The ability to successfully target the delivery of drugs and other therapeutic molecules has been a key goal of biomedical research for many decades. Despite highly promising in vitro results, however, successful translation of targeted drug delivery into clinical use has been extremely limited. This study investigates the significance of the characteristics of whole blood, which are rarely accounted for in vitro assays, as a possible explanation for the poor correlation between in vitro and in vivo experiments. It is shown using two separate model systems employing either biochemical or magnetic targeting that blood causes a substantial reduction in targeting efficiency relative to saline under the same flow conditions. This finding has important implications for the design of targeted drug delivery systems and the assays used in their development.  相似文献

Electronic commerce is an integral part of most businesses today. The perceived quality of the website and whether users intend to reuse the site are of primary concern to online businesses. There are various factors that influence customers' intentions to reuse a site. In this paper, we investigate two of these factors: whether the site is familiar to the user and whether there are online advertisements on the website. The results indicate that familiarity and online ads do have an impact on quality and reuse, although it is not entirely as expected.  相似文献

The modularity of the sentence processor, or lack thereof, remains a much-debated issue in psycholinguistics. The authors present evidence from a semantically impaired patient (DM) that bears on this issue. As demonstrated elsewhere (S. D. Breedin, E. M. Saffran, & H. B. Coslett, see record 1996-90000-001), DM suffered a significant loss of semantic knowledge. Here, the authors show that this impairment did not compromise DM's ability to process syntactic information. DM performed well on grammaticality judgment tasks and on sentence comprehension tasks that required the use of syntactic information for the assignment of thematic roles. The resistance of syntactic operations to semantic loss would seem to pose a challenge for models in which "the syntactic and conceptual aspects of processing are … inextricably intertwined" (J. L. We report use of a novel hyperspectral imaging system utilizing digital light processing (DLP) technology to noninvasively visualize in vivo tissue oxygenation during surgical procedures. The system's novelty resides in its method of illuminating tissue with precisely predetermined continuous complex spectra. The Texas Instruments digital micromirror device, DMD, chip consisting of 768 by 1024 mirrors, each 16 μm square, can be switched between two positions at 12.5 kHz. Switching the appropriate mirrors controls the intensity of light illuminating the tissue as a function of wavelength, active spectral illumination. Meaning, the tissue can be illuminated with a different spectrum of light within 80 μs. Precisely, predetermined spectral illumination penetrates into patient tissue, its chemical composition augments the spectral properties of the light, and its reflected spectra are detected and digitized at each pixel detector of a silicon charge-coupled device, CCD. Using complex spectral illumination, digital signal processing and chemometric methods produce chemically relevant images at near video rates. Specific to this work, tissue is illuminated spectrally with light spanning the visible electromagnetic spectrum (380 to 780 nm). Spectrophotometric images are detected and processed visualizing the percentage of oxyhemoglobin at each pixel detector and presented continuously, in real time, at 3 images per second. As a proof of principle application, kidneys of four live anesthetized pigs were imaged before, during, and after renal vascular occlusion. DLP Hyperspectral Imaging with active spectral illumination detected a 64.73 ± 1.5% drop in the oxygenation of hemoglobin within 30 s of renal arterial occlusion. Producing chemically encoded images at near video rate, time-resolved hyperspectral imaging facilitates monitoring renal blood flow during animal surgery and holds considerable promise for doing the same during human surgical interventions.  相似文献

The use of primary cells in human liver therapy is limited by a lack of cells. Induced pluripotent stem cells (iPSCs) represent an alternative to primary cells as they are infinitely expandable and can be differentiated into different liver cell types. The aim of our work was to demonstrate that simian iPSCs (siPSCs) could be used as a new source of liver cells to be used as a large animal model for preclinical studies. We first differentiated siPSCs into a homogenous population of hepatoblasts (siHBs). We then separately differentiated them into hepatocytes (siHeps) and cholangiocytes (siChols) expressing respective specific markers and displaying epithelial polarity. Moreover, we showed that polarized siChols can self-organize into 3D structures. These results should facilitate the deciphering of liver development and open the way to exploring co-culture systems that could be assessed during preclinical studies, including in autologous monkey donors, for regenerative medicine purposes.  相似文献

Building designers are increasingly relying on complex fenestration systems (CFS) to reduce energy consumed for lighting and HVAC in low-energy buildings. Radiance, a lighting simulation program, has been used to conduct daylighting simulations for CFS. Depending on the configurations, the simulation can take hours or even days using a personal computer. This paper describes how to accelerate the matrix multiplication portion of a Radiance three-phase daylight simulation by conducting parallel computing on heterogeneous hardware of a personal computer. The algorithm was optimized and the computational part was implemented in parallel using OpenCL. The speed of the new approach was evaluated using various daylighting simulation cases on a multi-core central processing unit (CPU) and a graphics processing unit (GPU). Based on the measurements and analysis of the time usage for the Radiance daylighting simulation, further speedups can be achieved using fast input/output devices and storing the data in a binary format.  相似文献

Dynamic modelling was used to quantify the impact of projected climate change, and potential changes in population and land use, on phosphorus (P) export from a sub-catchment in SW Ireland using the Generalised Watershed Loading Functions (GWLF) model. Overall the results indicated that the increase in annual total phosphorus loads attributable to climate change was greater than that from either population or land use change, and therefore that future climate variability will pose an increasingly significant threat to the successful long-term implementation of catchment management initiatives. The seasonal pattern in projected P export mirrored changes in streamflow, with higher rates between January and April and lower rates in summer. The potential reduction in export in summer was, however, negated when increases in population were included in simulations. A change in the slurry spreading period from that stipulated in national regulations to the months between April and September could potentially mitigate against future increases in dissolved P export in spring. The results indicate that projected changes in climate should be included when undertaking modelling exercises in support of decision making for catchment management plans.  相似文献

A novel V-junction microfluidic (VJM) device has been used to generate several types of particles from the break-up of bubbles. The flow rates of selected solutions and the gas pressure were adjusted in order to successfully generate monodisperse polymer coated microbubbles, which serve as a platform for particle generation. Uniform particles in both the nano and micro size ranges with different shapes were spontaneously generated from bubbles and have the potential to be used in several advanced technological applications.  相似文献

This paper describes a new in vivo Raman probe that allows investigation of areas of the body that are otherwise difficult to access. It is coupled to a previously described commercially available in vivo Raman spectrometer that samples the skin through an optical flat. In the work presented here, the laser light emerges from a smaller pen-shaped probe. It thus works on the same principles as the original spectrometer, while its relative performance in terms of signal-to-noise ratio of the spectra and obtained spatial resolution is only slightly diminished. It allows the window to be placed against the subject in more curved and recessed areas of subject's body and also for them to be more comfortable while the measurements take place. Results from three areas of the body that have previously been very difficult to study are described, the mouth, axilla, and scalp. Results from the scalp and axilla strata cornea (SC) show significant differences from the "normal" SC of the volar forearm. For instance, the scalp is observed to have lower amounts of natural moisturizing factors (NMF) compared to the volar forearm within the same subjects. Also for both the axilla and scalp the lipids show a change in order as compared to the lipids in the volar forearm and also differences from each other. The potential significance of these observations is discussed. Further, we show how we can probe the mouth, in this case observing the presence of the astringent tea polyphenol epigallocatechin gallate within the oral mucosa.  相似文献
