Parkinson’s disease (PD) is the most common movement disorder, characterized by progressive degeneration of the nigrostriatal pathway, which consists of dopaminergic cell bodies in substantia nigra and their neuronal projections to the striatum. Moreover, PD is associated with an array of non-motor symptoms such as olfactory dysfunction, gastrointestinal dysfunction, impaired regulation of the sleep-wake cycle, anxiety, depression, and cognitive impairment. Inflammation and concomitant oxidative stress are crucial in the pathogenesis of PD. Thus, this study aimed to model PD via intrastriatal injection of the inflammagen lipopolysaccharide (LPS)to investigate if the lesion causes olfactory and motor impairments, inflammation, oxidative stress, and alteration in synaptic proteins in the olfactory bulb, striatum, and colon. Ten µg of LPS was injected unilaterally into the striatum of 27 male C57BL/6 mice, and behavioural assessment was conducted at 4 and 8 weeks post-treatment, followed by tissue collection. Intrastriatal LPS induced motor impairment in C57BL/6 mice at 8 weeks post-treatment evidenced by reduced latency time in the rotarod test. LPS also induced inflammation in the striatum characterized by increased expression of microglial marker Iba-1 and astrocytic marker GFAP, with degeneration of dopaminergic neuronal fibres (reduced tyrosine hydroxylase immunoreactivity), and reduction of synaptic proteins and DJ-1 protein. Additionally, intrastriatal LPS induced inflammation, oxidative stress and alterations in synaptic proteins within the olfactory bulb, although this did not induce a significant impairment in olfactory function. Intrastriatal LPS induced mild inflammatory changes in the distal colon, accompanied by increased protein expression of 3-nitrotyrosine-modified proteins. This model recapitulated the major features of PD such as motor impairment and degeneration of dopaminergic neuronal fibres in the striatum, as well as some pathological changes in the olfactory bulb and colon; thus, this model could be suitable for understanding clinical PD and testing neuroprotective strategies.  相似文献

Detection of staphylococcal enterotoxin B in spiked food samples   总被引:9,自引:0,他引:9  
Contamination of food with infectious agents, intentional or not, is a global concern with far-reaching economic and social impact. Staphylococcal enterotoxins are a major cause of food poisoning, but most methods for the identification of these agents in food require extensive pretreatment or concentration of the sample prior to analysis. The array biosensor was developed as a portable device for the simultaneous analysis of multiple complex samples for multiple targets with minimal sample preparation. In this study, we use an array biosensor to expand and improve on a staphylococcal enterotoxin B (SEB) assay with the ultimate intent of incorporating testing for SEB into a battery of sensitive and convenient assays for food safety validation. In addition to buffer studies, six different types of food samples, including beverages, homogenates of fruit and meat, and carcass washings, were spiked with SEB, incubated for at least 2 h to permit antigen sequestration, and assayed. For all samples, there were differences in fluorescence intensity, but 0.5 ng of SEB per ml could be detected in <20 min with little if any pretreatment and no sample preconcentration.  相似文献

Common domestic solar water heating system usage patterns were investigated by a survey of 55 installations. These usage patterns were modelled by simulation based on the actual occupants' use of boiler or other auxiliary heating control strategies. These strategies were not optimal, as often assumed. The effectiveness of the technology was found to be highly sensitive to the time settings used for auxiliary water heating, and the 65% of solar householders using their boilers in the mornings were found to be forgoing 75% of their potential savings. Additionally, 92% of consumers were found to be small households, whose potential savings were only 23% of those of larger households, which use more hot water. Overall the majority (at least 60%) of the systems surveyed were found to be achieving no more than 6% of their potential savings. Incorporating consideration of Legionella issues, results indicate that if solar thermal technology is to deliver its potential to CO2 reduction targets: solar householders must avoid any use of their auxiliary water heating systems before the end of the main warmth of the day, grants for solar technology should be focused on households with higher hot water demands, and particularly on those that are dependent on electricity for water heating, health and safety requirements for hot water storage must be reviewed and, if possible, required temperatures should be set at a lower level, so that carbon savings from solar water heating may be optimized.  相似文献

Nine-analyte detection using an array-based biosensor   总被引:4,自引:0,他引:4  
A fluorescence-based multianalyte immunosensor has been developed for simultaneous analysis of multiple samples. While the standard 6 x 6 format of the array sensor has been used to analyze six samples for six different analytes, this same format has the potential to allow a single sample to be tested for 36 different agents. The method described herein demonstrates proof of principle that the number of analytes detectable using a single array can be increased simply by using complementary mixtures of capture and tracer antibodies. Mixtures were optimized to allow detection of closely related analytes without significant cross-reactivity. Following this facile modification of patterning and assay procedures, the following nine targets could be detected in a single 3 x 3 array: Staphylococcal enterotoxin B, ricin, cholera toxin, Bacillus anthracis Sterne, Bacillus globigii, Francisella tularensis LVS, Yersiniapestis F1 antigen, MS2 coliphage, and Salmonella typhimurium. This work maximizes the efficiency and utility of the described array technology, increasing only reagent usage and cost; production and fabrication costs are not affected.  相似文献

Reports an error in the article "Choice Processing in Emotionally Difficult Decisions' by Mary Frances Luce, James R. Bettman, and John W. Payne (Journal of Experimental Psychology: Learning, Memory, and Cognition, 1997, Vol. 23, No. 2, pp. 384–405; see record 1997-03378-007). In Table 7 on page 396, the high conflict/low trade-off difficulty mean for the PATTERN variable was printed as -0.13, but the actual mean is 0.13. A portion of the original abstract follows: Extends the standard effort-accuracy approach to explaining task influences on decision processing by arguing that coping goals will interact with effort-minimization goals for negatively emotion-laden decision tasks. These coping goals may involve both a desire to process in a thorough, accurate manner and a desire to avoid particularly distressing aspects of processing. The authors hypothesized and found in 3 experiments that decision processing under increasing negative emotion both becomes more extensive and proceeds more by focusing on one attribute at a time. In particular, increased negative emotion leads to more attribute-based processing at the beginning of the decision process. The results are inconsistent with views that negative emotion acts only as an incentive or only as a source of decision complexity. This article continues the series of electronic microscopical investigations of tick attachment and host's inflammatory reaction in the attachment site. In all this works we used larvae of different Ixodid ticks but the same species of the host (white mouse), Ixodid species having different type of attachment. Dermacentor marginatus (Amosova, 1989a) is characterized by the superficial penetration of mouth parts, the abundance of cement around them and on the surface of the host's skin, the mouth parts of Hyalomma asiaticum are fully inserted and the superficial cement is minimal (Amosova, 1989b) and Ixodes ricinus has mouth parts inserted more deeply in the skin tissue of the host and the cement substance localised only on the surface of the skin at the 1st and 2nd days of feeding period. For this investigation we choose Haemaphysalis longicornis because it is known (see Kemp et al., 1982) that genus Haemaphysalis has the most superficial localisation of the mouth parts in the attachment site. We have studied a fine structure of cement cone and the cement substance surrounding the mouth parts in the skin and have demonstrated that it is very similar to the Dermacentor marginatus cement structure. The pattern of cellular response is similar to D. marginatus lesion too.  相似文献
