The function of aspartic acid residue 101 in the active siteof Escherichia coli alkaline phosphatase was investigated bysite-specific mutagenesis. A mutant version of alkaline phosphatasewas constructed with alanine in place of aspartic acid at position101. When kinetic measurements are carried out in the presenceof a phosphate acceptor, 1.0 M Tris, pH 8.0, both the kcat andthe Km, for the mutant enzyme increase by –2-fold, resultingin almost no change in the kcat/Km ratio. Under conditions ofno external phosphate acceptor and pH 8.0, both the kcat andthe Km for the mutant enzyme decrease by {small tilde}2-fold,again resulting in almost no change in the kcat/Km ratio. Thekcat for the hydrolysis of 4-methyl-umbelliferyl phosphate andp-nitrophenyl phosphate are nearly identical for both the wild-typeand mutant enzymes, as is the K1 for inorganic phosphate. Thereplacement of aspartic acid 101 by alanine does have a significanteffect on the activity of the enzyme as a function of pH, especiallyin the presence of a phosphate acceptor. At pH 9.4 the mutantenzyme exhibits 3-fold higher activity than the wild-type. Themutant enzyme also exhibits a substantial decrease in thermalstability: it is half inactivated by treatment at 49°C for15 min compared to 71°C for the wild-type enzyme. The datareported here suggest that this amino acid substitution altersthe rates of steps after the formation of the phospho-enzymeintermediate. Analysis of the X-ray structure of the wild-typeenzyme indicates that the increase in catalytic rate of themutant enzyme in the presence of a phosphate acceptor may bedue to an increase in accessibility of the active site nearSerl02. The increased catalytic rate of this mutant enzyme maybe utilized to improve diagnostic tests that require alkalinephosphatase, and the reduced heat stability of the mutant enzymemay make it useful in recombinant DNA techniques that requirethe ability to heat-inactivate the enzyme after use.  相似文献

A tunable microwave phase shifter composed of a low-sintering-temperature, screen-printable barium strontium titanate (BST) film and silver metallization was fabricated on an alumina substrate and co-fired at 900 °C for 3 h. The dielectric properties of the films were characterized in a frequency range of 0.8–8 GHz using scattering parameter measurements and a quasi-static coplanar waveguide transmission line model. The temperature dependency of the films was measured through capacitance measurements in a frequency range of 0.5–2.5 GHz. The figure of merit (phase shift/dB of insertion loss) of the phase shifter was found to be 14.6 at 3 GHz with an applied bias field strength of 2.5 V/μm. The performance of the phase shifter is briefly discussed and compared with other phase shifters fabricated by direct screen-printing of BST films.  相似文献

Invasive aquatic species discharged through ballast water is one of the most serious problems posed nowadays in the marine environment. This review paper summarizes the available technologies applied for ballast water treatment. These technologies can be either port‐based or ship‐based, with the latter being easier to implement. Special emphasis was given to onboard treatment methods, which can be categorized as physical separation, mechanical or chemical methods. The efficiency of the methods, as well as the capacity of application and the target microorganisms were compiled and are presented in this review. Particle contamination at the head–disk interface (HDI) is a major concern for the long-term reliability of hard disk drives. In the current study, it is shown that, surprisingly, soft and lubricious talc particles have a significant damaging effect on the friction performance of media lubricants, nanometer-thick perfluoropolyethers. Thermogravimetric analysis (TGA) and the adsorption studies indicate that two mechanisms, Lewis acid–catalyzed decomposition and surface adsorption, are responsible for the observed tribological degradation. The potential impacts of the findings here on the design of next-generation media lubricant have been discussed.  相似文献

The importance of understanding and predicting the interactions of oxides with water vapor at high temperatures is demonstrated in this article. Methods for observing volatilization phenomena and identifying the chemical formulae for volatile metal hydroxides are discussed. In addition, techniques for obtaining accurate thermodynamic data for gaseous metal hydroxide species are described. Detailed examples of the stability of the principle structural and/or protective oxides chromia (Cr2O3), silica (SiO2), and alumina (Al2O3) in high-temperature water vapor are included.  相似文献

An anaerobic mixed microbial culture was enriched from soil and groundwater taken from a site contaminated with trichloroethene (TCE). This enrichment culture was divided into four subcultures amended separately with either perchloroethene (PCE), TCE, cis-dichloroethene (cDCE) or vinyl chloride (VC). In each of the four subcultures, the chlorinated ethenes were rapidly, consistently, and completely converted to ethene at rates of 30-50 micromol/l of culture per day, or an average 160 micro-electron equivalents/l of culture per day. These cultures were capable of sustained and rapid dechlorination of VC, and could not dechlorinate 1,2-dichloroethane, differentiating them from Dehalococcoides ethenogenes, the only known isolate capable of complete dechlorination of PCE to ethene. Chloroform (CF) and 1,1,1-trichloroethane, frequent groundwater co-contaminants with TCE and PCE, inhibited chlorinated ethene dechlorination. Most strongly inhibited was the final conversion of VC to ethene, with complete inhibition occurring at an aqueous CF concentration of 2.5 microM. Differences in rates and community composition developed between the different subcultures, including the loss of the VC enrichment culture's ability to dechlorinate PCE. Denaturing gradient gel electrophoresis of amplified bacterial 16S rRNA gene fragments identified three different DNA sequences in the enrichment cultures, all phylogenetically related to D. ethenogenes. Based on the PCR-DGGE results and substrate utilization patterns, it is apparent that significant mechanistic differences exist between each step of dechlorination from TCE to ethene, especially for the last important dechlorination step from VC to ethene.  相似文献

Immunocytochemistry, in situ hybridization, and radioimmunoassay were employed to examine the cellular distribution of mRNAs and proteins for IGF-I, II, IGF-II/M6P receptor, IGFBP2 as well as the levels of IGF-I and II in normal and unilaterally nephrectomized (Nx) adult rat kidneys. A similar distribution of immunoreactive IGF-I, and -II as well as IGF-II/M6P receptor was found in the principal cells of the cortical collecting duct and in all cells of the inner medullary collecting duct. In addition, immunostainable IGF-I and IGF-II/M6P receptor were noted in some inner medullary loops of Henle, while IGFBP2 was seen in the collecting ducts and loops of Henle of the inner medullar and the renal vasculature of all animals. By comparison, in situ hybridization revealed IGF-I mRNA only in the medullary thick ascending limbs while IGF-II mRNA was localized to the wall of the renal microvasculature in all kidneys. IGFBP2 mRNA was localized to the renal corpuscle and to inner medullary interstitial cells of all kidneys. These data suggest that renal IGF-I and IGFBP2 are synthesized at upstream sites along the nephron and then transported downstream for interaction with IGF receptors. Following nephrectomy, the renal levels of IGF-I peptide and mRNA were elevated at both 5 and 33 days post-nephrectomy, supporting a potential functional role for IGF-I in stimulating the structural and functional recovery in compensatory hypertrophy.  相似文献

We report single-molecule-transistor measurements on devices incorporating magnetic molecules. By studying the electron-tunneling spectrum as a function of magnetic field, we are able to identify signatures of magnetic states and their associated magnetic anisotropy. A comparison of the data to simulations also suggests that sequential electron tunneling may enhance the magnetic relaxation of the magnetic molecule.  相似文献

We present a method to control the work function of metallic carbon nanotube transparent electrodes by functionalization of the random network with metallic nanoparticles. Flexible functionalized electrodes with high transparency (~90%) can be obtained with work function values ranging from ~4.6 up to ~5.1 eV, depending on the nature of the nanoparticles. The work function values were obtained by comparative in situ Kelvin probe force microscopy under ultra high vacuum and ultraviolet photoelectron spectroscopy measurements. Interestingly, by appropriate choice of the metal source for functionalization, work function engineering can lead to work function values higher or lower than that for pristine metallic nanotubes. This could be of great interest for adjusting the work function of transparent electrodes to active layers in many optoelectronics devices.  相似文献

Current methods for lateral force calibration are often time-consuming, expensive, or cause significant wear of the tip. A quick and simple alternative is presented in which the linear relationship between force and voltage is exploited. The technique is independent of the shapes of the sample and cantilever and eliminates common problems, while maintaining better than 10% precision. This advance will facilitate quantitative comparisons between experiments.  相似文献
