Human exposure to methylmercury (MeHg) is currently high in regions such as the Amazon. Understanding the molecular changes associated with MeHg-induced neurotoxicity and the crosstalk with the periphery is essential to support early diagnoses. This work aimed to evaluate cellular and molecular changes associated with behavioral alterations in MeHg acute exposure and the possible changes in extracellular vesicles (EVs) number and S100β content. Adults male Wistar rats were orally treated with 5 mg/kg for four days. Behavioral performance, molecular and histological changes in the cerebellum, and plasma EVs were assessed. MeHg-intoxicated animals performed significantly worse in behavioral tests. MeHg increased the number of GFAP+ cells and GFAP and S100β mRNA expression in the cerebellum but no change in NeuN+ or IBA-1+ cells number was detected. The number of exosomes isolated from plasma were decreased by the metal. S100B mRNA was detected in circulating plasma EVs cargo in MeHg exposure. Though preliminary, our results suggest astrocytic reactivity is displaying a protective role once there was no neuronal death. Interestingly, the reduction in exosomes number could be a new mechanism associated with MeHg-induced neurotoxicity and plasma EVs could represent a source of future biomarkers in MeHg intoxication.  相似文献

In the offshore oil and gas industry, mainly focusing on the use of rigid or flexible pipes of subsea infrastructure applied to risers or flowlines, one of the greatest difficulties is the interpretation of the combined effects of the various correlated phenomena (hydrodynamic effects of intermittent flow, the effects of corrosivity of the environment in addition to variations in pressure, temperature, and dynamic loading). On the basis of this scenario, defining the degree of severity of each of the correlated system variables becomes of fundamental importance for establishing reliable criteria for selecting materials for subsea application. The established flow pattern directly affects the corrosion rate (or the pipe material mass loss), but the balance of other variables including possible changes in the physical and transported fluid chemical properties may increase the damage up to an order of magnitude, which is a piece of information normally not foreseen in design criteria. Therefore, to improve the understanding of the corrosion study influenced by multiphase flow, a testing loop was designed and assembled at the Corrosion and Protection Laboratory of the Institute for Technological Research, in which API X80 steel coupons were positioned in locations with a 0° and 45° inclinations. Tests were conducted by varying the partial pressure of the gaseous phase containing blends of CO2 and H2S with N2 balance, mixed with the liquid phase containing light oil and heavy oil in water with salinity (NaCl)-simulating oil well conditions with 80% water cut. The main objective of this study is to establish models that can predict the corrosion intensity in conditions close to those obtained experimentally. To achieve results, the multiple regression and Box–Cox transformation methods were applied. These models will make possible damage prediction and optimization of matrix parameters for the multiphase-loop test.  相似文献

The catalytic properties of 10-MR (membered ring) zeolites (ZSM-5, MCM-22, IM-5, ITQ-2, all with a similar Si/Al ratio of ca. 15) in hybrid Co/SiO2-zeolite catalysts for the direct conversion of syngas to mainly high-octane gasoline-range hydrocarbons has been studied under typical Fischer-Tropsch (FT) conditions: 250 °C, 2.0 MPa, and H2/CO = 2. Special emphasis has been given to the deactivation behavior and the characterization of the amount and nature of the carbonaceous deposits formed by a combination of techniques (elemental analysis, TGA (thermogravimetric analyses), GC–MS, and DR (diffuse reflectance) UV–vis spectroscopy). The presence of the medium-pore zeolite increases the gasoline yield by about 20–50%, depending on the particular zeolite, and enhances the formation of branched products with respect to the base Co/SiO2 catalyst, which is explained by the promotion of isomerization and cracking of long-chain (C13+) n-paraffins formed on the FT component. The initial zeolite activity is mostly determined by the surface acidity rather than by the total amount of Brønsted acid sites, pointing out to the existence of limitations for the diffusion of the long-chain n-paraffins through the 10-MR channels under FT conditions. Thus, ITQ-2 bearing the largest surface area presents the highest initial yield of branched gasoline-range products, followed by ZSM-5, IM-5, and MCM-22. All zeolites experience a loss of activity with TOS, particularly during the initial reaction stages. This deactivation is governed by the morphological and structural properties of the zeolite, which finally determine the amount and location of the coke species, and not by the acidity.  相似文献

This work aims to investigate the effect of adding vulcanized or partially devulcanized rubbers on recycled polypropylene (PPr), considering thermomechanical and morphological properties. The study proposes to better understand how structural changes underwent by rubber (after the devulcanization) contributed to improving the mechanical properties of the PPr. The PPr/rubber blends were prepared by a co-rotating twin-screw extruder and then were injected. The blends composed of the most devulcanized rubbers by microwaves with refined microstructure showed higher values of elongation at break and toughness. Data showed that the devulcanization process applied to the rubber interfered positively in its adhesion to the PPr. Data from dynamic mechanical analysis and atomic force microscopy indicated that the most devulcanized rubbers presented an interface more connected to PPr. These chemical interactions possibly impacted the mechanical properties of the PPr. Moreover, dilatation processes favored the fracture mechanisms of the PPr when rubber was added to it.  相似文献

This work deals with the rheological, morphological, and thermal properties of composites having poly(ethylene terephthalate) (PET), polyamide-6 (PA6), and their blends as matrices, and rice husk ash (RHA) as a filler. The study determines the effect of composition on the change in viscosity and rate of degradation during processing in a torque rheometer. Our data indicates that thermal stability and degradation during processing depend on matrix composition and filler concentration. SEM micrographs show both partial adhesion of the filler to the matrices and filler pullout. Optical microscopy shows particle agglomeration and that agglomerate size increased with filler content. FTIR investigates the shifting of absorption bands of PET/PA6 composite after the addition of RHA and attributes the selective dispersion of RHA to the formation of hydrogen bonds. Our data supports the idea that filler employed here is an option to develop polymer composites with improved properties.  相似文献

We aimed to compare detailed fat distribution and lipid profile between young adults with congenital adrenal hyperplasia due to 21-hydroxylase enzyme deficiency and a control group. We also verified independent associations of treatment duration and daily hydrocortisone dose equivalent (HDE) with lipid profile within patients. This case–control study included 23 patients (7 male and 16 female) matched by an age range of young adults (18–31 years) with 20 control subjects (8 male and 12 female). Dual energy X-ray absorptiometry was used to measure the fat distribution. Male patients demonstrated elevated indices of fat mass for total (7.7 ± 2.1 vs. 4.5 ± 1.3 kg/m2, p = 0.003), trunk (4.0 ± 1.2 vs. 2.2 ± 0.8 kg/m2, p = 0.005), android (0.63 ± 0.24 vs. 0.32 ± 0.15 kg/m2, p = 0.008), gynoid (1.34 ± 0.43 vs. 0.74 ± 0.24 kg/m2, p = 0.005), arm (0.65 ± 0.16 vs. 0.39 ± 0.10 kg/m2, p = 0.009), and leg regions (2.7 ± 0.8 vs. 1.6 ± 0.4 kg/m2, p = 0.005) than the control group, but not in females. However, female patients demonstrated elevated ratio of low-density lipoprotein cholesterol to high-density lipoprotein cholesterol (1.90 ± 0.46 vs. 1.39 ± 0.47, p = 0.009) than the control group, but not in males. Total fat mass was inversely correlated with total testosterone (r = −0.64, p = 0.014) and positively correlated with leptin in males (r = 0.75, p = 0.002). An elevated daily HDE (β = 0.43, p = 0.038 and β = 0.47, p = 0.033) and trunk to total fat mass ratio (β = 0.46, p = 0.025, and β = 0.45, p = 0.037) were independently correlated with impaired lipid profile markers. Although there is no altered lipid profile, male patients demonstrated an increased fat distribution. However, female patients presented with an impaired lipid profile marker but demonstrated close values of normal fat distribution. Interestingly, the dose of glucocorticoid therapy can have some role in the lipid mechanisms.  相似文献

While searching for healthier diets, people became more attentive to organic produce. Yet, organic foods may be more susceptible to microbiological contamination because of the use of organic fertilizers, a possible source of pathogenic bacteria. In this study, 130 samples of different organic and conventional vegetable varieties sold in Brazil were analyzed for mesophilic aerobic bacteria, yeasts and molds, total coliforms, Escherichia coli and Salmonella spp. Most of the mesophilic aerobic bacteria counts in organic and conventional vegetables ranged from 6 to 7 log10 CFU/g; most of the yeasts and molds counts ranged from 5 to 6 log10 CFU/g and most of the total coliforms counts ranged from 4 to 5 log10 CFU/g. E. coli was found in 41.5% of the organic and 40.0% of the conventional vegetables, and most samples had counts ranging from 1 to 2 log10 CFU/g. Salmonella spp. was not found in any sample. Comparative analyses of the microbial counts of organic and conventional vegetables showed that some organic varieties have greater counts. However, the global results show that this is not a trend. These results indicate the need of good farming practices, and proper sanitization before consumption, to ensure food quality and safety.  相似文献

A one-dimensional mathematical model is presented for flows in a rocket engine that has a regenerative cooling system. The problem involves the flow of a gas in a converging- diverging nozzle, the flow of a coolant in channels distributed around the engine, and the heat conduction through a wall between the gas and the coolant. The numerical model adopted is based on the finite-volume method with a second-order scheme. It was noted that it is important to use variable properties in order to predict the maximum wall temperature in the rocket engine and the drop in pressure of the coolant as it moves along the channels, whereas the thrust of the engine can be calculated with constant properties.  相似文献

The sporicidal effect of hydrogen peroxide (H2O2) in the sterilization of low density polyethylene film (LDPE) was evaluated using a Central Composite Design (CCD). The effects of contact time (5-19s), bath temperature (23–70 °C) and concentration of H2O2 (0–35%) in an immersion bath were investigated. A 16 cm2 film surface was evenly inoculated with 100 μL of the test microorganism Bacillus subtilis var. globigii ATCC9372 spores. The effective H2O2 sporicidal activity was demonstrated at concentrations from 18 to 35% and in a temperature range from 46 to 70 °C, resulting in 2–7 decimal reductions of B. subtilis spores. A quadratic mathematical model representative of the action of H2O2 on the B. subtilis spores was developed as a function of concentration, time and temperature. Test specimens sanitized with 28% H2O2 at 60 °C for 8 s showed 4 decimal reductions. In the same sterilization procedure, but extending the time to 16 s, this value increased to 7 decimal reductions, demonstrating the efficiency of H2O2 as a function of contact time. The sterilization system tested showed satisfactory performance in the sterilization of LDPE films, being capable of reaching up to 7 decimal reductions of the bacterial spore population.  相似文献
