BACKGROUND AND OBJECTIVE: Drug resistance has become a major cause of treatment failure in patients with acute leukemia. P-glycoprotein (Pgp), which is associated with the multidrug resistance (MDR) phenotype, has been reported to be an important predictor of treatment outcome. The aim of this study was to analyze the value of Pgp expression in bone marrow or peripheral blood as a predictor of the response to remission induction chemotherapy as well as the duration of remission in patients with de novo acute myeloid leukemia (AML). DESIGN AND METHODS: We examined the expression of Pgp in 82 patients with de novo AML using an immunocytochemical assay with the C219 monoclonal antibody. RESULTS: Twenty-seven of the 82 patients (33%) were C219-positive in from 1% to 100% of their cells. Thirteen cases (16%) showed a positive reaction in more than 50% of the leukemic cells. Only hyperleukocytosis was significantly associated with higher expression of Pgp. Although 8 of the 13 cases (62%) with more than 50% of cells having Pgp expression were CD34-positive, this association was not statistically significant. A univariate analysis of resistance to induction therapy showed a significantly higher resistance rate in patients with increased Pgp expression (P = 0.01) as well as in those patients with decreased reactivity to myeloperoxidase. The multivariate analysis revealed the independent prognostic value of Pgp expression. C219 reactivity did not have an influence on remission duration. INTERPRETATION AND CONCLUSIONS: Our data indicate that P-glycoprotein expression is a reliable marker of resistance to induction treatment in patients with de novo AML. 相似文献

The notion of a linogram is introduced. It corresponds to the notion of a sinogram in the conventional representation of projection data in image reconstruction. In the sinogram, points which correspond to rays which go through a fixed point in the cross section to be reconstructed all fall on a sinusoidal curve. In the linogram, however, these points fall on a straight line. Thus, backprojection corresponds to integration along straight lines in the linogram. A theorem is proved expressing the backprojection operator in terms of the Radon transform and simple changes of variables. Consequences of this theorem are discussed. A novel image reconstruction method based on the theorem is presented. 相似文献

Summary Monoglyceride concentrates are quantitatively separated into mono-, di-, and triglyceride components on silica gel columns
by an adsorption chromatographic technique. The separated glycerides are determined gravimetrically. The adsorption on silica
gel is dependent on the number of hydroxyl groups in the molecule, and the influence of unsaturation and chain length is minimized.
Combinations of benzene and ethyl ether are used for elution, thus preventing isomerization, which frequently results when
polar solvents such as alcohols are used.
The procedure for the chromatographic separation is simple and straightforward. The equipment used is easily obtainable. The
silica gel adsorbent requires only adjustment of its water content before use.
Mixtures of both saturated and unsaturated glycerides and those having different fatty acid chain length have been separated.
Known mixtures have also been analyzed. For a known mixture containing 15% tripalmitin, 45% distearin, and 38% monopalmitin

[1-14C]Heptane was aromatized under 5 atm pressure over a “nonacidic” 3% platinum on alumina catalyst maintained at 500 °C. The reactant was pumped in at a HLSV of 0.88, and the hydrogen-to-heptane mole ratio was 5.1. The samples of product were collected (one during the first 90 min and another during the next 90 min), the toluene was separated from the respective samples, and the radioactivity in each position of the toluene was determined by a degradation sequence. The distribution of radioactivity in sample 1 was as follows: in position 1–2.6%, 2–38.6%; 3–14.5%; 4–4.2%, and in the methyl group—40.1%. Approximately similar distribution of radioactivity was found in sample 2. 1,6-Ring closure is the most important pathway leading to the formation of toluene from [1-14C]heptane; lesser amounts of repetitive, 1,5-ring closure and hydrogenolysis, and bond shift reactions followed by 1,6-ring closure and dehydration can explain the distribution of radioactivity. Zircon, ZrSiO4, is a natural mineral used for various applications as a refractory bulk material. It is an excellent feedstock for the plasma
spraying of protective coatings and free-standing bodies. Zircon decomposes on spraying into t-ZrO2 and glassy SiO2, which can be preserved in deposits by fast cooling. This combination of zirconia and silica exhibits properties such as
a high thermal shock resistance, good corrosion resistance, low wettability, etc. The final properties of deposits can be
further enhanced by the addition of other materials such as alumina. For instance, alumina-zircon plasma-sprayed free-standing
