Apical membrane antigen 1 is a microneme protein which plays an indispensable role during Apicomplexa parasite invasion. The detailed mechanism of AMA-1 molecular interaction with its receptor on bovine erythrocytes has not been completely defined in Babesia bovis. This study was focused on identifying the minimum B. bovis AMA-1-derived regions governing specific and high-affinity binding to its target cells. Different approaches were used for detecting ama-1 locus genetic variability and natural selection signatures. The binding properties of twelve highly conserved 20-residue-long peptides were evaluated using a sensitive and specific binding assay based on radio-iodination. B. bovis AMA-1 ectodomain structure was modelled and refined using molecular modelling software. NetMHCIIpan software was used for calculating B- and T-cell epitopes. The B. bovis ama-1 gene had regions under functional constraint, having the highest negative selective pressure intensity in the Domain I encoding region. Interestingly, B. bovis AMA-1-DI (100YMQKFDIPRNHGSGIYVDLG119 and 120GYESVGSKSYRMPVGKCPVV139) and DII (302CPMHPVRDAIFGKWSGGSCV321)-derived peptides had high specificity interaction with erythrocytes and bound to a chymotrypsin and neuraminidase-treatment sensitive receptor. DI-derived peptides appear to be exposed on the protein’s surface and contain predicted B- and T-cell epitopes. These findings provide data (for the first-time) concerning B. bovis AMA-1 functional subunits which are important for establishing receptor-ligand interactions which could be used in synthetic vaccine development. 相似文献

Acquiring the skills needed to solve certain types of problems is one of the main challenges of distance university education, demanding extra motivation from students to tackle it. New technology should be one of our great allies in addressing these problems. This article proposes an expert system with a web‐based problem‐solving simulator for a multidevice environment in order to allow students to access an unlimited number of problems with their corresponding solutions, immediately, anytime and anywhere. The types of problems that can be used are those based on quantitative methods with a fixed methodology to be followed. A successful case study was carried out for the subject Operations Management at the Distance University of Madrid (UDIMA), with three different problem simulators having been developed. The results were very satisfactory compared with previous academic years in terms of motivation. A decrease in students that did not sit the final examination was observed, as was an increase in the number of visits to the Moodle classroom. Ultranarrowband organic photodiodes (OPDs) are demonstrated for thin film solid state materials composed of tightly packed dipolar merocyanine dyes. For these dyes the packing arrangement can be controlled by the bulkiness of the donor substituent, leading to either strong H‐ or strong J‐type exciton coupling in the interesting blue (H‐aggregate) and NIR (J‐aggregate) spectral ranges. Both bands are shown to arise from one single exciton band according to fluorescence measurements and are not just a mere consequence of different polymorphs within the same thin film. By fabrication of organic thin‐film transistors, these dyes are demonstrated to exhibit hole transport behavior in spin‐coated thin films. Moreover, when used as organic photodiodes in planar heterojunctions with C60 fullerene, they show wavelength‐selective photocurrents in the solid state with maximum external quantum efficiencies of up to 11% and ultranarrow bandwidths down to 30 nm. Thereby, narrowing the linewidths of optoelectronic functional materials by exciton coupling provides a powerful approach to produce ultranarrowband organic photodiodes. 相似文献

In this work, we report a derivative of N-(piperidin-4-yl)-1H-pyrrole-2-carboxamide as a new inhibitor for adenylyl cyclase of Giardia lamblia which was obtained from a study using structural data of the nucleotidyl cyclase 1 (gNC1) of this parasite. For such a study, we developed a model for this specific enzyme by using homology techniques, which is the first model reported for gNC1 of G. lamblia. Our studies show that the new inhibitor has a competitive mechanism of action against this enzyme. 2-Hydroxyestradiol was used as the reference compound for comparative studies. Results in this work are important from two points of view. on the one hand, an experimentally corroborated model for gNC1 of G. lamblia obtained by molecular modelling is presented; on the other hand, the new inhibitor obtained is an undoubtedly excellent starting structure for the development of new metabolic inhibitors for G. lamblia. 相似文献

The development of inhibitors for hydrochloric acidic media is very common. Potassium iodide, in combination with some organic inhibitors, may transform a mediocre inhibitor into an excellent corrosion inhibitor based on a synergetic effect. There is a need to study what happens at the interface in the presence of this compound. The dynamics of the metal electrolyte interface when KI concentration increases has been characterized. The present study has been carried out based on electrochemical impedance spectroscopy and electrochemical noise analysis in the light of nonlinear phenomena by applying recurrence plots and fractal geometry. The fractal dimension of the current–time series shows a direct relationship with the fractal dimension of the surface of steel. The phenomenon of self-organization has been observed on the metal surface. The nonlinear analysis carried out by means of recurrence plots showed that the iodide ions' inhibitory effect is not only simple adsorption but also a nonlinear phenomenon that auto-organizes. The degree of complexity increases, consequently causing an interaction between the microcells and resulting in a lower charge-transfer rate. 相似文献

Thrombocytes and platelets, beyond their primary function in hemostasis, seem to play an active role in inflammation. As regards their phagocytic ability, the results to date are confusing, incomplete, and somewhat contradictory. Whereas the interaction of avian thrombocytes or mammalian platelets with bacteria both in vitro and in vivo has received wide attention, almost no information exists on the topic in "lower" vertebrates. The aim of this work is to review the available information on the phagocytic properties of thrombocytes and platelets. Particular attention is payed to the ontogeny of these cells, the soluble factors involved in the inflammatory process derived from them, and their interaction with particulate material, mainly with bacteria. 相似文献

This research describes a novel approach for fault detection in industrial processes, by means of unsupervised and projectionist techniques. The proposed method includes a visual tool for the detection of faults, its final aim is to optimize system performance and consequently obtaining increased economic savings, in terms of energy, material, and maintenance. To validate the new proposal, two datasets with different levels of complexity (in terms of quantity and quality of information) have been used to evaluate five well‐known unsupervised intelligent techniques. Management in wireless backhaul networks is a challenging task, especially in rural and isolated environments. In these scenarios, the backhaul network usually consists of a set of heterogeneous wireless links that provide limited and variable bandwidth to the access networks, often 3G/4G small cells. Because of the highly constrained nature of this type of backhaul network, intelligent and joint management in both the backhaul network and the access network is crucial in order to avoid performance degradation caused by traffic congestion. In order to avoid the saturation in the backhaul network, access networks should consider the backhaul state when taking decisions in the admission control and scheduling procedures. However, no standardized mechanisms currently exist for sharing management information between both networks. In this work, we propose to use the Explicit Congestion Notification (ECN) bit in the outer IP headers present in the Iuh 3GPP IPSec‐enabled interfaces in order to notify the backhaul congestion state to the access network. We analyze for the referred scenario, compatibility and security details, validating our approach by running numerically simulations and implementing the notification mechanism. Our low complexity approach offers 2% accuracy and backhaul update latency lower than 10 ms during 80% of the time, which makes the solution appropriate for admission control and scheduling intervals in small cells. 相似文献

We analyse instabilities due to aliasing errors when solving one dimensional non-constant advection speed equations and discuss means to alleviate these types of errors when using high order discontinuous Galerkin (DG) schemes. First, we compare analytical bounds for the continuous and discrete version of the PDEs. Whilst traditional \(L^2\) norm energy bounds applied to the discrete PDE do not always predict the physical behaviour of the continuous version of the equation, more strict elliptic norm bounds correctly bound the behaviour of the continuous PDE. Having derived consistent bounds, we analyse the effectiveness of two stabilising techniques: over-integration and split form variations (conservative, non-conservative and skew-symmetric). Whilst the former is shown to not alleviate aliasing in general, the latter ensures an aliasing-free solution if the splitting form of the discrete PDE is consistent with the continuous equation. The success of the split form de-aliasing is restricted to DG schemes with the summation-by-parts simultaneous-approximation-term properties (e.g. DG with Gauss–Lobatto points).
