This investigation aimed to determine the role of general stress-response alternative sigma factors σS (RpoS) and σB (SigB) in heat resistance and the occurrence of sublethal injuries in cell envelopes of stationary-phase Escherichia coli BJ4 and Listeria monocytogenes EGD-e cells, respectively, as a function of treatment medium pH. Given that microbial death followed first-order inactivation kinetics (R2 > 0.95) the traditional DT and z values were used to describe the heat inactivation kinetics.Influence of rpoS deletion was constant at every treatment temperature and pH, making a ΔrpoS deletion mutant strain approximately 5.5 times more heat sensitive than its parental strain for every studied condition. Furthermore, the influence of the pH of the treatment medium on the reduction of the heat resistance of E. coli was also constant and independent of the treatment temperature (average z value = 4.9 °C) in both parental and mutant strains.L. monocytogenes EGD-e z values obtained at pH 7.0 and 5.5 were not significantly different (p > 0.05) in either parental or the ?sigB deletion mutant strains (average z value = 4.8 °C). Nevertheless, at pH 4.0 the z value was higher (z = 8.4 °C), indicating that heat resistance of both L. monocytogenes strains was less dependent on temperature at pH 4.0. At both pH 5.5 and 7.0 the influence of sigB deletion was constant and independent of the treatment temperature, decreasing L. monocytogenes heat resistance approximately 2.5 times. In contrast, the absence of sigB did not decrease the heat resistance of L. monocytogenes at pH 4.0.The role of RpoS in protecting cell envelopes was more important in E. coli (4 times) than SigB in L. monocytogenes (1.5 times). Moreover, the role of σS in increasing heat resistance seems more relevant in enhancing the intrinsic resilience of the cytoplasmic membrane, and to a lesser extent, outer membrane resilience.Knowledge of environmental conditions related to the activation of alternative sigma factors σS and σB and their effects on heat resistance would help us to avoid and/or identify situations that increase bacterial stress resistance. Therefore, more efficient food preservation processes might be designed.  相似文献

The processing parameters related with chia oil extraction employing screw press have not been studied yet. A Box–Behnken experimental design was used to study the optimization process by response surface analysis. The independent variables considered were seed moisture content, restriction die, screw press speed and barrel temperature, while the response variables measured were oil yield, fines content in oil and oil quality (acidity, peroxide index, K232, K270, values, antioxidant activity and total tocopherol content). Since chemical quality data of chia seeds oil pressed at different conditions was not affected, the response was optimize to maximize oil yield. The results suggested that 0.113 g/g dry solids (0.101 g/g seed), 6 mm restriction die, 20 rpm screw press speed and 30 °C barrel temperature were the best processing combination to maximize oil yield.  相似文献

The goal of this work was to analyze the effect of different treatments on functional properties (Color parameters, water holding capacity, oil holding capacity, water imbibition capacity, protein solubility and apparent viscosity) and microstructure of three cowpea varieties. Proteins present in flours obtained from heat-treated seeds exhibited a significant increase in water and oil absorption values, water imbibition capacity, and apparent viscosity, whereas solubility decreased. Color determinations on flours showed a reduction in L* and H*, and an increment in C* with treatment intensity. Structural changes in seeds would explain the variations in the functionality of flours. This study shows that cooking and autoclaving affect significantly the functional properties of flours, while soaking had a lower impact. The knowledge about these changes will be very useful for developing and obtaining new preparations based on cowpea flour.  相似文献

The effects of fat supplements that differed in fatty acid composition (chain length and degree of saturation) and chemical form (free fatty acids, Ca salts of fatty acids, and triacylglyceride) on digestible energy (DE) concentration of the diet and DE intake by lactating cows were measured. Holstein cows were fed a control diet [2.9% of dry matter (DM) as long-chain fatty acids] or 1 of 3 diets with 3% added fatty acids (that mainly replaced starch). The 3 fat supplements were (1) mostly saturated (C18:0) free fatty acids (SFA), (2) Ca-salts of fatty acids (CaFA), and (3) triacylglyceride high in C16:0 fatty acids (TAG). Cows fed CaFA (22.8 kg/d) consumed less DM than cows fed the control (23.6 kg/d) and TAG (23.8 kg/d) diets but similar to cows fed SFA (23.2 kg/d). Cows fed fat produced more fat-corrected milk than cows fed the control diet (38.2 vs. 41.1 kg/d), mostly because of increased milk fat percentage. No differences in yields of milk or milk components were observed among the fat-supplemented diets. Digestibility of DM, energy, carbohydrate fractions, and protein did not differ between diets. Digestibility of long-chain fatty acids was greatest for the CaFA diet (76.3%), intermediate for the control and SFA diets (70.3%), and least for the TAG diet (63.3%). Fat-supplemented diets had more DE (2.93 Mcal/kg) than the control diet (2.83 Mcal/kg), and DE intake by cows fed supplemented diets was 1.6 Mcal/d greater than by cows fed the control, but no differences were observed among the supplements. Because the inclusion rate of supplemental fats is typically low, large differences in fatty acid digestibility may not translate into altered DE intake because of small differences in DM intake or digestibility of other nutrients.  相似文献

BACKGROUND: The efficacy of common fungicides to control downy mildew (Plasmopara viticola) in vines decreases when the same commercial formulations are applied over several years. For this reason, new commercial formulations (containing new fungicides) have to offer to grape‐growers to increase the efficacy of antifungal treatments. The efficacy of five new commercial formulations (Cabrio Top, Equation Pro, Fantic M, Mikal Premium and Fobeci) to control downy mildew in an experimental vineyard producing white grapes (Vitis vinifera cv. Godello) was studied. All these products have in common a new fungicide (pyraclostrobin, famoxadone, benalaxyl‐M, iprovalicarb and benalaxyl, respectively) in their chemical formulation. RESULTS: Two commercial formulations, Cabrio Top and Fobeci, gave the highest efficacy to control this fungal disease. Once treated white grapes were harvested, the fate of these fungicides from vine to wine was studied. The white winemaking process allowed the dissipation of fungicide residues in high percentages (~95%), except for iprovalicarb (45%). Concentrations in final filtered white wines were close to or lower than 0.01 mg L?1 for all detected fungicides except iprovalicarb and fludioxonil. The following dissipation rate was found: famoxadone > cyprodinil > pyraclostrobin > benalaxyl > folpet > fludioxonil > benalaxyl‐M > penconazole > iprovalicarb. CONCLUSION: From the data obtained in the vinification process, estimated maximum residue levels for white wines were proposed for future EU legislation to control the presence of pesticides in white wines. The behavioral and electroantennographic responses of Prorhinotermes canalifrons to its soldier frontal gland secretion, and two separated major components of the secretion, (E)-1-nitropentadec-1-ene and (E,E)-α-farnesene, were studied in laboratory experiments. Behavioral experiments showed that both the frontal gland secretion and (E,E)-α-farnesene triggered alarm reactions in P. canalifrons, whereas (E)-1-nitropentadec-1-ene did not affect the behavior of termite groups. The alarm reactions were characterized by rapid walking of activated termites and efforts to alert and activate other members of the group. Behavioral responses to alarm pheromone differed between homogeneous and mixed groups, suggesting complex interactions. Antennae of both soldiers and pseudergates were sensitive to the frontal gland secretion and to (E,E)-α-farnesene, but soldiers showed stronger responses. The dose responses to (E,E)-α-farnesene were identical for both soldiers and pseudergates, suggesting that both castes use similar receptors to perceive (E,E)-α-farnesene. Our data confirm (E,E)-α-farnesene as an alarm pheromone of P. canalifrons.  相似文献

The decolorisation and mineralisation of a model mixture of reactive dyes, under Fenton and photo‐Fenton conditions, have been investigated. A kinetic study was performed and rate constants and half‐life times determined from batch experimental data according to a pseudo‐first‐order degradation kinetic model. Moreover, the use of natural or artificial light, as well as the effect of temperature, was evaluated with the aim of finding the best Fenton process to promote the efficient degradation of the standard trichromatic system. The experimental results showed that the Fenton process run under solar light was the most effective. It is suggested that Fenton‐ and photo‐Fenton‐type reactions are viable techniques for the treatment of these types of dye mixtures, according to the high levels of colour, chemical oxygen demand and total organic carbon removal.  相似文献

The methanolic extract of the marine sponge Ircinia felix has yielded nine novel fatty acid esters, (7E, 12E, 18R, 20Z)-variabilin (5Z, 9Z)-22-methyltricosadienoate, (7E, 12E, 18R, 20Z)-variabilin (5Z, 9Z)-tetracosadienoate, (7E, 12E, 18R, 20Z)-variabilin hexadecanoate, (7E, 12E, 18R, 20Z)-variabilin 10-methylhexadecanoate, (7E, 12E, 18R, 20Z)-variabilin 15-methylhexadecanoate, (7E, 12E, 18R, 20Z)-variabilin 14-methylhexadecanoate, (7E, 12E, 18R, 20Z)-variabilin 9-octadecenoate, (7E, 12E, 18R, 20Z)-variabilin octadecanoate, and (7E, 12E, 18R, 20Z)-variabilin 2,11-dimethyloctadecanoate, along with the recently described (7E, 12E, 18R, 20Z)-variabilin 11-methyloctadecanoate. The characterization of the new fatty acids (5Z, 9Z)-22-methyltricosadienoic and 2,11-dimethyloctadecanoic acids is also described. The chemical structures were determined by extensive spectroscopic, chromatographic, and chemical analyses.  相似文献

The nucleation and growth mechanism of some homopolymers of aniline (six monomers were studied: 3,5‐dichloroaniline, 2,5‐dichloroaniline, 2,6‐dichloroaniline, 2,3‐dichloroaniline, 2,5‐dibromoaniline, and 2,6‐dibromoaniline), synthesized by potentiostatic methods, was determined with a mathematical model that considers different contributions from current–time transients with a gold‐disc electrode. Deconvolution of the transients for the dichlorinated monomers showed IN3Dct and PN3Ddif contributions (where IN3Dct refers to an instantaneous nucleation and three‐dimensional growth mechanism under charge‐transfer control and PN3Ddif refers to a progressive nucleation and three‐dimensional growth mechanism under diffusion control), whereas IN2D and PN3Ddif components and IN2D, IN3Dct, and PN3Ddif components (where IN2D refers to an instantaneous nucleation and two‐dimensional growth mechanism) were needed for 2,5‐dibromoaniline and 2,6‐dibromoaniline, respectively. The percentage of the contribution of the current–time transient to the total charge was worked out for each monomer. The effect of the scan rate on the voltammetric profile during the potentiodynamic electrosynthesis of the polymers was studied too. Curves of the current versus the square root of the potential scan rate were recorded for a selected group of monomers, and the slope was considered an estimation of the diffusion coefficient of the respective monomer. Furthermore, the electrosynthesized polymers were characterized with Fourier transform infrared, ultraviolet–visible, X‐ray photoelectron spectroscopy, and scanning electron microscopy. Microanalysis, used to establish the ratio of the atomic percentages of P and N for each polymer synthesized at a constant potential, was performed for doped and undoped polymers. This parameter was a measure of the degree of electrochemical doping. The conductivity of the doped and undoped polymers was also measured. Hence, the systematic characterization of this analogue series of monomers allows, before generalization, an adequate experimental design to prepare polymers with the properties required for their use. © 2009 Wiley Periodicals, Inc. Different high impact polystyrenes were synthesized using styrene/butadiene copolymers (SB) with PS/PB composition: 30/70 and 20/80 as the precursor rubber, benzoyl peroxide (BPO) as the initiator, and ter‐dodecyl mercaptane (TDM) as the chain transfer agent. During the polymerization, several samples were taken and analyzed under different techniques to evaluate the phase inversion (PI) phenomenon. The PI was determined through transmission electron microscopy (TEM) and through dynamic oscillatory behavior, where the PI takes places when the relaxation process presents the lower value of activation energy. Finally, the Choi and Schowalter emulsion model was employed to elucidate the PI, and relevant information was revealed about the interfacial tension in the PI. POLYM. ENG.
