Legislation and the motor car

Vehicle legislation for safety and emission control is led by the USA where half the world's cars and one‐third of the world's road deaths are found. The 1975/6 standards will cause confrontations between industry and government in the U.S.A. and have costly implications for the American people. The rest of the world is affected by what happens in the U.S.A. but for language and other reasons fragmented laws are normal with conflicting requirements for manufacturers who supply the many markets. The greatest force for harmonization of legislation may be the enlarged Common Market including, as it does, the four major European Car manufacturing countries. The Common Market Commission may displace WP 29 of the United Nations as the leading law‐making body. Type approval is the compliance system used in Europe, but the growth in scope of safety and other legislation is bringing into focus a weakness of the system which is reducing production efficiency to an alarming degree. American style self‐certification is not acceptable to European governments.     

Untersuchungen zur Sorptionsreversibilität von organischen Schadstoffen in Aktivkohle, Holzkohle und Zeolith Y-200

Numerous studies have shown that sorption of organic contaminants in soils is dominated by the natural organic carbon content (C _org) of the soil. However, it is still under discussion whether sorption processes are fully reversible or whether an irreversibly sorbed contaminant fraction remains in the soil. This is especially important when considering soil remediation measures and its targets. In multi-stage sorption-desorption batch experiments with TCE, PCE, ortho-xylene and para-xylene and with the sorbents activated carbon, charcoal and a hydrophobic zeolite Y-200, the reversibility of sorption was studied. It could be shown that the structural features of the sorbents are of ample importance for the occurrence of a desorption-resistant fraction. While sorption was mainly reversible for the micro-porous zeolite Y-200 with a rigid pore network, charcoal and the activated carbon showed significant desorption hysteresis. However, following a subsequent sorption step, this fraction eventually desorbs and is re-mobilized.     

Entwicklung eines optisch zugänglichen Reaktors zur Thermographiemessung in einer Katalysatorschüttung

To measure the microkinetics in heterogeneously catalyzed strongly exothermic gas phase reaction heat and mass transfer limitations should be excluded. Also an unequal distribution of temperature in the catalyst fixed bed, like a hot spot, distorts the results of microkinetic investigations. A promising approach to measure the temperature contactless is thermography. For this work, an optically accessible reactor is developed to measure the temperature in a catalytically powder fixed bed. In the scope of this work, this technique is used to investigate the temperature development and distribution during the exothermal CO₂ methanation depending on the bed and gas dilution and the reactor performance.     

Identification and Structure–Activity Relationship Studies of Small‐Molecule Inhibitors of the Methyllysine Reader Protein Spindlin1

The methyllysine reader protein Spindlin1 has been implicated in the tumorigenesis of several types of cancer and may be an attractive novel therapeutic target. Small‐molecule inhibitors of Spindlin1 should be valuable as chemical probes as well as potential new therapeutics. We applied an iterative virtual screening campaign, encompassing structure‐ and ligand‐based approaches, to identify potential Spindlin1 inhibitors from databases of commercially available compounds. Our in silico studies coupled with in vitro testing were successful in identifying novel Spindlin1 inhibitors. Several 4‐aminoquinazoline and quinazolinethione derivatives were among the active hit compounds, which indicated that these scaffolds represent promising lead structures for the development of Spindlin1 inhibitors. Subsequent lead optimization studies were hence carried out, and numerous derivatives of both lead scaffolds were synthesized. This resulted in the discovery of novel inhibitors of Spindlin1 and helped explore the structure–activity relationships of these inhibitor series.     

4‐Biphenylalanine‐ and 3‐Phenyltyrosine‐Derived Hydroxamic Acids as Inhibitors of the JumonjiC‐Domain‐Containing Histone Demethylase KDM4A

Overexpression of the histone lysine demethylase KDM4A, which regulates H3K9 and H3K36 methylation states, has been related to the pathology of several human cancers. We found that a previously reported hydroxamate‐based histone deacetylase (HDAC) inhibitor (SW55) was also able to weakly inhibit this demethylase with an IC₅₀ value of 25.4 μm . Herein we report the synthesis and biochemical evaluations, with two orthogonal in vitro assays, of a series of derivatives of this lead structure. With extensive chemical modifications on the lead structure, also by exploiting the versatility of the radical arylation with aryldiazonium salts, we were able to increase the potency of the derivatives against KDM4A to the low‐micromolar range and, more importantly, to obtain demethylase selectivity with respect to HDACs. Cell‐permeable derivatives clearly showed a demethylase‐inhibition‐dependent antiproliferative effect against HL‐60 human promyelocytic leukemia cells.
