Saccharomyces cerevisiae Sly1 protein is a member of the Sec1/Munc18-family proteins, which are essential for vesicular trafficking, but their exact biological roles are yet to be determined. A temperature-sensitive sly1 mutant arrests the vesicular transport from the ER to Golgi compartments at 37 degrees C. We screened for multicopy suppressor genes that restore the colony formation of the sly1(ts) mutant to discover functionally interacting components. The suppressor genes obtained were classified as: (1) those that encode a multifunctional suppressor, SSD1; (2) heat shock proteins, SSB1 and SSB2; (3) cell surface proteins, WSC1, WSC2 and MID2; (4) ER-Golgi transport proteins, USO1 and BET1; and (5) an as-yet-uncharacterized protein, HSD1 (high-copy suppressor of SLY1 defect 1). By epitope tagging of the gene product, we found that Hsd1 protein is an ER-resident membrane protein. Its overproduction induced enlargement of ER-like membrane structures.  相似文献

Effective thermal conductivity of sintered spherical particles is estimated by a computer simulation. The simulation consists of (i)simulated random packing of equal spheres by a method of "rigid sphere free fall into a virtual box," (ii) finite element method (FEM) estimation of the thermal resistance of a "sintered" pair of spheres, and (iii) simulated heat conduction tests of a "random network," as a model of sintered particles, of thermal resistors with the estimated resistance; these tests yield the effective conductivity of sintered spherical particle aggregates. Statistical structures of the random packings of spheres are examined. The random packings constructed are standard "loose random packings." The cumulative diameter distribution of circles appearing on cross-sections of the packings is in complete agreement with the theoretical prediction for all three orthogonal directions, implying that the packing structures are isotropic. And, despite this result, the zenithal distribution of branch orientations deviates from a uniform one; more spheres are in contact with their neighbors at the zenithal angles of theta approxequal 45 degrees. The effective conductivities obtained are not isotropic but transversely isotropic, which is due to the deviation of branch orientations.  相似文献

Mechanical recycling of plastics from waste from electrical and electronical equipment (WEEE) is increasingly expected by regulators and demanded by original equipment manufacturers (CEMs); however, mechanical recycling is generally recognized to be the most economically costly and technically challenging method of recovering WEEE plastics. With 12% of WEEE plastics requiring the use of flame-retardants in order to ensure appropriate levels of consumer fire safety, there is a distinct need for data from comparative tests on recyclability of various flame-retarded plastics. Ten commercially available flame-retarded plastic grades commonly used in electronic equipment (eight "halogen-free" grades and two grades containing brominated flame-retardants (BFRs)) were subjected to two different recycling scenarios. A standard recycling scenario was carried out by repeatedly extruding the materials and an accelerated hydrolysis scenario was carried out to study the influence of humidity from air during use on the process. Both, virgin and recycled materials were tested for a potential formation of polybrominated dibenzodioxins/furans (PBDD/Fs), their mechanical properties were assessed and the fire safety rating was determined. Results indicate that none of the tested materials showed a potential to form the PBDD/Fs regulated by the German Chemicals Banning Ordinance. The halogen-free plastic grades showed a significant deterioration of mechanical properties after recycling, whereas those plastics containing BFRs were able to pass all test criteria, thus maintaining their original properties. With respect to the fire safety rating, none of the eight tested halogen-free plastic grades could maintain their fire safety rating after five recycling loops, whereas both BFR plastics continued to achieve their fire safety ratings. Therefore the tested BFR containing plastic materials showed superior recycling properties compared to the tested halogen-free plastic grades with respect to all investigated parameters.  相似文献

A new reactor concept of innovative water reactor for flexible fuel cycle (FLWR) is under development at Japan Atomic Energy Agency in cooperation with Japanese reactor suppliers. A design of 1,356 MWe high conversion boiling water reactor-type FLWR core, which has an instantaneous conversion ratio of 1.04, negative void coefficient, high burnup of 65 GWd/t, and 15-month operational cycle length, has been constructed. So far, studies on thermal-hydraulic characteristics have been performed for tight lattice core. Evaluation methods for the critical power and the pressure drop under both the steady and the transient states have been established, and a modified TRAC-BF1 code has been developed for the thermal-hydraulic design of the FLWR. In this paper, the thermal feasibility of the designed 1356MWe FLWR core is analyzed by using the modified TRAC-BF1 code. The analysis is first carried out for the current core design. It is confirmed that no boiling transition (BT) occurs under the steady state. However, the minimum critical power ratio (MCPR) is only about 1.08, and the BT is confirmed occurring under the postulated abnormal transient processes. Therefore, concretizations of the conditions that ensure the thermal feasibility of a natural circulation-type FLWR and a forced circulation-type FLWR are performed. As for the results, for a forced circulation-type FLWR, the operation-limited MCPR (OLMCPR) is 1.32, and the necessary minimum core coolant flow rate is 640 kg/(m2s). For a natural circulation-type FLWR, the OLMCPR is 1.19, and the necessary minimum core coolant flow rate is 560 kg/(m2s).  相似文献

To clarify the effects of food sediments on ultraviolet-C (254 nm) sanitation in food-related environments, we examined the resistance of pathogenic bacteria (Salmonella Typhimurium and Staphylococcus aureus) cells, in wet and dried suspensions adhered with 1.5-15% w/v egg albumen, 1.5-15% yolk or 3.0-30% whole egg solutions, against UV-C irradiation. Bacterial suspensions (0.1 ml of 8 log CFU/ml) were put on 47 mmφ glass dishes and dried at room temperature (20-24 °C) for 180 min in a bio safety cabinet with ventilation. Viable S. Typhimurium and S. aureus cells in distilled water decreased during the drying period from 7.2 to 3.2 and from 8.0 to 6.5 log CFU/dish, respectively,. On the other hand, the bacteria cells were protected from drying by egg compounds, even by the lowest concentration. The UV-C treatment (0.16 mW/cm2 for 10 min) showed a clear bactericidal effect in the absence of egg compounds. However, the bactericidal effect was inhibited by 15% yolk and 30% whole egg. Results in this study suggested that the small food sediment protect bacteria on the surfaces from dryness and UV-C irradiation and it might introduce cross contamination.  相似文献

Solid electrolyte interface (SEI) films formed on Li1−xCoO2 electrodes were observed with hard X-ray photoelectron spectroscopy (HX-PES). This paper particularly focuses on film thickness estimation using HX-PES with theoretical calculation. The validity of the calculation was proven by experiments using model SEI films. The native film formed on a LiCoO2 composite electrode was estimated to be LiF with its thickness of 5 nm. Formation of Co (II) species on top of LiCoO2 was also indicated. Storage of the electrode at 60 °C brought about considerable film growth (30-40 nm) with carbonate compounds formation. SEI film changes during charging of the LiCoO2 electrode were also examined. The main component in the film was deduced to be LiF or a kind of fluorite, with its thickness decreased during charging. The SEI formation mechanisms are also elucidated.  相似文献

Biohydrogen production process from glucose using extreme-thermophilic H2-producing bacteria enriched from digested sewage sludge was investigated for five cycles of repeated batch experiment at 70 °C. Heat shock pretreatment was used for preparation of hydrogen-producing bacteria comparing to an untreated anaerobic digested sludge for their hydrogen production performance and responsible microbial community structures. The results showed that the heat shock pretreatment completely repressed methanogenic activity and gave the maximum hydrogen production yield of 355-488 ml H2/g COD in the second cycle of repeated batch cultivation with more stable gas production during the cultivation when compared with control. Hydrogen production was accompanied by production of acetic acid. The average specific hydrogen in five cycles experiment ranged from 150 to 200 ml H2/g VSS. PCR-DGGE profiling showed that the extreme-thermophilic culture predominant species were closely affiliated to Thermoanaerobacter pseudethanolicus.  相似文献

Food processing environments are suspected to be the primary source of Listeria monocytogenes contamination of foods. We investigated the survival of L. monocytogenes on stainless steel that was soiled with food components (minced tuna, ground pork, and cabbage) followed by dehydration. Stainless steel coupons were inoculated with 107 CFU/coupon and stored up to 30 days. In comparison with Staphylococcus aureus and Salmonella Typhimurium, L. monocytogenes demonstrated the highest survivability where about 3 log CFU/coupon remained at the end of the experimental period. While survival during the early storage period was markedly affected by both food-component attachment and food type, a long dehydration period diminished the effect of those.  相似文献

The glucosyl transfer reaction of kojibiose phosphorylase (KP; EC 2.4.1.230) was examined using glycerol or myo-inositol as an acceptor. In the case of glycerol, KP produced two main transfer products: saccharides A and B. The structure of saccharide A was O-alpha-D-glucopyranosyl-(1-->1)-glycerol and that of saccharide B was O-alpha-D-glucopyranosyl-(1-->2)-O-alpha-D-glucopyranosyl-(1-->1)-glycerol. These results show that KP transferred a glucose residue to the hydroxyl group at position 1 of glycerol. On the other hand, when myo-inositol was used as an acceptor, KP produced four transfer products: saccharides 1-4. The structures of saccharides 1 and 2 were O-alpha-D-glucopyranosyl-(1-->1)- and O-alpha-D-glucopyranosyl-(1-->5)-myo-inositol, respectively; those of saccharides 3 and 4 were O-alpha-D-glucopyranosyl-(1-->2)-O-alpha-D-glucopyranosyl-(1-->1)- and O-alpha-D-glucopyranosyl-(1-->2)-O-alpha-D-glucopyranosyl-(1-->5)-myo-inositol, respectively. KP transferred a glucose residue to the hydroxyl group at position 1 or 5 of myo-inositol. On the basis of the structures of their glucosyl transfer products, glycerol and myo-inositol were found to have a common structure with three hydroxyl groups corresponding to the hydroxyl group of the glucose molecule at positions 2, 3 and 4. The conformation of these three hydroxyl groups in the structure is equatorial. This structure is the substrate recognition site of KP. It has been suggested that KP strictly recognizes the structures of glycerol and myo-inositol, and catalyzes the transfer reaction of a glucose residue to the hydroxyl group at position 1 in glycerol, and at position 1 or 5 in myo-inositol, corresponding to position 2 in glucose.  相似文献
