Heterotrophic marine protists known as thraustochytrids can synthesize polyunsaturated fatty acids (PUFAs) such as docosahexaenoic acid (DHA). The biosynthetic pathways of PUFAs in thraustochytrids are poorly understood, however. In this study, we attempted to reveal the enzymes involved in DHA synthesis in thraustochytrids. Nine thraustochytrid strains representing 3 genera (Aurantiochytrium, Schizochytrium, and Thraustochytrium) were used for PCR-based detection of the genes encoding Δ5-elongase and Δ4-desaturase and for fatty acid analysis. The degenerate primers were designed to amplify the Δ5-elongase and Δ4-desaturase genes, and the partial sequences of the enzymes were obtained from the genera Thraustochytrium and Schizochytrium. These fragments were identical to those of known Δ5-elongase and Δ4-desaturase. Neither Δ5-elongase nor Δ4-desaturase was detected in the strains belonging to the genus Aurantiochytrium, however, suggesting that this group likely synthesizes DHA not via the elongation/desaturation pathway but via an alternate pathway such as the polyketide synthase pathway. The fatty acid profiles of thraustochytrids were consistent with the presence of genes involved in PUFA biosynthesis in thraustochytrid genera. Thus, our findings suggest that two biosynthetic pathways for PUFAs exist in these organisms.  相似文献

In this study, complexes composed of poly-l-tyrosine (pLT) and single-walled carbon nanotubes (SWCNTs) were produced and the dispersibility of the pLT/SWCNT complexes in water by measuring the ζ potential of the complexes and the turbidity of the solution were investigated. It is found that the absolute value of the ζ potential of the pLT/SWCNT complexes is as high as that of SWCNTs modified with double-stranded DNA (dsDNA) and that the complexes remain stably dispersed in the water at least for two weeks. Thermogravimetry analysis (TGA) and visualization of the surface structures of pLT/SWCNT complexes using an atomic force microscope (AFM) were also carried out.  相似文献

The degradation and the recovery behavior of the device performance for SiGe diodes and p-MOSFETs irradiated by 2-MeV electrons are reported. For diodes, it is noted that both the reverse and forward current increase by irradiation. An interesting observation is that the forward current decreases after irradiation for a forward voltage larger than ~ 0.7 V. This reduction can be explained by an increased resistivity of the Si substrate. The degradation recovers by thermal annealing after irradiation. For a fluence of 1 × 1015 e/cm2, the diode performance almost recovers to the initial condition after 250 °C annealing. For the transistors, after irradiation, a slight negative shift of the threshold voltage and a decrease of the drain current for input and output characteristics have been observed together with a decrease of hole mobility. This is mainly due to the increase of the threshold voltage induced by positive charge trapping in the gate oxide.  相似文献

Hexagonal boron carbonitride (h-BCN) hybrid films have been synthesized on highly oriented pyrolytic graphite by radiofrequency plasma enhanced chemical vapor deposition using tris-(dimethylamino)borane as a single-source molecular precursor. The films were characterized by X-ray photoelectron spectroscopy (XPS), near-edge X-ray absorption fine structure (NEXAFS) and Raman spectroscopic measurements. XPS measurement showed that the B atoms were bonded to C and N atoms to form the sp2-B-C-N atomic hybrid chemical environment. The atomic composition estimated from the XPS of the typical sample was found to be almost B1C1N1. NEXAFS spectra of the B K-edge and the N K-edge had the peaks due to the π* and σ* resonances of sp2 hybrid orbitals implying the existence of the sp2 hybrid configurations of h-BCN around the B atoms. The G band at 1592 and D band at 1352 cm− 1 in the Raman spectra also suggested the presence of the graphite-like sp2-B-C-N atomic hybrid bonds. The films consisted of micrometer scale crystalline structure of around 10 µm thick has been confirmed by the field emission scanning electron microscopy.  相似文献

When an amphiphilic block copolymer is directly spread from an organic solution, the morphology of the surface micelles is not in general controlled because the structure is rapidly frozen after the solvent evaporation. In contrast, we have developed a new versatile method to generate highly regular ordered nanopatterns of the surface micelles on water, which involves the process of two dimensional (2D) hybridization with a polar liquid crystal molecule, 4′-pentyl-4-cyanobiphenyl (5CB) [Langmuir 22 (2006) 5233]. The present work extends this approach using a family of diblock copolymers of polystyrene-block-poly(4-vinylpyridine) changing the chain length of the two polymer blocks. It is found here that the dot-to-dot distance and the dot size can be precisely controlled on-demand by the length of the polymer blocks. Such structural regulations cannot be attained without the 5CB hybridization. Due to the high reproducibility of the morphology formation, this process should be of practical significance to ‘engineer’ the 2D patterns in the range of some tens of nanometers.  相似文献

Mimics of natural antimicrobial peptides are promising compounds to fight the rising threat of multi-drug resistant bacteria. Here we report the design, synthesis and conformational analysis of a new class of antimicrobial peptide mimetics incorporating a diphenylacetylene scaffold. Within a small set of compounds, we observe a correlation between amphiphilicity, the efficiency of partitioning into negatively charged membranes and antibacterial activity. The most amphiphilic compound, which contains four isoleucine residues and four lysine residues, displays species-selective antibacterial activity (most active against Bacillus subtills) and low haemolytic activity. Solution-phase conformational analysis of this compound indicates that a defined structure is adopted in the presence of negatively charged phospholipid membranes and aqueous 2,2,2-trifluoroethanol but not in water. A conformation model indicates that the cationic and hydrophobic functional groups are segregated. These results may inform the development of highly selective antimicrobial peptide mimetics for therapeutic applications.  相似文献

High-speed and low-power divide-by-252 or -256 circuit have been fabricated by using high-transconductance GaAs enhancement-mode MESFETs. This variable-modulus divider is able to operate up to a clock frequency of 3.7 GHz. The total power dissipation at the maximum frequency is 180 mW, and it is as low as 42 mW and 30 mW at 3 GHz and 2.5 GHz, respectively.  相似文献

We experienced 12 cases of congenital cystic adenomatoid malformation of the lung (CCAM) including 6 cases diagnosed antenatally. They were classified into three groups according to the clinical manifestations. Group A was associated with hydrops fetalis (n = 3), group B presented with respiratory distress symptoms after birth (n = 6), and group C showed no respiratory symptoms (n = 3). All cases of group A were lost because of hydrops and respiratory failure due to pulmonary hypoplasia. Because a compression of the mass is thought to be a cause of hydrops, this group is considered to be a good indication for fetal treatment. All cases of group B showed progressive respiratory symptoms a few days after birth which were successfully treated surgically. In 2 of 3 cases of group C, the lesions decreased in size both antenatally and postnatally. We conclude that serial sonographic evaluations for fetal CCAM are important. If the fetus develops hydrops, fetal surgery is to be considered. If not, however, fetal surgery should not be done, because some lesions can shrink in size, or even disappear, while others can be treated successfully after birth by lobectomy or even segmentectomy.  相似文献
