Peutz-Jeghers syndrome (PJS) is an autosomal dominant disease characterized by mucocutaneous pigmentation and hamartomatous polyps. There is an increased risk of benign and malignant tumors in the gastrointestinal tract and in extraintestinal tissues. One PJS locus has been mapped to chromosome 19p13.3; a second locus is suspected on chromosome 19q13.4 in a minority of families. The PJS gene on 19p13.3 has recently been cloned, and it encodes the serine/threonine kinase LKB1. The gene, which is ubiquitously expressed, is composed of 10 exons spanning 23 kb. Several LKB1 mutations have been reported in heterozygosity in PJS patients. In this study, we screened for LKB1 mutations in nine PJS families of American, Spanish, Portuguese, French, Turkish, and Indian origin and detected seven novel mutations. These included two frameshift mutations, one four-amino-acid deletion, two amino-acid substitutions, and two splicing errors. Expression of mutant LKB1 proteins (K78I, D176N, W308C, and L67P) and assessment of their autophosphorylation activity revealed a loss of the kinase activity in all of these mutants. These results provide direct evidence that the elimination of the kinase activity of LKB1 is probably responsible for the development of the PJS phenotypes. In two Indian families, we failed to detect any LKB1 mutation; in one of these families, we previously had detected linkage to markers on 19q13.3-4, which suggests locus heterogeneity of PJS. The elucidation of the molecular etiology of PJS and the positional cloning of the second potential PJS gene will further elucidate the involvement of kinases/phosphatases in the development of cancer-predisposing syndromes.  相似文献

We have recently characterised the genomic organisation of a novel interphotoreceptor matrix proteoglycan, IMPG1, and have mapped the gene locus to chromosome 6q13-q15 by fluorescence in situ hybridisation. As the interphotoreceptor matrix (IPM) is thought to play a critical role in retinal adhesion and the maintenance of photoreceptor cells, it is conceivable that a defect in one of the IPM components may cause degenerative lesions in retinal structures and thus may be associated with human retinopathies. By genetic linkage analysis, several retinal dystrophies including one form of autosomal dominant Stargardt-like macular dystrophy (STGD3), progressive bifocal chorioretinal atrophy (PBCRA), and North Carolina macular dystrophy (MCDR1) have previously been localised to a region on proximal 6q that overlaps the IMPG1 locus. We have therefore assessed the entire coding region of IMPG1 by exon amplification and subsequent single stranded conformational analysis in patients from 6q linked multigeneration families diagnosed with PBCRA and MCDR1, as well as a single patient from an autosomal dominant STGD pedigree unlinked to either of the two known STGD2 and STGD3 loci on chromosomes 13q and 6q, respectively. No disease associated mutations were identified. In addition, using an intragenic polymorphism, IMPG1 was excluded by genetic recombination from both the PBCRA and the MCDR1 loci. However, as the autosomal dominant Stargardt-like macular dystrophies are genetically heterogeneous, other forms of this disorder, in particular STGD3 previously linked to 6q, may be caused by mutations in IMPG1.  相似文献

One‐dimensional (1D) porous materials have shown great potential for gas storage and separation, sensing, energy storage, and conversion. However, the controlled approach for preparation of 1D porous materials, especially porous organic materials, still remains a great challenge due to the poor dispersibility and solution processability of the porous materials. Here, carbon nanotube (CNT) templated 1D conjugated microporous polymers (CMPs) are prepared using a layer‐by‐layer method. As‐prepared CMPs possess high specific surface areas of up to 623 m2 g?1 and exhibit strong electronic interactions between p‐type CMPs and n‐type CNTs. The CMPs are used as precursors to produce heteroatom‐doped 1D porous carbons through direct pyrolysis. As‐produced ternary heteroatom‐doped (B/N/S) 1D porous carbons possess high specific surface areas of up to 750 m2 g?1, hierarchical porous structures, and excellent electrochemical‐catalytic performance for oxygen reduction reaction. Both of the diffusion‐limited current density (4.4 mA cm?2) and electron transfer number (n = 3.8) for three‐layered 1D porous carbons are superior to those for random 1D porous carbon. These results demonstrate that layered and core–shell type 1D CMPs and related heteroatom‐doped 1D porous carbons can be rationally designed and controlled prepared for high performance energy‐related applications.  相似文献

Zusammenfassung Gegenstand dieses Beitrages ist die Ermittlung von Distanzindizes, die als Maß für die Ähnlichkeit von durch quantitative Merkmalsausprägungen charakterisierbaren Objekten verwandt werden können. Im Gegensatz zu dem üblichen Vorgehen, bei dem ein Distanzindex gewählt wird, der mehr oder weniger wünschenswerte Eigenschaften besitzt, werden zunächst Bedingungen formuliert, die als sinnvoll für den gesuchten Distanzindex angesehen werden. Anschließend wird die Klasse derjenigen Distanzindizes ermittelt, die diesen Bedingungen genügt. Es wird gezeigt, daß die Forderung von vier derartigen Bedingungen die gewichtete Lr-Distanz impliziert.
Summary Subject of this paper is the determination of a distance function defined on the set of observed characteristics of objects (individuals, goods, enterprises etc.). In contrast to the usual approach, where a dissimilarity coefficient exhibiting some more or less desirable properties is chosen, at first conditions to be met by the dissimilarity coefficient are set. Then the class of dissimilarity coefficients that meets these conditions is determined. It is shown, that four such conditions imply the weighted Lr-metric.

Staphylococcus aureus genotype B (GTB) is a contagious mastitis pathogen in cattle, occurring in up to 87% of individuals. Because treatment is generally insufficient, culling is often required, leading to large economic loss in the Swiss dairy industry. As the detection of this pathogen in bulk tank milk (BTM) would greatly facilitate its control, a novel real-time quantitative PCR-based assay for BTM has previously been developed and is now being evaluated for its diagnostic properties at the herd level. Herds were initially classified as to their Staph. aureus GTB status by a reference method. Using BTM and herd pools of single-quarter and 4-quarter milk, the herds were then grouped by the novel assay, and the resulting classifications were compared. A total of 54 dairy herds were evaluated. Using the reference method, 21 herds were found to be GTB positive, whereas 33 were found to be negative. Considering the novel assay using both herd pools, all herds were grouped correctly, resulting in maximal diagnostic sensitivities (100%) and specificities (100%). For BTM samples, diagnostic sensitivities and specificities were 90 and 100%, respectively. Two herds were false negative in BTM, because cows with clinical signs of mastitis were not milked into the tank. Besides its excellent diagnostic properties, the assay is characterized by its low detection level, high efficiency, and its suitability for automation. Using the novel knowledge and assay, eradication of Staph. aureus GTB from a dairy herd may be considered as a realistic goal.  相似文献

Discovery on a protein electrophoregram of a bisalbuminemia can orientate according to its migration fast or slow to an hereditary mutation of an amino acid, or an acquired form by excess of beta lactamines due to renal insufficiency or by the rupture of a pancreatic pseudocyst in the peritoneum. The synthesis of a series (1–5) of fullerene and Zn(II)-porphyrin amphiphiles with polar dibenzo[24]crown-8 headgroups is described. Their ability to form Langmuir monolayers at the air-water interface was investigated in a systematic study. The Langmuir films were characterized by their surface pressure versus molecular area isotherms, compression and expansion cycles, and Brewsterangle microscopy. Complexation of larger alkali metal cations (K+ and Cs+) by the polar headgroups leads to higher molecular area requirements and a better anchoring to the aqueous subphase. The monolayers of the porphyrin-(dibenzo[24]crown-8) conjugates were transferred as Langmuir—Blodgett films onto glass slides and the films characterized by UV-vis spectroscopy and grazing-incidence X-ray diffraction. Good evidence was obtained that the porphyrin-fullerene-crown ether triad 5 adopts a sandwich geometry in the LB films. In such an arrangement, the electron-attracting carbon sphere benefits from attractive interactions with the sandwiching electron-rich porphyrin and crown ether chromophores. The described films could have potential applications as functional materials in optical and electronic technology.  相似文献
