Flavanols of Camellia sinensis exhibit uric acid (UA) lowering effect, through the modulation of both xanthine oxidase and urate excretion. In order to investigate the potential benefit of Camellia Sinenis products in asymptomatic hyperuricemia, a meta-analysis of long-term Randomized Controlled Trials (RCT) with tea or tea extract has been conducted. From 20 human intervention studies selected only 5 RCT (13 interventions) were suitable for meta-analysis (n = 472). The current “normal” range set for hyperuricemia fails to identify patients with potential metabolic disorders. Therefore on the basis of the literature data, we fixed cut-off limits for UA baseline levels of 4.5 mg/dl for women, 6.1 mg/dl for men, and 5.5 mg/dl for studies involving mixed populations. Statistically significant effects were not found, but subgroup analysis revealed that the Pooled Estimate effect was different in subjects with baseline levels under [MD (95% CI): 0.1078 (?0.0528 to 0.2684)] and over the cut-off [MD (95% CI): ?0.0239 (0.3311 to 0.2833)]. However, due to the low number of RCT and to the lack of data on bioavailability, it is difficult to draw any firm conclusion and more studies are needed to establish if tea flavanols could be useful in asymptomatic hyperuricemia treatment. 相似文献

Magnesium oxide (MgO) nanoparticles were dispersed via solution processing in poly(1-trimethylsilyl-1-propyne) (PTMSP) to form polymer nanocomposites. Transmission electron microscopy was used to determine the extent of particle aggregation in the composites. Both nanocomposite density and CO2, CH4, N2, and H2 permeability were influenced by nanoparticle loading. Nanocomposite densities were markedly lower than predicted by a two phase additive model. For example, in films containing 75 nominal volume percent MgO, the polymer-particle composite density was 68 percent lower than expected based on an additive model. At this loading, gas permeability coefficients were, depending on the gas, 17-50 times higher than in unfilled PTMSP at similar conditions. The changes in permeability with particle content were interpreted in terms of measured changes in gas solubility with particle content and diffusion coefficients calculated from the permeability and solubility data. 相似文献

Flavonoids, substantial components of the human diet, are generally considered to be beneficial. However, they may possess possible pro-oxidative effects, which could be based on their reducing potential. The aims of this study were to evaluate the ability of 26 flavonoids to reduce ferric ions at relevant pH conditions and to find a possible relationship with potentiation of hydroxyl radical production. A substantial ferric ions reduction was achieved under acidic conditions, particularly by flavonols and flavanols with the catecholic ring B. Apparently corresponding bell-shaped curves displaying the pro-oxidant effect of flavonols quercetin and kaempferol on iron-based Fenton reaction were documented. Several flavonoids were efficient antioxidants at very low concentrations but rather inefficient or pro-oxidative at higher concentrations. Flavonols, morin and rutin were progressively pro-oxidant, while 7-hydroxyflavone and hesperetin were the only flavonoids with dose-dependent inhibition of hydroxyl radical production. Tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β) are two cytokines involved in the perpetuation of the chronic inflammation state characterizing rheumatoid arthritis (RA). Significant advances in the treatment of this pathology have been made over the past ten years, partially through the development of anti-TNF and anti-IL-1 therapies. However, major side effects still persist and new alternative therapies should be considered. The formulation of the micro-immunotherapy medicine (MIM) 2LARTH® uses ultra-low doses (ULD) of TNF-α, IL-1β, and IL-2, in association with other immune factors, to gently restore the body’s homeostasis. The first part of this review aims at delineating the pivotal roles played by IL-1β and TNF-α in RA physiopathology, leading to the development of anti-TNF and anti-IL-1 therapeutic agents. In a second part, an emphasis will be made on explaining the rationale of using multiple therapeutic targets, including both IL-1β and TNF-α in 2LARTH® medicine. Particular attention will be paid to the ULD of those two main pro-inflammatory factors in order to counteract their overexpression through the lens of their molecular implication in RA pathogenesis. 相似文献

Inherited retinal diseases (IRDs) are a heterogeneous group of conditions that include retinitis pigmentosa (RP) and Leber congenital amaurosis (LCA) and early-onset severe retinal dystrophy (EO[S]RD), which differ in severity and age of onset. IRDs are caused by mutations in >250 genes. Variants in the RPE65 gene account for 0.6–6% of RP and 3–16% of LCA/EORD cases. Voretigene neparvovec is a gene therapy approved for the treatment of patients with an autosomal recessive retinal dystrophy due to confirmed biallelic RPE65 variants (RPE65-IRDs). Therefore, the accurate molecular diagnosis of RPE65-IRDs is crucial to identify ‘actionable’ genotypes—i.e., genotypes that may benefit from the treatment—and is an integral part of patient management. To date, hundreds of RPE65 variants have been identified, some of which are classified as pathogenic or likely pathogenic, while the significance of others is yet to be established. In this review, we provide an overview of the genetic diagnostic workup needed to select patients that could be eligible for voretigene neparvovec treatment. Careful clinical characterization of patients by multidisciplinary teams of experts, combined with the availability of next-generation sequencing approaches, can accelerate patients’ access to available therapeutic options. 相似文献

Total polyphenols, carotenoids and vitamin C were assessed in three different peaches cultivars (Prunus persica): ‘Luisa Berselli’ (LB), ‘Stark Earlyglo’ (SE) and ‘Maria Serena’ (MS). Total antioxidant capacity of fruits and plasma antioxidant
activity after ingestion of fruits were determined as Trolox equivalent antioxidant capacity (TEAC) and as plasma total radical-trapping
potential (TRAP) respectively. The amount of polyphenols, carotenoids and vitamin C resulted to be generally similar in the
three cultivars. TEAC, measured from harvest to 7 days postharvest, resulted to be influenced mainly by vitamin C content,
whereas polyphenols and carotenoids seemed to play a secondary role. Although TEAC was similar in the three cultivars, only
LB significantly increased the TRAP in human plasma at 1, 2 and 4 h after ingestion of peaches. Sugar moiety, condensed and
glycoside phenols are suggested to be involved in the higher effect on plasma TRAP of LB. 相似文献

Noble metal nanoparticles of different sizes and shapes combined with conjugated functional polymers give rise to advanced

paper, a versatile and facile synthesis of core shell systems based on noble metal nanoparticles (AuNPs, AgNPs, PtNPs), coated
by copolymers belonging to the class of substituted polyacetylenes has been developed. The polymeric shells containing functionalities
such as phenyl, ammonium, or thiol pending groups have been chosen in order to tune hydrophilic and hydrophobic properties
and solubility of the target core shell hybrids. The Au, Ag, or Pt nanoparticles coated by poly(dimethylpropargylamonium chloride),
or poly(phenylacetylene-co-allylmercaptan). The chemical structure of polymeric shell, size and size distribution and optical
properties of hybrids have been assessed. The mean diameter of the metal core has been measured (about 10-30 nm) with polymeric
