Step changes in length (between -3 and +5 nm per half-sarcomere) were imposed on isolated muscle fibers at the plateau of an isometric tetanus (tension T0) and on the same fibers in rigor after permeabilization of the sarcolemma, to determine stiffness of the half-sarcomere in the two conditions. To identify the contribution of actin filaments to the total half-sarcomere compliance (C), measurements were made at sarcomere lengths between 2.00 and 2.15 microm, where the number of myosin cross-bridges in the region of overlap between the myosin filament and the actin filament remains constant, and only the length of the nonoverlapped region of the actin filament changes with sarcomere length. At 2.1 microm sarcomere length, C was 3.9 nm T0(-1) in active isometric contraction and 2.6 nm T0(-1) in rigor. The actin filament compliance, estimated from the slope of the relation between C and sarcomere length, was 2.3 nm microm(-1) T0(-1). Recent x-ray diffraction experiments suggest that the myosin filament compliance is 1.3 nm microm(-1) T0(-1). With these values for filament compliance, the difference in half-sarcomere compliance between isometric contraction and rigor indicates that the fraction of myosin cross-bridges attached to actin in isometric contraction is not larger than 0.43, assuming that cross-bridge elasticity is the same in isometric contraction and rigor.  相似文献

In the 1st session of each of 2 studies, 48 undergraduates' accessible traits were elicited by asking them to list the characteristics of different people, with accessibility defined as frequency of output (Study 1) or primacy of output (Study 2). In the 2nd session, held 1 or 2 wks later, Ss read an essay describing the behaviors of a target person. The essay contained both accessible and inaccessible trait-related information for each S, with different traits being accessible or inaccessible for different Ss. Both studies found that Ss deleted significantly more inaccessible than accessible trait-related information in their impressions and reproductions of the target information. This effect on impressions and reproductions was evident 2 wks after exposure to the target information. Implications of this approach for personality differences, interpersonal conflict and attraction, similarity of self and other judgments, and therapeutic intervention are discussed. Metabolomics and systems biology require the acquisition of reproducible, robust, reliable, and homogeneous biological data sets. Therefore, we developed and validated standard operating procedures (SOPs) for quenching and efficient extraction of metabolites from Escherichia coli to determine the best methods to approach global analysis of the metabolome. E. coli was grown in chemostat culture so that cellular metabolism could be held in reproducible, steady-state conditions under a range of precisely defined growth conditions, thus enabling sufficient replication of samples. The metabolome profiles were generated using gas chromatography/time-of-flight mass spectrometry (GC/TOF-MS). We employed univariate and multivariate statistical analyses to determine the most suitable method. This investigation indicates that 60% cold (-48 degrees C) methanol solution is the most appropriate method to quench metabolism, and we recommend 100% methanol, also at -48 degrees C, with multiple freeze-thaw cycles for the extraction of metabolites. However, complementary extractions would be necessary for coverage of the entire complement of metabolites as detected by GC/TOF-MS. Finally, the observation that metabolite leakage was significant and measurable whichever quenching method is used indicates that methods should be incorporated into the experiment to facilitate the accurate quantification of intracellular metabolites.  相似文献

Previous research has investigated age differences in complex social perception tasks such as theory of mind and emotion recognition, with predominant findings of age-related declines. The present study investigated whether there are also age-related changes in basic aspects of social perception. Individuals' ability both to detect subtle differences in eye-gaze direction (e.g., where someone is looking in the environment) and to subsequently use these gaze cues to engage in joint attention with others was assessed. Age-related declines were found in the detection of the most subtle differences in gaze aversion. The ability to engage in joint attention by following gaze cues also declined with age. These age differences were not solely attributable to age impairments in visual perception and visual attention. The potential role of age-related neural declines in social perception problems was considered, along with the implications that age deficits in these basic social skills may have for older adults' social perception. Organochlorine pesticides in the soils and atmosphere of Costa Rica   总被引:3,自引:0,他引:3  
A survey of the contamination of the physical environment of Costa Rica with banned organochlorine pesticides (OCPs) relied on sampling air and soil at 23 stations acrossthe country in 2004. Average annual air concentrations, determined with XAD-based passive samplers, and surface soil concentrations were generally low when compared to values reported for North and Central America, which is consistent with relatively low historical domestic use and little atmospheric inflow from neighboring countries. Statistical analysis and concentration maps reveal three types of spatial distribution: alpha-hexachlorocyclohexane and p,p'-DDD had a relatively uniform distribution across the country; other DDT-related species were greatly elevated over the national average at Manuel Antonio, a National Park on the Pacific coast; and dieldrin, lindane, and chlordane-related species had higher concentrations in Costa Rica's populated Central Valley. An altitudinal transect of stations in the Central Valley shows declining air-soil concentration ratios with elevation for lindane, likely driven by atmospheric inversions and soil organic carbon content. Enantiomeric composition of chiral OCPs in air and soil was close to racemic, with slight depletion of (-)-alpha-HCH, (-)-cis-chlordane, and (+)-trans-chlordane. Estimated air-soil fugacity fractions are highly uncertain but indicate equilibrium conditions for most OCPs, net volatilization of lindane at some sites, and net deposition for p,p'-DDE. The study demonstrates an approach for quickly evaluating the spatial distribution of OCPs in an understudied area, identifying regionally important contaminants and areas of elevated concentrations.  相似文献

With a rapidly aging population, strategies for improving driver safety are beginning to emerge that focus on changing driving behaviors and knowledge. We examined the perceptions of risk, beliefs and attitudes, and openness to change of 86 older participants voluntarily attending a driver education program. It appeared that most people attending these sessions were not necessarily concerned about their own driving, safety or abilities, but were interested in maintaining mobility. They were conservative and reasonably consistent in their attitudes toward traffic regulations and safe driving practices. Some gender differences emerged with more men than women being resistant to changing their driving habits, more men than women reporting that they drive after consuming alcohol and more women than men identifying a role for their families in decision-making regarding driving cessation. This suggests that educational material may need to be targeted differently for men and women. It is anticipated that psychosocial factors related to driving such as driver perception, beliefs and openness to change will be useful for maximizing the fit between education program content and outcomes.  相似文献

Three fluorescence signaling DNA enzymes (deoxyribozymes or DNAzymes) were successfully immobilized within a series of sol-gel-derived matrixes and used for sensing of various metal ions. The DNAzymes are designed such that binding of appropriate metal ions induces the formation of a catalytic site that cleaves a ribonucleotide linkage within a DNA substrate. A fluorophore (fluorescein) and a quencher (DABCYL, [4-(4-dimethylaminophenylazo)benzoic acid]) were placed on the two deoxythymidines flanking the ribonucleotide to allow the generation of fluorescence upon the catalytic cleavage at the RNA linkage. In general, all DNAzymes retained at least partial catalytic function when entrapped in either hydrophilic or hydrophobic silica-based materials, but displayed slower response times and lower overall signal changes relative to solution. Interestingly, it was determined that maximum sensitivity toward metal ions was obtained when DNAzymes were entrapped into composite materials containing approximately 40% of methyltrimethoxysilane (MTMS) and approximately 60% tetramethoxysilane (TMOS). Highly polar materials derived from sodium silicate, diglycerylsilane, or TMOS had relatively low signal enhancements, while materials with very high levels of MTMS showed significant leaching and low signal enhancements. Entrapment into the hybrid silica material also reduced signal interferences that were related to metal-induced quenching; such interferences were a significant problem for solution-based assays and for polar materials. Extension of the solid-phase DNAzyme assay toward a multiplexed assay format for metal detection is demonstrated, and shows that sol-gel technology can provide new opportunities for the development of DNAzyme-based biosensors.  相似文献

Solvent-cast films from three polymers, carboxymethylcellulose (CMC), sodium alginate (SA), and xanthan gum, were prepared by drying the polymeric gels in air. Three methods, (a) passive hydration, (b) vortex hydration with heating, and (c) cold hydration, were investigated to determine the most effective means of preparing gels for each of the three polymers. Different drying conditions [relative humidity – RH (6–52%) and temperature (3–45°C)] were investigated to determine the effect of drying rate on the films prepared by drying the polymeric gels. The tensile properties of the CMC films were determined by stretching dumbbell-shaped films to breaking point, using a Texture Analyser. Glycerol was used as a plasticizer, and its effects on the drying rate, physical appearance, and tensile properties of the resulting films were investigated. Vortex hydration with heating was the method of choice for preparing gels of SA and CMC, and cold hydration for xanthan gels. Drying rates increased with low glycerol content, high temperature, and low relative humidity. The residual water content of the films increased with increasing glycerol content and high relative humidity and decreased at higher temperatures. Generally, temperature affected the drying rate to a greater extent than relative humidity. Glycerol significantly affected the toughness (increased) and rigidity (decreased) of CMC films. CMC films prepared at 45°C and 6% RH produced suitable films at the fastest rate while films containing equal quantities of glycerol and CMC possessed an ideal balance between flexibility and rigidity.  相似文献
