Recently, combined carbon and hydrogen isotope fractionation investigations have emerged as a powerful tool for the characterization of reaction mechanisms relevant for the removal of organic pollutants. Here, we applied this approach in order to differentiate benzene biodegradation pathways under oxic and anoxic conditions in laboratory experiments. Carbon and hydrogen isotope fractionation of benzene was studied with four different aerobic strains using a monooxygenase or a dioxygenase for the initial benzene attack, a facultative anaerobic chlorate-reducing strain as well as a sulfate-reducing mixed culture. Carbon and hydrogen enrichment factors (epsilon(C), epsilon(H)) varied for the specific pathways and degradation conditions, respectively, so that from the individual enrichment factors only limited information could be obtained for the identification of benzene biodegradation pathways. However, using the slope derived from hydrogen vs carbon isotope discriminations or the ratio of hydrogen to carbon enrichment factors (lambda = deltaH/ deltaC approximately epsilon(H)/epsilon(C)), benzene degradation mechanisms could be distinguished. Although experimentally determined lambda values partially overlapped, ranges could be determined for different benzene biodegradation pathways. Specific lambda values were < 2 for dihydroxylation, between 7 and 9 for monohydroxylation, and > 17 for anaerobic degradation. Moreover, variations in lambda values suggest that more than one reaction mechanism exists for monohydroxylation as well as for anaerobic benzene degradation under nitrate-reducing, sulfate-reducing, or methanogenic conditions. Our results show that the combined carbon and hydrogen isotope fractionation approach has potential to elucidate biodegradation pathways of pollutants in field and laboratory microcosm studies.  相似文献

Moringa oleifera Lam is a multipurpose tree cultivated to use as a vegetable, for spice, for cooking and cosmetic oil and as a medicinal plant. Owing to the use of its seeds as food and as a clarifying agent of turbid water some nutritional and antinutritional characteristics were studied. The mature seeds contained 332.5 g crude protein, 412.0 g crude fat, 211.2 g carbohydrate and 44.3 g ash per kg dry matter. The essential amino acid profile compared with the FAO/WHO/UNU scoring pattern requirements for different age groups showed deficiency of lysine, threonine and valine. The content of methionine + cysteine (43.6 g kg−1 protein), however, was exceptionally higher and close to that of human milk, chicken egg and cow's milk. The seed extract agglutinated rabbit erythrocytes but did not show trypsin inhibitor and urease activities. Feeding rats with a diet containing the seed meal showed loss of appetite, impaired growth, lower NPU and enlargement of stomach, small intestine, caecum + colon, liver, pancreas, kidneys, heart and lungs and atrophy of thymus and spleen in comparison with rats fed on an egg‐white diet. The results indicated that consumption of M oleifera raw mature seeds should be viewed with some caution until suitable processing methods are developed to abolish the yet unknown adverse factors. © 1999 Society of Chemical Industry  相似文献

In automatic milking systems (AMS), sensors can measure cow behavior and milk composition at every milking. The aim of this observational study of previously collected data was to gain insight into the differences in dynamics of udder inflammation indicators between cows that recover and those that do not recover after detection of an initial inflammation. Milk diversion (milk separated from the bulk tank and thus indicating farmer intervention), conductivity, and somatic cell count (SCC) data from 4 wk before the initial inflammation to 12 wk after the initial inflammation were used to analyze 2,584 cases of udder inflammation. An udder inflammation case was defined as an initial observation of SCC ≥200,000 cells/mL as well as 1 additional SCC measurement >200,000 cells/mL within 10 d after the initial case, among other requirements. The data originated from 15 AMS herds in 6 countries. Four subsets of cows were created based on whether milk was diverted after the initial inflammation and whether the udder inflammation cases recovered, using a 10-d rolling average SCC threshold of 200,000 cells/mL and checking whether this rolling mean was below the threshold within 90 d after the initial inflammation as the indication of recovery. This formed the following subsets of cow lactations: milk diverted–recovered, milk diverted–not recovered, no milk diverted–not recovered, no milk diverted–recovered. Thresholds of 100,000 SCC/mL and 300,000 SCC/mL for the definition of case and recovery were also applied in a sensitivity analysis but with no substantial difference in results. Linear mixed models were used for each subset to study the variation in SCC (natural logarithm of SCC divided by 1,000) and σ-conductivity (natural logarithm of standard deviation of quarter conductivities). When observing the fraction of cows with SCC <200,000 cells/mL in the recovery subsets, most cows recovered within 20 d after the initial inflammation. In the recovery subsets, both σ-conductivity and SCC stabilized, mostly within 3 to 4 wk after the initial inflammation. σ-Conductivity stabilized above the pre-onset level in all subsets and did not show a clear increase in the no-milk-diverted subgroups, whereas SCC stabilized closer to the pre-onset level. Overall, this study indicated a cutoff point between nonchronic and chronic changes in indicators 3 to 4 wk after the initial inflammation for SCC and σ-conductivity.  相似文献

Aquifer storage and recovery (ASR) is an aquifer recharge technique in which water is injected in an aquifer during periods of surplus and withdrawn from the same well during periods of deficit. It is a critical component of the long-term water supply plan in various regions, including Florida, USA. Here, the viability of ASR as a safe and cost-effective water resource is currently being tested at a number of sites due to elevated arsenic concentrations detected during groundwater recovery. In this study, we developed a process-based reactive transport model of the coupled physical and geochemical mechanisms controlling the fate of arsenic during ASR. We analyzed multicycle hydrochemical data from a well-documented affected southwest Floridan site and evaluated a conceptual/numerical model in which (i) arsenic is initially released during pyrite oxidation triggered by the injection of oxygenated water (ii) then largely complexes to neo-formed hydrous ferric oxides before (iii) being remobilized during recovery as a result of both dissolution of hydrous ferric oxides and displacement from sorption sites by competing anions.  相似文献

The presence of components of the renin angiotensin system (RAS) and specific receptors of angiotensin II in the female and male reproductive tract supports the hypothesis that reproductive functions may be controlled by RAS. Therefore, the present study investigated the influence of ACE and angiotensins on sperm functions and the sperm-egg interaction. The experiments did not indicate direct effects of ACE on the capacitation process or acrosome reaction. Release of ACE from human spermatozoa during capacitation was not related to their ability to undergo acrosome reaction after stimulation with ionophore. Therefore, ACE release does not seem to be a useful clinical marker for human sperm capacitation. However, decreased binding of human spermatozoa to the oolemma of zonafree hamster oocytes after inhibition of ACE by captopril indicates that kininase II is involved in sperm-egg interactions. In contrast to other studies, incubation with captopril had no influence on sperm binding to the zona pellucida. Because effects of ACE on sperm-egg interactions but not on capacitation or acrosome reaction were observed, several experiments were performed to study the influence of substrates and products on the acrosome reaction. Angiotensin II induced the acrosome reaction dose-dependently, whereas angiotensin I had no effect on the acrosome reaction. The effect of angiotensin II on acrosome reaction seems to be calcium-dependent and mediated by protein kinases. Since a specific type 2 angiotensin II receptor inhibits the acrosome reaction induced by angiotensin II, this subtype of receptors may be present at the surface of sperm heads. Another clue for the presence of type 2 receptors on human spermatozoa is the finding that pertussis toxin did not inhibit the angiotensin II induced acrosome reaction. In contrast to type 1 angiotensin II receptors, type 2 receptors are known to be G-protein independent.  相似文献
