant against herbivorous insects.  相似文献

SETD7 is a histone H3K4 lysine methyltransferase involved in human gene regulation. Aberrant expression of SETD7 has been associated with various diseases, including cancer. Therefore, SETD7 is considered a good target for the development of new epigenetic drugs. To date, few selective small‐molecule inhibitors have been reported that target SETD7, the most potent being (R)‐PFI‐2. Herein we report structure–activity relationship studies on (R)‐PFI‐2 and its analogues. A library of 29 structural analogues of (R)‐PFI‐2 was synthesized and evaluated for inhibition of recombinantly expressed human SETD7. The key interactions were found to be a salt bridge and a hydrogen bond formed between (R)‐PFI‐2′s NH2+ group and SETD7′s Asp256 and His252 residue, respectively.  相似文献

We examined the effects of water and nutrient availability on the expression of the defense pyrrolizidine alkaloids (PAs) in Senecio jacobaea and S. aquaticus. Senecio jacobaea, and S. aquaticus are adapted to different natural habitats, characterized by differing abiotic conditions and different selection pressures from natural enemies. We tested if PA concentration and diversity are plastic over a range of water and nutrient treatments, and also whether such plasticity is dependent on plant species. We also tested the hypothesis that hybridization may contribute to PA diversity within plants, by comparing PA expression in parental species to that in artificially generated F1 hybrids, and also in later generation natural hybrids between S. jacobaea and S. aquaticus. We showed that total PA concentration in roots and shoots is not dependent on species, but that species determines the pattern of PA diversification. Pyrrolizidine alkaloid diversity and concentration are both dependent on environmental factors. Hybrids produce a putatively novel PA, and this PA is conserved in natural hybrids, that are backcrossed to S. jacobaea. Natural hybrids that are backcrossed several times to S. jacobaea are with regard to PA diversity significantly different from S. jacobaea but not from S. aquaticus, while F1 hybrids are in all cases more similar to S. jacobaea. These results collectively suggest that PA diversity is under the influence of natural selection.  相似文献
