Increasing evidence has shown that vascular aging has a key role in the pathogenesis of vascular diseases. P300/CBP-associated factor (PCAF) is involved in many vascular pathological processes, but the role of PCAF in vascular aging is unknown. This study aims to explore the role and underlying mechanism of PCAF in vascular aging. The results demonstrated that the expression of PCAF was associated with age and aging, and remarkably increased expression of PCAF was present in human atherosclerotic coronary artery. Downregulation of PCAF could reduce angiotensin II (AngII)-induced senescence of rat aortic endothelial cells (ECs) in vitro. In addition, inhibition of PCAF with garcinol alleviated AngII-induced vascular senescence phenotype in mice. Downregulation of PCAF could alleviate AngII-induced oxidative stress injury in ECs and vascular tissue. Moreover, PCAF and nuclear factor erythroid-2-related factor 2 (Nrf2) could interact directly, and downregulation of PCAF alleviated vascular aging by promoting the activation of Nrf2 and enhancing the expression of its downstream anti-aging factors. The silencing of Nrf2 with small interfering RNA attenuated the protective effect of PCAF downregulation from vascular aging. These findings indicate that downregulation of PCAF alleviates oxidative stress by activating the Nrf2 signaling pathway and ultimately inhibits vascular aging. Coronary artery spasm (CAS) plays an important role in the pathogenesis of many ischemic heart entities; however, there are no established diagnostic biomarkers for CAS in clinical and forensic settings. This present study aimed to identify such serum biomarkers by establishing a rabbit CAS provocation model and integrating quantitative serum proteomics, parallel reaction monitoring/mass spectrometry-based targeted proteomics, and partial least-squares discriminant analysis (PLS-DA). Our results suggested that SELENBP1 and VCL were potential candidate biomarkers for CAS. In independent clinical samples, SELENBP1 and VCL were validated to be significantly lower in serum but not blood cells from CAS patients, with the reasons for this possibly due to the decreased secretion from cardiomyocytes. The areas under the curve of the receiver operating characteristics (ROC) analysis were 0.9384 for SELENBP1 and 0.9180 for VCL when diagnosing CAS. The CAS risk decreased by 32.3% and 53.6% for every 10 unit increases in the serum SELENBP1 and VCL, respectively. In forensic samples, serum SELENBP1 alone diagnosed CAS-induced deaths at a sensitivity of 100.0% and specificity of 72.73%, and its combination with VCL yielded a diagnostic specificity of 100.0%, which was superior to the traditional biomarkers of cTnI and CK-MB. Therefore, serum SELENBP1 and VCL could be effective biomarkers for both the clinical and forensic diagnosis of CAS. 相似文献

The growing region of herbaceous peony (Paeonia lactiflora) has been severely constrained due to the intensification of global warming and extreme weather events, especially at low latitudes. Assessing and selecting stress-tolerant and high-quality peony germplasm is essential for maintaining the normal growth and application of peonies under adverse conditions. This study proposed a modified multi-criteria decision-making (MCDM) model for assessing peonies adapted to low-latitude climates based on our previous study. This model is low-cost, timesaving and suitable for screening the adapted peony germplasm under hot and humid climates. The evaluation was conducted through the analytic hierarchy process (AHP), three major criteria, including adaptability-related, ornamental feature-related and growth habits-related criteria, and eighteen sub-criteria were proposed and constructed in this study. The model was validated on fifteen herbaceous peonies cultivars from different latitudes. The results showed that ‘Meiju’, ‘Hang Baishao’, ‘Hongpan Tuojin’ and ‘Bo Baishao’ were assessed as Level I, which have strong growth adaptability and high ornamental values, and were recommended for promotion and application at low latitudes. The reliability and stability of the MCDM model were further confirmed by measuring the chlorophyll fluorescence of the selected adaptive cultivars ‘Meiju’ and ‘Hang Baishao’ and one maladaptive cultivar ‘Zhuguang’. 在设计强度为C35的煤矸石保温混凝土中分别掺入0.05%、0.1%、0.15%和0.2%的聚丙烯纤维(PPF),并对3 d、7 d、14 d及28 d龄期下的混凝土试件进行立方体抗压强度试验。试验结果表明,在各龄期下,立方体抗压强度随着PPF掺量的增加,均呈现出先上升后下降的趋势,在纤维掺量为0.05%时提高最大;纤维的掺入可以有效提高煤矸石保温混凝土的早期抗压强度。根据试验结果建立了PPF煤矸石保温混凝土的抗压强度预测模型,依据7 d的抗压强度可以较好地预测28 d抗压强度。 相似文献

为提升多股碳纤维导线的质量,研究基于高精度温度传感器的多股碳纤维导线潜伏性缺陷检测方法。利用布里渊光时域反射技术设计高精度温度传感器,并将单根光纤植入多股碳纤维导线中,通过注入窄谱光源来实现温度采集。采用外差检测法对窄谱光源进行处理,获取多股碳纤维导线的温度采集结果。采用平均处理方法滤除温度数据内部的噪声。基于滤波后的温度数据,建立多股碳纤维导线的温度场逆问题模型,并利用牛顿迭代法对模型进行求解,从而得到多股碳纤维导线的热导率。实验证明,该方法能够有效地采集多股碳纤维导线的温度,并成功滤除99%的内部噪声。检测无缺陷多股碳纤维导线的热导率是235 W/m·k,基于此,该方法也能够根据与无缺陷导线的热导率的差异有效地检测导线的潜伏性缺陷。值得注意的是,当导线的负荷电流增加时,该方法的潜伏性缺陷检测效果更佳。 相似文献
