The objectives of the study were to estimate the reproducibility and repeatability of milk coagulation properties (MCP) measured by a computerized renneting meter (CRM) and to evaluate the predictive ability of mid-infrared spectroscopy (MIRS) as an innovative technology for the assessment of rennet coagulation time (RCT, min) and curd firmness (a30, mm). Four samples without addition of preservative (NP) and 4 samples with Bronopol addition (PS) were collected from each of 83 Holstein-Friesian cows. Six hours after collection, 2 replicated measures of MCP were obtained with CRM using 1 NP and 1 PS sample from each cow. Mid-infrared spectra of the remaining NP and PS samples from each animal were recorded after 6 h, 4 d, and 8 d after sampling. Two groups of calibration equations were developed using MIRS spectra and CRM measures of MCP as reference data obtained from analysis of NP and PS, respectively. Reproducibility and repeatability of CRM measures were obtained from REML estimation of variance components on the basis of a linear model including the fixed effects of herd and days in milk class and the random effects of cows, sample treatment (addition or no addition of preservative), and the interaction between cow and sample treatment. Coefficient of reproducibility is an indicator of the agreement between 2 measurements of MCP for the same milk sample preserved with or without addition of Bronopol. Coefficient of repeatability is an indicator of the agreement between repeated measures of MCP. Pearson correlations between MCP measures for NP and PS were 0.97 and 0.83 for RCT and a30, respectively. Reproducibility of CRM measures under different preserving conditions of milk was 93.5% for RCT and 64.6% for a30. Repeatabilities of RCT and a30 measures were 95.7 and 77.3%, respectively. Based on the estimated cross-validation standard errors and coefficients of determination and ratios of standard errors of cross-validation to standard deviation of reference data, the predictive ability of MIRS calibration equations was moderate for RCT and unsatisfactory for a30. Predictive ability of equations based on spectra and MCP measures of PS was greater than that of equations based on data of NP. The study did not provide conclusive evidence on the effectiveness of MIRS as a predictive tool for MCP and it requires an enlargement of the variability of milk sampling circumstances. Because the relevance of MIRS predictions in relation to breeding programs for MCP based on indicator traits relies on the genetic variation of MIRS predictions and on phenotypic and genetic correlations between MIRS predictions and MCP measures, additional specific investigations on these topics are needed.  相似文献

Two gas diffusion layers based on the same carbon cloth substrate, produced by an Italian Company (SAATI), and coated with microporous layers of different hydrophobicities, were assembled in a polymer electrolyte membrane fuel cell and its performances assessed. For comparison the cell mounting the carbon cloth without microporous layer was also tested. The membrane electrode assembly was made of Nafion® 212 with Pt load 0.3/0.6 mg cm−2 (anode/cathode). The cell testing was run at 60 °C and 80 °C with fully humidified air (100%RH) and 80%RH hydrogen feedings. The assembly of gas diffusion layers and membrane with electrodes was compressed to 30% and 50% of its initial thickness. For each configuration polarization and power curves were recorded; in order to evaluate the role of different GDLs, AC impedance spectroscopy of the running cell was also performed.The higher compression ratio caused the worsening of cell performances, partially mitigated when the operating temperature was raised to 80 °C. The presence of the microporous layer onto the carbon cloth resulted extremely beneficial for the operations especially at high current density; moreover, it sensibly reduces the high frequency resistance of the overall assembly.  相似文献

A new method specific for the determination and unambiguous confirmation of the antibiotic valnemulin in feed for all animal species is described. Simple clean-up based on solvent extraction and liquid partition is carried out prior to determination by liquid chromatography coupled to electrospray tandem mass spectrometry on a hybrid QTRAP 4000 system, in multiple reaction monitoring mode. Valnemulin is licensed in the European Union as a veterinary drug for use in feed for pigs and rabbits, but unavoidable carryover in feed for non-target species should be monitored. The method is rapid and reliable, and was validated in-house at 10, 100 and 750 ng g1, evaluating the analytical performances according to Commission Regulation (EC) No. 882/2004. Good mean recoveries from 79.7% to 92.6%, and within-laboratory reproducibility RSDs, ranging from 6.2% to 12.1%, were measured; the limit of quantification at 10.0 ng g1 also allows for sensitive evaluation of undesirable carryover in feed for non-target species. The results of monitoring 24 compound feeds for several animal species are also reported.  相似文献

Steaks from gluteus medius m. (GM) and longissimus dorsi m. (LD) were packaged in atmospheres containing two low levels of CO (0.2 and 0.4%) in combination with 21% O2 or without oxygen. The other components of the modified atmosphere packaging systems were carbon dioxide (30%) and nitrogen (balance). Steaks were evaluated on 0, 7, 14 and 21 days of storage at about 2oC to investigate the effects of the gas mixtures on the microflora, instrumental (CIE L*a*b*) and visual colour and odour. The preservation of colour and odour of GM and LD steaks packaged under low carbon monoxide was best achieved using anoxic mixtures containing 0.2% CO and 30% CO2. Steaks packaged without oxygen retained a desirable colour and practically did not present a visible discoloration or perceptive off‐odour along the entire storage time. Colour stabilization with 0.2% CO and 30% CO2, without O2, following storage up to 21 days at 2°C did not increase microbial risk of products. The increase in CO from 0.2 to 0.4% had a negative effect on the colour of LD steaks which was regarded as ‘artificial’ by some panellists. The higher stability achieved in this study using 0.2% CO may result from increased stability of the colour from Bos indicus × Bos Taurus cross. Discoloration and off‐odour were evident in the GM and LD steaks packaged aerobically after 14 days of storage when the counts of deteriorant bacteria achieved approximately 6.5 to 9.7 log CFU/g. Highest bacteriostatic effect could be obtained if the volume of injected CO2 was sufficient to saturate the meat and keep in balance around the product during storage. ABSTRACT:  Deoxynivalenol, zearalenone, and aflatoxin concentrations in Sicilian durum wheat were determined through ELISA tests. The results highlighted the safety of the grain samples at harvest because deoxynivalenol, zearalenone, and aflatoxin levels did not exceed European legal limits. With regard to aflatoxins, reliability of the ELISA test was evaluated, comparing it with HPLC analyses. The comparison of HPLC and ELISA data showed a tendency to overestimate aflatoxin concentrations with respect to chromatographic determinations. The usefulness of ELISA was confirmed as a rapid screening method; however, when contamination levels are close to legal limits, chromatographic analyses are necessary to quantify aflatoxins with greater accuracy and specificity.  相似文献

Osteosarcoma (OS) is the most common primary bone tumor mainly occurring in young adults and derived from primitive bone-forming mesenchyme. OS develops in an intricate tumor microenvironment (TME) where cellular function regulated by microRNAs (miRNAs) may affect communication between OS cells and the surrounding TME. Therefore, miRNAs are considered potential therapeutic targets in cancer and one of the goals of research is to accurately define a specific signature of a miRNAs, which could reflect the phenotype of a particular tumor, such as OS. Through NGS approach, we previously found a specific molecular profile of miRNAs in OS and discovered 8 novel miRNAs. Among these, we deepen our knowledge on the fifth candidate renamed now miR-CT3. MiR-CT3 expression was low in OS cells when compared with human primary osteoblasts and healthy bone. Through TargetScan, VEGF-A was predicted as a potential biological target of miR-CT3 and luciferase assay confirmed it. We showed that enforced expression of miR-CT3 in two OS cell lines, SAOS-2 and MG-63, reduced expression of VEGF-A mRNA and protein, inhibiting tumor angiogenesis. Enforced expression of miR-CT3 also reduced OS cell migration and invasion as confirmed by soft agar colony formation assay. Interestingly, we found that miR-CT3 behaves inducing the activation of p38 MAP kinase pathway and modulating the epithelial-mesenchymal transition (EMT) proteins, in particular reducing Vimentin expression. Overall, our study highlights the novel role of miR-CT3 in regulating tumor angiogenesis and progression in OS cells, linking also to the modulation of EMT proteins.  相似文献
