The development of insulin resistance and type 2 diabetes is determined by various factors, including defects within the insulin signaling pathway. Mediators of insulin resistance operate through activation of various protein kinase C isoforms, IκB kinase β (IKKβ), and/or c‐Jun N‐terminal kinase, and subsequent inhibition of the proximal insulin signaling pathway via the insulin receptor substrate 1 and Akt. These mechanisms are still largely unresolved because of the complexity of the molecular events. In this study, an expression and activation state profiling of multiple known key signaling biomolecules involved in insulin metabolic and mitogenic signaling pathways was evaluated using a phosphospecific antibody array platform. The results of the arrayed antibodies were verified by the multiplexed bead array assay and conventional Western blot analysis, and confirmed the well‐known inhibitory effects of phorbol esters on insulin signaling pathway activation. Of interest, the increase in protein kinase C signaling responses with phorbol esters was associated with activation of the lipid phosphatase PTEN and a 27 kDa HSP. Thus, this insulin signaling antibody array provides a powerful and effective way to investigate the mechanism of insulin resistance and likely assist the development of innovative therapeutic drugs for type 2 diabetes. 相似文献

In order to utilize the potential advantages of replicated collaborative CAD system to support natural, free, fast and less-constrained multi-user human-to-human interaction, a local-locking mechanism, which can provide fast modeling response, is adopted as concurrency control mechanism for a replicated on-line collaborative CAD system. A human–human interactive modeling is achieved by immediate local execution of modeling operations and exchange of modeling operations across all collaborative sites real-time. In particular, an approach to achieve topological entity correspondence across collaborative sites during modeling procedure, which is critical to guarantee the correctness and consistency of collaborative modeling result, is proposed. Previous trust models are mainly focused on reputational mechanism based on explicit trust ratings. However, the large amount
of user-generated content and community context published on Web is often ignored. Without enough information, there are several
problems with previous trust models: first, they cannot determine in which field one user trusts in another, so many models
assume that trust exists in all fields. Second some models are not able to delineate the variation of trust scales, therefore
they regard each user trusts all his friends to the same extent. Third, since these models only focus on explicit trust ratings,
so the trust matrix is very sparse. To solve these problems, we present RCCtrust – a trust model which combines Reputation-,
Content- and Context-based mechanisms to provide more accurate, fine-grained and efficient trust management for the electronic
community. We extract trust-related information from user-generated content and community context from Web to extend reputation-based
trust models. We introduce role-based and behavior-based reasoning functionalities to infer users' interests and category-specific trust relationships. Following the study in sociology, RCCtrust exploits similarities between pairs of users to depict differentiated
trust scales. The experimental results show that RCCtrust outperforms pure user similarity method and linear decay trust-aware

目的建立采用高效液相色谱法同时测定调节血脂类保健食品中烟酸、普伐他汀钠、瑞舒伐他汀钙、苯扎贝特、阿托伐他汀钙、氟伐他汀钠、吉非罗齐、洛伐他汀及辛伐他汀9种非法添加药物的方法。方法以甲醇为样品提取溶剂,采用Inertsil ODS-3色谱柱,以0.02 mol/L乙酸铵-甲醇为流动相梯度洗脱;流速为1.0m L/min;检测波长为230 nm,柱温为30℃,利用保留时间定性、外标法定量,分析9种非法添加药物。结果本方法可以在20 min内定性并定量分析烟酸、普伐他汀钠、瑞舒伐他汀钙、苯扎贝特、阿托伐他汀钙、氟伐他汀钠、吉非罗齐、洛伐他汀及辛伐他汀9种化学药物。9种非法添加药物在1.0~100μg/m L之间线性关系良好,相关系数均大于0.999;除阿托伐他汀钙在袋泡茶中回收率较低外,平均加标回收率范围为77.9%~102.9%,相对标准偏差范围为0.42%~5.8%。结论该方法线性关系良好,线性范围广,准确度及精密度高,可同时鉴定是否非法添加上述9种化学药物。 相似文献

Effect of easily degradable substrate on anaerobic degradation of pentachlorophenol in an upflow anaerobic sludge blanket (UASB) reactor 总被引:7,自引:0,他引:7
The effect of microbial easily degradable substrate (MEDS) on the anaerobic degradation of pentachlorophenol (PCP) in two upflow anaerobic sludge blanket (UASB) reactors was investigated. The results indicated that glucose-utilizing activity decreased with the increase of PCP concentration in the mixed culture, and MEDS promoted PCP-dechlorination and degrading activities. The concentration of MEDS increased from 917 to 4580 mg L(-1) with the increase of PCP concentration from 100 to 181 mg L(-1)in influent, which was necessary for maintenance of steady operation of the experimental reactors, the removal rate of PCP and COD ranged up to 99.5 and 90.0% and the concentration of PCP in the effluent was less than 0.5 mg L(-1). The concentration of PCP in effluent was linearly or logarithmically related to sucrose concentration in the influent while PCP was less than the maximum permissible PCP concentration. The activity of anaerobic sludge in the reactor decreased as the concentration of PCP increased, but it could be recovered step by step as time progressed. In the lowest layer of the reactor, the activity of sludge was the highest. So it is feasible to accelerate the degradation of the organic toxic compounds like PCP, by adding suitable quantities of microbial easily degradable substrate to the system. 相似文献

Anchoring single metal atom to carbon supports represents an exceptionally effective strategy to maximize the efficiency of catalysts. Recently, dual-atom catalysts (DACs) emerge as an intriguing candidate for atomic catalysts, which perform better than single-atom catalysts (SACs). However, the clarification of the polynary single-atom structures and their beneficial effects remains a daunting challenge. Here, atomically dispersed triple Zn-Co-Fe sites anchored to nitrogen-doped carbon (ZnCoFe-N-C) prepared by one-step pyrolysis of a designed metal-organic framework precursor are reported. The atomically isolated trimetallic configuration in ZnCoFe-N-C is identified by annular dark-field scanning transmission electron microscopy and spectroscopic techniques. Benefiting from the synergistic effect of trimetallic single atoms, nitrogen, and carbon, ZnCoFe-N-C exhibits excellent catalytic performance in bifunctional oxygen reduction/evolution reactions in an alkaline medium, outperforming other SACs and DACs. The ZnCoFe-N-C-based Zn-air battery exhibits a high specific capacity (liquid state: 931.8 Wh kgZn−1), power density (liquid state: 137.8 mW cm−2; all-solid-state: 107.9 mW cm−2), and good cycling stability. Furthermore, density-functional theory calculations rationalize the excellent performance by demonstrating that the ZnCoFe-N-C catalyst has upshifted d-band center that enhances the adsorption of the reaction intermediates. 相似文献
