The physicochemical properties of crude Nigella seed (Nigella sativa L.) oil which was extracted using Soxhlet, Modified Bligh–Dyer and Hexane extraction methods were determined. The effect of different extraction methods which includes different parameters, such as temperature, time and solvent on the extraction yield and the physicochemical properties were investigated. The experimental results showed that temperature, different solvents and extraction time had the most significant effect on the yield of the Nigella oil extracts. The fatty acid (FA) compositions of Nigella seed oil were further analyzed by gas chromatography to compare the extraction methods. The C16:0, C18:1 and C18:2 have been identified to be the dominant fatty acids in the Nigella seed oils. However, the main triacylglycerol (TAG) was LLL followed by OLL and PLL. The FA and TAG content showed that the composition of the Nigella seed oil extracted by different methods was mostly similar, whereas relative concentration of the identified compounds were apparently different according to the extraction methods. The melting and crystallization temperatures of the oil extracted by Soxhlet were −2.54 and −55.76 °C, respectively. The general characteristics of the Nigella seed oil obtained by different extraction methods were further compared. Where the Soxhlet extraction method was considered to be the optimum process for extracting Nigella seed oil with a higher quality with respect to the other two processes.  相似文献

A transesterified experimental solid frying shortening was prepared from a palm stearin/palm kernel olein blend at 1∶1 ratio (by weight) by using Rhizomucor miehei lipase at 60°C for 6 h. The fatty acid (FA) and triacylglycerol compositions, polymorphic forms, melting and cooling characteristics, slip melting point (SMP), and solid fat content (SFC) of the transesterified blend were then compared with five commercial solid frying shortenings (three domestic and two imported) found in Malaysia. All the domestic shortenings contained nonhydrogenated palm oil or palm olein and palm stearin as the hard stock, whereas the imported frying shortenings were formulated from soybean oil and cottonseed oil and contained high level of β′ crystals. Trans FA were also found in these samples. The lipase-transesterified blend was found to be more β′-tending than the domestic samples. The SMP of the transesterified blend (47.0°C) fell within the range of the domestic samples (37.8–49.7°C) but was higher than the imported ones (42.3–43.0°C). All samples exhibited similar differential scanning calorimetry cooling profiles, with a narrow peak at the higher temperatures and a broad peak at the lower temperatures, even though their heating thermograms were quite different. Imported samples had flatter SFC curves than both the experimental and domestic samples. The domestic samples were found to have better workability or plasticity at higher temperatures than the imported ones, probably because they were formulated for a tropical climate.  相似文献

Recently, the use of controlled release fertilizers in agriculture has resulted in huge benefits in plant growth and cultivation. Superabsorbent polymer (SAP)-coated fertilizers have the added advantage in retaining water in soil after irrigation and also reduce the nutrient release rate from soil in a controlled manner. This study aimed to produce a nitrogen–phosphorus–potassium (NPK) fertilizer coated with superabsorbent carbonaceous microspheres polymer (SPC) by inverse suspension polymerization method with water-retention and controlled release properties. Two sets of experiments were conducted: (1) three different weight percentages and (2) different materials. NPK coated with SPC showed increasing water-retention ability with respect to carbon microsphere percentages and retains >80% water at the 30th day of experiment compared with pure NPK and NPK coated with SAP. The slow release behavior of all samples was investigated by induced coupled plasma mass spectrometry spectrometry and results showed that NPK coated with SAP and SPC has a low release rate with <50% nutrient release compared with uncoated NPK at the 30th day. The release mechanism kinetics of NPK coated with SAP and SPC were studied based on the Kosmeyer–Peppas model. It has recently been shown that tocotrienols are the components of vitamin E responsible for inhibiting the growth of human breast cancer cells in vitro, through an estrogen-independent mechanism. Although tocotrienols act on cell proliferation in a dose-dependent manner and can induce programmed cell death, no specific gene regulation has yet been identified. To investigate the molecular basis of the effect of tocotrienols, we injected MCF-7 breast cancer cells into athymic nude mice. Mice were fed orally with 1 mg/d of tocotrienol-rich fraction (TRF) for 20 wk. At end of the 20 wk, there was a significant delay in the onset, incidence, and size of the tumors in nude mice supplemented with TRF compared with the controls. At autopsy, the tumor tissue was excised and analyzed for gene expression by means of a cDNA array technique. Thirty out of 1176 genes were significantly affected. Ten genes were down-regulated and 20 genes up-regulated with respect to untreated animals, and some genes in particular were involved in regulating the immune system and its function. The expression of the interferon-inducible transmembrane protein-1 gene was significantly up-regulated in tumors excised from TRF-treated animals compared with control mice. Within the group of genes related to the immune system, we also found that the CD59 glycoprotein precursor gene was up-regulated. Among the functional class of intracellular transducers/effectors/modulators, the c-myc gene was significantly down-regulated in tumors by TRF treatment. Our observations indicate that TRF supplementation significantly and specifically affects MCF-7 cell response after tumor formation in vivo and therefore the host immune function. The observed effect on gene expression is possibly exerted independently from the antioxidant activity typical of this family of molecules.  相似文献

This work proposes an effective method for dispersion of zirconia suspension for dental block preparation and optimizes the cold isostatic pressing (CIP) pressure to improve the densification of slip-casted zirconia blocks. Two batches of 44 wt% zirconia suspension were prepared using distilled water in a pH 2 medium containing 0.5 wt% polyethyleneimine as dispersant. The first batch was sonicated for different durations (from 5 min to 30 min), and the second batch was dispersed through ball milling at rotational speeds of 200, 300, and 400 rpm for 60, 90, and 120 min. All suspensions were subjected to sedimentation test and particle size measurement. Results revealed that the optimum ultrasonication duration was 10 min, which yielded the smallest particle size of 133 nm. Ball milling at 300 rpm for 120 min achieved the maximum dispersion of particles, with an average size of 75 nm. Under the optimum conditions of ultrasonication duration, ball milling duration, and ball milling speed, the particle size decreased to 48 nm, which is close to the primary particle size. These dispersion techniques and parameters were selected for preparing a suspension to be consolidated into blocks through slip casting and were enhanced through CIP at pressure ranging from 100 MPa to 300 MPa. CIP compaction at 250 MPa significantly increased the shrinkage percentage of green zirconia blocks, with pore radius decreased to 18 nm. The density of zirconia pressed at 250 MPa and presintered at a low temperature of 950 °C was 59% of the theoretical density and was higher than that of commercial presintered blocks. Thus, CIP should be conducted under a compaction pressure of 250 MPa to produce dense and homogeneous zirconia blocks.  相似文献

This article presents a mathematical model to assess and optimize the separation performance of an enantioselective inclusion complexation‐organic solvent nanofiltration process. Enantiomer solubilities, feed concentrations, solvent compositions, permeate solvent volumes, and numbers of nanofiltrations were identified as key factors for process efficiency. The model was first tested by comparing calculated and experimental results for a nonoptimized process, and then, calculations were carried out to select the best operating conditions. An important finding was that the optimal configuration varied with the objective function selected, e.g., resolvability versus yield, with a boundary on product optical purity. The model also suggested that the process efficiency could benefit from diafiltration of the distomer and from the use of higher feed concentrations. However, the latter strategy would result in higher losses of eutomer. Obesity and hyperlipidemia are major risk factors for developing vascular diseases. Bee bread (BB) has been reported to exhibit some biological actions, including anti-obesity and anti-hyperlipidemic. This study aims to investigate whether bee bread can ameliorate vascular inflammation and impaired vasorelaxation activity through eNOS/NO/cGMP pathway in obese rats. Forty male Sprague-Dawley rats were randomly divided into four groups (n = 10/group), namely: control (normal group), obese rats (OB group), obese rats treated with bee bread (0.5 g/kg/day, OB/BB group) and obese rats treated with orlistat (10 mg/kg/day, OB/OR group). The latter three groups were given a high-fat diet (HFD) for 6 weeks to induced obesity before being administered with their respective treatments for another 6 weeks. After 12 weeks of the total experimental period, rats in the OB group demonstrated significantly higher Lee obesity index, lipid profile (total cholesterol, triglyceride, low-density lipoprotein), aortic proinflammatory markers (tumor necrosis factor-α, nuclear factor-κβ), aortic structural damage and impairment in vasorelaxation response to acetylcholine (ACh). Bee bread significantly ameliorated the obesity-induced vascular damage manifested by improvements in the lipid profile, aortic inflammatory markers, and the impaired vasorelaxation activity by significantly enhancing nitric oxide release, promoting endothelial nitric oxide synthase (eNOS) and cyclic guanosine monophosphate (cGMP) immunoexpression. These findings suggest that the administration of bee bread ameliorates the impaired vasorelaxation response to ACh by improving eNOS/NO/cGMP-signaling pathway in obese rats, suggesting its vascular therapeutic role.  相似文献

The stability of nano-zirconia 3YSZ powder in suspension was extensively studied by the colloidal method, and the optimum sintering temperature of the green sample fabricated through slip casting was determined. Zirconia suspensions with 10 vol% powder loading were prepared with distilled water, and HNO3 was used to adjust the pH of the suspension to pH 1–6. All of the suspensions were subjected to sedimentation test, and the results showed that the suspensions adjusted to pH 2 had the lowest sediment volume. This finding indicates that a suspension with pH 2 produces higher packing density. Viscosity test was carried out for the suspensions added with dispersant ranging from 0.3 wt% to 0.7 wt% polyethyleneimine (PEI) with and without pH adjustment. The suspension containing 0.5 wt% PEI with pH 2 adjustment produced the lowest viscosity because of interparticle bond breakage in the aggregates, thus forming colloidally stable suspensions. The zirconia suspension containing 0.5 wt% PEI and whose pH was adjusted to pH 2 was chosen to be slip casted into cylindrical shape. Green samples were sintered at various sintering temperatures that ranged from 1100 °C to 1500 °C through a two-step sintering method. The sample sintered at 1500 °C was found to be porosite-free, and its highest relative density was 99.6% of the theoretical density. Morphological studies detected pores in the microstructure of the samples sintered at low sintering temperatures (1100 and 1200 °C). By contrast, the samples sintered at 1400 and 1500 °C were fully densified. However, the grain size of the sample sintered at 1500 °C was 230 nm, which indicated excessive grain growth. The Vickers hardness of the sample sintered at 1400 °C was found to be highest (12.9 GPa) and comparable to results found in the literature.  相似文献

Pullulanase (EC.3.2.1.41) was used to generate more linear-chain dextrin from sago starch (24.9% amylose) such that the resulting product could act as a high amylose starch. A starch suspension of 5.0% (w/v) sago starch was heated at 100 °C for 45 min and, after cooling, the gelatinized sago starch was hydrolyzed with 2.0% (v/dry weight starch) pullulanase (Promozyme 400L, Novozymes A/S, Denmark) for 24 h. The linear long-chain dextrin (LLD) content of the hydrolysate after drying, was then compared with the initial LLD content. The surface morphology of the starch granules was observed with a scanning electron microscope (SEM). The effects of gelatinization, time of reaction, pretreatment with different strengths of hydrochloric acid prior to enzyme hydrolysis, and starch and enzyme concentrations were studied. Raw sago starch was resistant to the action of pullulanase, but caused an increase in the LLD of that sago starch from an initial concentration of 24.9–33.2% following gelatinization. The best conditions to maximize the amount of LLD were 5.0% (w/v) sago starch, 2.0% (v/w) enzyme and 12 h reaction time. Acid pretreatment of the sago starch did not cause greater improvement in the accessibility and susceptibility of pullulanase as the LLD content, following pullulanase action did not change significantly. Shrinkage on the surface of the starch granules was observed with the SEM.  相似文献
