In consumer food‐sensory studies, sorting and closely related methods (for example, projective mapping) have often been applied to large product sets which are complex and fatiguing for panelists. Analysis of sorting by Multi‐Dimensional Scaling (MDS) is common, but this method discards relevant individual decisions; analysis by DISTATIS, which accounts for individual differences, is gaining acceptance. This research posits that replication can improve DISTATIS analysis by stabilizing consumer sensory maps, which are often extremely unstable. As a case study a fatiguing product set was sorted: 10 American whiskeys—5 bourbons and 5 ryes—were sorted into groups by 21 consumers over 2 replications. These products were chosen because American whiskeys are some of the most important distilled beverages in today's market; in particular, “bourbon” (mashbill more than 50% corn) and “rye” (more than 50% rye) whiskeys are important and assumed to be products with distinct sensory attributes. However, there is almost no scientific information about their sensory properties. Data were analyzed using standard and aggregated DISTATIS and MDS. No significant relationship between mashbill and consumer categorization in whiskeys was found; instead, there was evidence of producer and aging effects. aggregated DISTATIS was found to provide more stable results than without replication, and DISTATIS results provided a number of benefits over MDS, including bootstrapped confidence intervals for product separation. In addition, this is the first published evidence that mashbill does not determine sensory properties of American whiskey: bourbons and ryes, while legally distinct, were not separated by consumers.  相似文献

Pork loin color categories were established with the intent of demonstrating differences in eating quality between them. Center-cut boneless pork loins (n=64) were divided into three color classifications. Category A included National Pork Producers Council color standards 1 and 2; category B, 3 and 4; and category C, 5 and 6. L* means for categories A, B, and C were 57.00, 50.24, and 45.54, respectively. Warner-Bratzler shear force values were not affected by category (P>0.05), but were negatively correlated with a* and b* values (P<0.05). The "in-home" portion of this study consisted of 47 households that prepared and evaluated chops randomly distributed by category. Consumers reported differences in liking of juiciness and liking of tenderness (P<0.05) due to color category. Eighty-nine percent of the households participated in a simulated retail display where, 20.8% chose chops from category A, 26.4% from category B, and 52.8% from category C. A trained panel evaluated the cooked chops and perceived category C to be more tender, more juicy, and less dry than both A and B (P<0.05). Consumers responded similarly to the trained panel in their perceptions of tenderness and juiciness.  相似文献

Denitrification of groundwater with methane as sole hydrogen donor   总被引:2,自引:0,他引:2  
It was examined, whether methane can be used as hydrogen donor for an in situ denitrification of groundwater. It is demonstrated, that groundwater can serve as liquid medium and that the denitrification can occur at 10 degrees C. Efforts to enrich methanotrophic bacteria under anoxic conditions have not been successful. No methane oxidation occurred in the absence of oxygen. For this reason, the denitrification with methane must be performed in a two-stage process with aerobic methanotrophic bacteria producing metabolites, that are used as hydrogen donor by non-methanotrophic bacteria in anoxic areas. This kind of indirect denitrification was proved by quantifying nitrogen and nitrous oxide in enrichment cultures that were not stirred or shaken. Large numbers of non-methanotrophic bacteria being able to denitrify with methanol, acetate or proteins as sole hydrogen donor were enriched besides the methanotrophic bacteria under these conditions.  相似文献

Integrins belong to a large family of heterodimeric membrane glycoproteins which mediate cell-cell or cell-extracellular matrix interactions. These interactions could play a major role during the migration of tumor cells across the extracellular matrix and vascular endothelium and would thus appear to be requisite for the metastatic process. Pretreatment of the Foss human melanoma cell line with HILDA/LIF or OSM, two cytokines involved in acute-phase response, increased the expression of membrane alpha v beta 1 1.5-2-fold. The same phenomenon was observed on the SK-N-SH human neuroblastoma cell line. alpha v beta 1 upmodulation was concomitant with improved tumor cells attachment to the fibronectin matrix. This greater adhesion of tumor cells to fibronectin was inhibited by specific monoclonal antibodies against alpha v or beta 1 integrin subunits. Similar results were obtained after TNF-alpha treatment. Our findings demonstrate the ability of HILDA/LIF and OSM to modulate tumor cell capacity to adhere to the matrix component, suggesting a potential role for these cytokines in modulation of tumoral progression.  相似文献

In order to understand protein structure to a sufficient extent for, e.g., drug discovery, no single technique can provide satisfactory information on both the lowest-energy conformation and on dynamic changes over time (the ‘four-dimensional’ protein structure). Instead, a combination of complementary techniques is required. Mass spectrometry methods have shown promise in addressing protein dynamics, but often rely on the use of high-end commercial or custom instruments. Here, we apply well-established chemistry to conformation-sensitive oxidative protein labelling on a timescale of a few seconds, followed by analysis through a routine protein analysis workflow. For a set of model proteins, we show that site selectivity of labelling can indeed be rationalised in terms of known structural information, and that conformational changes induced by ligand binding are reflected in the modification pattern. In addition to conventional bottom-up analysis, further insights are obtained from intact mass measurement and native mass spectrometry. We believe that this method will provide a valuable and robust addition to the ‘toolbox’ of mass spectrometry researchers studying higher-order protein structure.  相似文献

The solubilities of C60 and C70 at 25°C in seven normal alcohols obey the relationship InY = a + bX + cX2, where Y is solubility and X is the Hildebrand solubility parameter of the solvent. Extrapolation to the solubility parameter of water yields solubilities in water of 1.3'10-11 (C60) and 1.3'10-10(C70) ng/ml with an uncertainty of one order of magnitude.  相似文献
