Syndecans act as independent co-receptors to exert biological activities and their altered function is associated with many pathophysiological conditions. Here, syndecan-1 and -4 were examined in lesional skin of patients with psoriasis. Immunohistochemical staining confirmed altered syndecan-1 distribution and revealed absence of syndecan-4 expression in the epidermis. Fibronectin (FN)—known to influence inflammation and keratinocyte hyperproliferation via α5β1 integrin in psoriasis—was also decreased. Syndecan-1 and -4 expression was analyzed in freshly isolated lesional psoriatic human keratinocytes (PHK) characterized based on their proliferation and differentiation properties. mRNA levels of syndecan-1 were similar between healthy and PHK, while syndecan-4 was significantly decreased. Cell growth and release of the pro-inflammatory Tumor Necrosis Factor-alpha (TNFα) were selectively and significantly induced in PHKs plated on FN. Results from co-culture of healthy keratinocytes and psoriatic fibroblasts led to the speculation that at least one factor released by fibroblasts down-regulate syndecan-1 expression in PHK plated on FN. To assay if biological treatments for psoriasis target keratinocyte proliferation, gelatin-based patches enriched with inteleukin (IL)-17α or TNFα blockers were prepared and tested using a full-thickness healthy epidermal model (Phenion®). Immunohistochemistry analysis showed that both blockers impacted the localisation of syndecan-1 within the refined epidermis. These results provide evidence that syndecans expression are modified in psoriasis, suggesting that they may represent markers of interest in this pathology. 相似文献

Cancer is a set of diseases characterized by several hallmark properties, such as increased angiogenesis, proliferation, invasion, and metastasis. The increased angiogenic activity constantly supplies the tumors with nutrients and a plethora of cytokines to ensure cell survival. Along these cytokines is a newly discovered protein, called irisin, which is released into the circulation after physical exercise. Irisin is the product of fibronectin type III domain-containing protein 5 (FNDC5) proteolytic cleavage. Recently it has been the topic of investigation in several types of cancer. In this study, we conducted a systematic review and meta-analysis to investigate its implication in different types of cancer. Our results suggest that irisin expression is decreased in cancer patients, thus it can be used as a valid biomarker for the diagnosis of several types of cancer. In addition, our results indicate that irisin may have an important role in tumor progression and metastasis since it is involved in multiple signaling pathways that promote cell proliferation and migration. 相似文献

Mediterranean dried fruits that are cultivated and produced in Greece; that is, Corinthian currants, figs, prunes, cherries, apricots and peaches were evaluated in terms of total polar phenols, individual simple phenolics, anthocyanins and antioxidant capacity in vitro. The potential release of dried fruit polar phenols among the different fractions of an in vitro digestion model was also determined. Total polar phenol, flavanol, flavone/flavonol content and antioxidant capacity was in the range 86–551 mg GAE/100g, 0.2–57 mg CE/100g, 9–71 mg RE/100g and 6–47 mg AAE/100g, respectively. A 12–82% release of total phenolics was observed post-mastication, which further increased post-gastric digestion. The same trend was also followed in the case of total flavanols and flavones/flavonols. Total polar phenols and flavones/flavonols were found to enter the simulated epithelial cell wall. Simple polar phenolics and anthocyanins were identified and quantified in all dried fruit extracts and in some of the digestion fractions. 相似文献

Cardiovascular disease (CVD) is the major cause of death in patients with type-2 diabetes mellitus (T2DM), although the factors that accelerate atherosclerosis in these patients are poorly understood. The identification of the altered quantity and quality of lipoproteins, closely related to atherogenesis, is limited in routine to a pattern of high triglycerides and low HDL-cholesterol (HDL-C) and in research as dysfunctional HDLs. We used the emerging NMR-based lipidomic technology to investigate compositional features of the HDLs of healthy individuals with normal coronary arteries, drug-naïve; recently diagnosed T2DM patients with normal coronary arteries; and patients with recent acute coronary syndrome. Patients with T2DM and normal serum lipid profiles even at diagnosis presented significant lipid alterations in HDL, characterized by higher triglycerides, lysophosphatidylcholine and saturated fatty acids; and lower cholesterol, phosphatidylcholine, phosphatidylethanolamine, sphingomyelin, plasmalogens and polyunsaturated fatty acids, an atherogenic pattern that may be involved in the pathogenesis of atherosclerosis. These changes are qualitatively similar to those found, more profoundly, in normolipidemic patients with established Coronary Heart Disease (CHD). We also conclude that NMR-based lipidomics offer a novel holistic exploratory approach for identifying and quantifying lipid species in biological matrixes in physiological processes and disease states or in disease biomarker discovery. 相似文献

Abstract In this work, in order to describe the impact of ozone on ferric oxide – a common constituent of building materials – in purely scientific terms, a novel methodology that of Reversed Flow–Inverse Gas Chromatography (RF–IGC or RF–GC) is proposed. Five important physicochemical quantities concerning adsorption of ozone on the solid surface of ferric oxide are determined, in five temperatures, in a time-resolved way. By means of a simple PC-program, the values of local adsorption energy, local adsorption isotherm, local monolayer capacity, probability density function for adsorption energy, and energy from lateral interactions are calculated. Thus, the role of surface heterogeneity, lateral interactions and different distributions of active sites (surface topography) on the adsorption of ozone on the surface of ferric oxide is analyzed in the way the RF–IGC method has it developed. 相似文献

Emergent behaviors occur when simple interactions between a system's constituent elements produce properties that the individual elements do not exhibit in isolation. This article reports tunable emergent behaviors observed in domain wall (DW) populations of arrays of interconnected magnetic ring-shaped nanowires under an applied rotating magnetic field. DWs interact stochastically at ring junctions to create mechanisms of DW population loss and gain. These combine to give a dynamic, field-dependent equilibrium DW population that is a robust and emergent property of the array, despite highly varied local magnetic configurations. The magnetic ring arrays’ properties (e.g., non-linear behavior, “fading memory” to changes in field, fabrication repeatability, and scalability) suggest they are an interesting candidate system for realizing reservoir computing (RC), a form of neuromorphic computing, in hardware. Although a milestone in the history of Latin American photography, the work of Horacio Coppola is little known outside Argentina. His unorthodox angles and tilting frames, his attention to industrial and urban themes, and his experimentation with different lenses in the period between 1927 and 1931 were in absolute synchrony with the precepts of European New Vision and New Objectivity photography, and with American straight photography. That Coppola’s visual idiom was in tune with the latest photographic developments in interwar Europe is obvious. What remains unclear is how exactly he achieved that synchrony. Several scholars have wrestled with this question, which nevertheless remains elusive due to lack of evidence of direct contact with European photography prior to his studies at the Berlin Bauhaus in 1932 and his repeated attempts to downplay the influence contemporary photographers had on his work. The article provides a systematic study of Coppola’s aesthetic genealogy and his photographic borrowings through a critical examination of his early pictures and theoretical writings. It teases out possible direct and indirect channels of dialogue with the New Vision, New Objectivity, and straight photography in the second half of the 1920s, and probes the reasons behind Coppola’s reluctance to admit a connection with contemporary European photography in particular. 相似文献
