Nonthermal technologies are becoming more popular in food processing; however, little detailed research has been conducted on the study of the lethal effect of these technologies on certain microorganisms. Saccharomyces cerevisiae is a yeast related to spoilage of fruit products such as juices; novel technologies have been explored to inactivate this yeast. Three nonthermal technologies, high hydrostatic pressure (HHP), pulsed electric fields (PEF) and thermo-sonication (TS), were used to evaluate and to compare the structural damage of yeast cells after processing. Processing conditions were chosen based on previous experiments to ensure the death of cells; HHP was conducted at 600 MPa for 7 min (room temperature, 21 °C); for PEF, 30.76 kV/cm at 40 °C and 21 pulses (2 μs each), and finally for TS the conditions were 120 μm, 60 °C and 30 min in continuous and pulsed modes; all treatments were applied in apple juice. Cells were prepared for electron microscopy using an innovative and short microwave assisted dehydration technique. Scanning electron microscopy showed the degree of damage to the cells after processing and illustrated the important and particular characteristics of each technology. Cells treated with high hydrostatic pressure showed a total disruption of the cell membrane, perforation, and release of the cell wall; scars were also observed on the surface of the pressurized cells. PEF treated cells showed less superficial damage, with the main changes being the deformation of the cells, apparent fusion of cells, the formation of pores, and the breakdown of the cell wall in some cells. Finally, the thermo-sonicated cells showed a similar degree of cellular damage to their structure regardless of whether the TS was applied continuously or pulsed. The main characteristics of cellular death for this technology were the erosion and disruption of the cellular membrane, formation of orifices on the surface, lysis of cells causing the release of intracellular contents, roughness of the cell membrane, and displacement of cell debris to the surface of other cells. This study confirms some theories about cell inactivation and presents new and detailed results about nonthermal technologies, but also shows that after using the above mentioned conditions, recovery of cells, specifically those that are pressurized and thermo-sonicated, it is not possible to do it following the high extent of damage observed in the entire population. Furthermore, a faster methodology that was used in sample preparation for electron microscopy provided high quality resolution images, allowing closer study of the detail of structural lethal effects on treated cells.  相似文献

The last several decades have seen decreases in SO(4)(2-) deposition across the northeastern United States. As a result, SO(4)(2-) concentrations in lakes and streams have also decreased and many surface water bodies have become less acidic. During the same time period, there has been a concurrent increase in dissolved organic carbon (DOC) concentrations in many lakes and streams. We used fluorescence spectroscopy to characterize the dissolved organic matter (DOM) quality of archived samples from nine acid-sensitive lakes in Maine collected between 1993 and 2009, and determined that increased DOM contributions to lakes were primarily derived from litter and soil. All five lakes with increasing DOC trends demonstrated significant decreasing (i.e., more terrestrial) trends in fluorescence index (FI) and significant positive correlations between SO(4)(2-) and FI. This study used the chemical signature of terrestrial DOM to support the hypothesis that increased DOC concentrations in lakes and streams are driven by declining acid deposition and increased solubility of soil organic matter across a large area of the landscape.  相似文献

Red band bidirectional reflectance factor data from the NASA MODerate resolution Imaging Spectroradiometer (MODIS) acquired over the southwestern United States were interpreted through a simple geometric-optical (GO) canopy reflectance model to provide maps of fractional crown cover (dimensionless), mean canopy height (m), and aboveground woody biomass (Mg ha− 1) on a 250 m grid. Model adjustment was performed after dynamic injection of a background contribution predicted via the kernel weights of a bidirectional reflectance distribution function (BRDF) model. Accuracy was assessed with respect to similar maps obtained with data from the NASA Multiangle Imaging Spectroradiometer (MISR) and to contemporaneous US Forest Service (USFS) maps based partly on Forest Inventory and Analysis (FIA) data. MODIS and MISR retrievals of forest fractional cover and mean height both showed compatibility with the USFS maps, with MODIS mean absolute errors (MAE) of 0.09 and 8.4 m respectively, compared with MISR MAE of 0.10 and 2.2 m, respectively. The respective MAE for aboveground woody biomass was ~ 10 Mg ha− 1, the same as that from MISR, although the MODIS retrievals showed a much weaker correlation, noting that these statistics do not represent evaluation with respect to ground survey data. Good height retrieval accuracies with respect to averages from high resolution discrete return lidar data and matches between mean crown aspect ratio and mean crown radius maps and known vegetation type distributions both support the contention that the GO model results are not spurious when adjusted against MISR bidirectional reflectance factor data. These results highlight an alternative to empirical methods for the exploitation of moderate resolution remote sensing data in the mapping of woody plant canopies and assessment of woody biomass loss and recovery from disturbance in the southwestern United States and in parts of the world where similar environmental conditions prevail.  相似文献

We prove that a group G has a word problem that is accepted by a deterministic counter automaton with a weak inverse property if and only if G is virtually abelian. We extend this result to larger classes of groups by considering a generalization of finite state automata, counter automata and pushdown automata. Natural corollaries of our general result include a restricted version of Herbst's classification of groups for which the word problem is a one counter language and a new classification of automata that accept context-free word problems.  相似文献

There has been relatively little research on the role of grandparents as a source of support for children during and following their parents' marital transitions. In this study, we examined children's contact with and closeness to grandparents in different family types (i.e., two biological parents, single mother, stepparent). Participants included 155 children from the Avon Brothers and Sisters Study. Parent and child interviews and questionnaires regarding the children's relationships with maternal and paternal biological and stepgrandparents were examined. There were family type differences in rates of contact with grandparents as well as children's closeness to grandparents. Furthermore, children's and parents' view about these relationships with grandparents were modestly correlated, suggesting that children often held different views about their closeness to their grandparents than did their parents. Applies the criteria in the Procedural and Coding Manual for Review of Evidence-Based Interventions, established by the Task Force on Evidence-Based Interventions (EBIs) in School Psychology, to a group-based design intervention study. Specifically, the School Psychology EBI group-based criteria were applied to a research study conducted by the Conduct Problems Prevention Research Group (CPPRG, 1999a, 1999b) that examined the effects of the Fast Track intervention program on Grade 1 students. The group-based coding criteria are explicated and described through demonstrating the basis for coding decisions. In general, the application of the Procedural and Coding Manual criteria to the CPPRG investigation of the Fast Track program suggested promising evidence in support of the program. Considerations for interpreting the results of the coding process are discussed with particular attention to implications for practitioners and researchers. Large river systems create challenges when sampling zooplankton. The dynamics of large rivers, spatial heterogeneity of biota, and behaviours of zooplankton can all contribute to large variation in estimation. The gear utilized for zooplankton collections may also be a factor, and there is not a well‐studied gear, nor industry standard, for collection in riverine systems. The lack of consistent sampling methods makes it difficult to compare zooplankton within and across systems. A standard method for zooplankton collection would be valuable for comparison among studies. To optimize zooplankton assessment in large river habitats, we tested how community estimations varied between four common zooplankton sampling devices across three river habitat types. We tested four gears: the Schindler–Patalas trap, integrated tube sampler, powered water pump, and horizontal tow net. Each device was used to collect samples in thalweg, channel border, and backwater river habitats within Pool 8 of the Upper Mississippi River in June, July, and August of 2017. Our results support that there are qualitative and quantitative differences in zooplankton estimates among gear in different habitats. The powered pump most often yielded highest abundances of total and individual zooplankton taxa allowing for more reliable community comparisons. With some modification, the pump is recommended as the most appropriate sampling gear when performing quantitative studies of zooplankton composition and abundance in large river habitats.  相似文献

There is a positive relationship between student participation in computer-supported collaborative learning (CSCL) environments and improved complex problem-solving strategies, increased learning gains, higher engagement in the thinking of their peers, and an enthusiastic disposition toward groupwork. However, student participation varies from group to group, even in contexts where students and teachers have had extensive training in working together. In this study, we use positioning theory and interaction analysis to conceptualize and investigate relationships between student interactions across two partner pairs working with technology in an all-female cryptography summer camp and their negotiated positions of power and status. The analysis resulted in uneven participation patterns, unequal status orderings, and an imbalance of power in both comparison cases. We found a reflexive relationship between partner interactions around shared technology resources and negotiated positions of power and status, which leads us to conclude that interactions around technology function as an important indicator of negotiated positionings of power and status in CSCL settings, and vice-versa. With that said, we found qualitative differences in the ways emergent status problems impacted each team’s productivity with the cryptography challenge, which has important implications for future research on CSCL settings and classroom practice.  相似文献
