5-Fluoroanthranilic acid (FAA)-resistant mutants were selected in homothallic diploids of three Saccharomyces species, taking care to isolate mutants of independent origin. Mutations were assigned to complementation groups by interspecific complementation with S. cerevisiae tester strains. In all three species, trp3, trp4 and trp5 mutants were recovered. trp1 mutants were also recovered if the selection was imposed on a haploid strain. Thus, FAA selection may be more generally applicable than was previously described.  相似文献

The geomagnetic field (GMF) is an environmental factor affecting the mineral nutrient uptake of plants and a contributing factor for efficient iron (Fe) uptake in Arabidopsis seedlings. Understanding the mechanisms underlining the impact of the environment on nutrient homeostasis in plants requires disentangling the complex interactions occurring among nutrients. In this study we investigated the effect of GMF on the interplay between iron (Fe) and sulfur (S) by exposing Arabidopsis thaliana plants grown under single or combined Fe and S deficiency, to near-null magnetic field (NNMF) conditions. Mineral analysis was performed by ICP-MS and capillary electrophoresis, whereas the expression of several genes involved in Fe and S metabolism and transport was assayed by qRT-PCR. The results show that NNMF differentially affects (i) the expression of some Fe- and S-responsive genes and (ii) the concentration of metals in plants, when compared with GMF. In particular, we observed that Cu content alteration in plant roots depends on the simultaneous variation of nutrient availability (Fe and S) and MF intensity (GMF and NNMF). Under S deficiency, NNMF-exposed plants displayed variations of Cu uptake, as revealed by the expression of the SPL7 and miR408 genes, indicating that S availability is an important factor in maintaining Cu homeostasis under different MF intensities. Overall, our work suggests that the alteration of metal homeostasis induced by Fe and/or S deficiency in reduced GMF conditions impacts the ability of plants to grow and develop.  相似文献

Generation of the 3′ overhang is a critical step during homologous recombination (HR) and replication fork rescue processes. This event is usually performed by a series of DNA nucleases and/or helicases. The nuclease NurA and the ATPase HerA, together with the highly conserved MRE11/RAD50 proteins, play an important role in generating 3′ single-stranded DNA during archaeal HR. Little is known, however, about HerA-NurA function and activation of this fundamental and complicated DNA repair process. Herein, we analyze the functional relationship among NurA, HerA and the single-strand binding protein SSB from Saccharolubus solfataricus. We demonstrate that SSB clearly inhibits NurA endonuclease activity and its exonuclease activities also when in combination with HerA. Moreover, we show that SSB binding to DNA is greatly stimulated by the presence of either NurA or NurA/HerA. In addition, if on the one hand NurA binding is not influenced, on the other hand, HerA binding is reduced when SSB is present in the reaction. In accordance with what has been observed, we have shown that HerA helicase activity is not stimulated by SSB. These data suggest that, in archaea, the DNA end resection process is governed by the strictly combined action of NurA, HerA and SSB.  相似文献

This work proposes a fully BDD-based approach based on: mixing forward and backward traversals, dovetailing approximate and exact methods, adopting guided and partitioned searches, and using conjunctive decompositions and generalized-cofactor-based BDD simplifications. The method is exact, i.e., it does not produce false negatives or positives, and reaps relevant performance enhancements from an appropriate tradeoff among the component methodologies.The resulting verification strategy is able, on one hand, to improve standard BDD-based algorithms, and, on the other hand, to compete with SAT-based tools on a broader range of circuit sizes.We experimentally compare our approach with three state-of-the-art freely available techniques. The first and second ones are SAT-based strategies, i.e., bounded model checking implemented within the NuSMV and the VIS tools (using Chaff as SAT engine). The third one is a BDD-based methodology, i.e., approximate reachability dont cares model checking implemented within the VIS tool. Results show interesting improvements in terms of both efficiency and power. They demonstrate how BDDs are able to accomplish larger verification tasks and to efficiently deal with high sequential depths.  相似文献

Greenhouse farming, where energy consumptions are mainly related to the greenhouses heating, is one of the sectors consuming the most energy in the agricultural industry. High costs and the uncertain availability of fossil fuels constrain the use of heating applications. Among possible solutions, the utilization of renewable heating systems such as geothermal energy through ground-source heat pump systems (GSHPs) at competitive prices has to be taken in consideration. The competitiveness of these systems depends mainly on the characteristics of the end-users, i.e., the annual heating loads. Few studies focusing on the potential of using these systems start with an analysis of the thermal requirements and end with a cost evaluation in tune with local assets, geo-climatic conditions, and landscape protection. This paper analyzes the greenhouse crop industry in the Apulia region in southern Italy, as a potential end-user of GSHP systems. Data collected from an area mainly devoted to greenhouse crop production have been used to (a) describe greenhouse farms, (b) define the heating requirements of a greenhouse model representative of the most used typology in the investigated area, and (c) examine the economic viability of greenhouse heating with GSHP systems. Both vertical and horizontal ground heat exchanger (GHE) configurations are compared with conventional fossil-fuel heating systems. In all scenarios considered, the observed payback periods appear reasonable and worthy of consideration. The results suggest that these technologies can fully satisfy the winter heating requirements in a cost-effective way and they can support the planning of measures aimed to improve the sector competitiveness.  相似文献

Progressive multifocal leukoencephalopathy is a demyelinating disease of the human central nervous system that results from lytic infection of oligodendrocytes by the polyomavirus JC (JCV). Originally, JCV was thought to replicate exclusively in human glial cells, specifically oligodendrocytes. However, we have recently shown that JCV can replicate in cells of lymphoid origin such as hematopoietic precursor cells, B lymphocytes, and tonsillar stromal cells. To determine whether tonsils harbor JCV, we tested a total of 54 tonsils, 38 from children and 16 from adult donors. Nested PCRs with primer sets specific for the viral T protein and regulatory regions were used for the detection of JCV DNA. JCV DNA was detected in 21 of 54 tonsil tissues, or 39% (15 of 38 children and 6 of 16 adults) by using regulatory-region primers and in 19 of 54 tonsil tissues, or 35% (13 of 38 children and 6 of 16 adults) by using the T-protein primers. The DNA extracted from children's nondissected tonsil tissue, isolated tonsillar lymphocytes, and isolated stromal cells that demonstrated PCR amplification of the JCV regulatory region underwent cloning and nucleotide sequencing. Of the regulatory-region sequences obtained, nearly all contained tandem repeat arrangements. Clones originating from nondissected tonsil tissue and tonsillar lymphocytes were found to have sequences predominantly of the Mad-1 prototype strain, whereas the majority of clones from the DNA of tonsillar stromal cells had sequences characteristic of the Mad-8br strain of JCV. A few clones demonstrated structures other than tandem repeats but were isolated only from tonsillar lymphocytes. These data provide the first evidence of the JCV genome in tonsil tissue and suggest that tonsils may serve as an initial site of viral infection.  相似文献
