Transient complexes are crucial for diverse biological processes such as biochemical pathways and signaling cascades in the cell. Here, we give an overview of the transient interactions; the importance of transient interactions as drug targets; and the structural characterization of transient protein-protein complexes based on the geometrical and physicochemical features of the transient complexes' interfaces. To better understand and eventually design transient protein-protein interactions (TPPIs), a molecular perspective of the protein-protein interfaces is necessary. Obtaining high-quality structures of protein-protein interactions could be one way of achieving this goal. After introducing the association kinetics of TPPIs, we elaborate on the experimental techniques detecting TPPIs in combination with the computational methods which classify transient and/or non-obligate complexes. In this review, currently available databases and servers that can be used to identify and predict TPPIs are also compiled.  相似文献

Circulating nucleic acids (CNAs) are under investigation as a liquid biopsy in cancer as potential non-invasive biomarkers, as stable structure in circulation nucleosomes could be valuable sources for detection of cancer-specific alterations in histone modifications. Our interest is in histone methylation marks with a focus on colorectal cancer, one of the leading cancers respective the incidence and mortality. Our previous work included the analysis of trimethylations of lysine 9 on histone 3 (H3K9me3) and of lysine 20 on histone 4 (H4K20me3) by chromatin immuno- precipitation-related PCR in circulating nucleosomes. Here we asked whether global immunologic measurement of histone marks in circulation could be a suitable approach to show their potential as biomarkers. In addition to H3K9me3 and H4K20me3 we also measured H3K27me3 in plasma samples from CRC patients (n = 63) and cancer free individuals (n = 40) by ELISA-based methylation assays. Our results show that of three marks, the amounts of H3K27me3 (p = 0.04) and H4K20me3 (p < 0.001) were significantly lower in CRC patients than in healthy controls. For H3K9me3 similar amounts were measured in both groups. Areas under the curve (AUC) in receiver operating characteristic (ROC) curves indicating the power of CRC detection were 0.620 for H3K27me3, 0.715 for H4K20me3 and 0.769 for the combination of both markers. In conclusion, findings of this preliminary study reveal the potential of blood-based detection of CRC by quantification of histone methylation marks and the additive effect of the marker combination.  相似文献

The effects of halogen lamp–microwave combination baking on the quality of cookies in terms of texture, color and spread ratio were studied. In addition, gelatinization and pasting properties of cookies baked in different ovens were assessed by using a Rapid Visco Analyzer. The hardness values of the cookies increased with increasing baking time and/or halogen power. The microwave power also contributed to cookie hardness. The lightness values of the cookies decreased, while the a* and E values increased linearly during baking at different halogen lamp and microwave powers. The spread ratio of conventionally baked cookies was significantly lower than that of the other cookies. Halogen lamp power, microwave power and baking time were found to have a significant effect on the spread ratio of the cookies. The moisture contents of the cookies decreased during baking as the halogen lamp and/or the microwave power level increased. The best baking condition in a halogen lamp–microwave combination oven to produce cookies having similar quality parameters as conventionally baked ones was baking at 70% halogen lamp and 20% microwave power levels for 5.5 min. The baking time of these cookies is half of that required in conventional baking.  相似文献

In this paper, we present our system design, operational procedure, testing process, field results, and lessons learned for the valve-turning task of the DARPA Robotics Challenge (DRC). We present a software framework for cooperative traded control that enables a team of operators to control a remote humanoid robot over an unreliable communication link. Our system, composed of software modules running on-board the robot and on a remote workstation, allows the operators to specify the manipulation task in a straightforward manner. In addition, we have defined an operational procedure for the operators to manage the teleoperation task, designed to improve situation awareness and expedite task completion. Our testing process, consisting of hands-on intensive testing, remote testing, and remote practice runs , demonstrates that our framework is able to perform reliably and is resilient to unreliable network conditions. We analyze our approach, field tests, and experience at the DRC Trials and discuss lessons learned which may be useful for others when designing similar systems.  相似文献

Variations in pore structure cause significant changes in diffusion resistance and adsorption characteristics during the reaction of SO2 with activated soda. Breakthrough and moment analysis of the SO2-activated soda ash reaction showed that the effective diffusion coefficient of SO2 in the solid product (mostly Na2SO3) was about three times smaller than its value in the original activated soda. About 30% of the diffusion flux of SO2 in the solid product was found to be due to surface diffusion. The adsorption equilibrium constant of SO2 on the solid product was found to be about half the value of the adsorption equilibrium constant on activated soda. It was also determined that SO2 was irreversibly adsorbed on the solid product Na2SO3 at 200°C and 3.7 atm. The gaseous product CO2 was not adsorbed on the solid product while it was slightly adsorbed on activated soda.  相似文献

The present work proposes to fabricate a composite hydrogel material that well characterized, transparent, biocompatible, and self‐antibacterial as potential soft contact lens material. For this purpose, poly(2‐hydroxyethyl methacrylate) (PHEMA)/boric acid (BA) composite hydrogels were successfully prepared by chemical crosslinking with BA through in situ polymerization using different BA ratios between 1 and 10% w/w. Afterward, the compositions, thermal stability, transparence, oxygen permeability, water uptake capacity, swelling ratio as well as morphological and rheological properties, in vitro degradability, in vitro cytotoxicity, and antibacterial properties of the all prepared materials were analyzed using a series of different techniques. The thermal stability, hydrophilicity, water uptake, oxygen permeability gradually increased depending ratio of BA, which is desirable for biomaterial. While the transparence and refractive index decreased, the composite hydrogels, except for BA content of 10 wt %, maintained enough transparency to be used for contact lens. In addition, PHEMA/BA composite hydrogels exhibited good cytocompatibility (PHEMA‐1%BA and PHEMA‐3%BA) and excellent antibacterial activity against Gram‐positive (Staphylococcus aureus and Enterococcus faecium) and Gram‐negative (Escherichia coli and Pseudomonas aeruginosa) bacteria. Overall, the results demonstrated that the obtained PHEMA/BA composite hydrogels could be considered as self‐antibacterial contact lens and a potential composite biomaterial for other applications. © 2018 Wiley Periodicals, Inc.
