Although skin melanoma (SKM) represents only one-quarter of newly diagnosed skin malignant tumors, it presents a high mortality rate. Hence, new prognostic and therapeutic tools need to be developed. This study focused on investigating the prognostic value of the subcellular expression of BRAF, KRAS, and KIT in SKM in correlation with their gene-encoding interactions. In silico analysis of the abovementioned gene interactions, along with their mRNA expression, was conducted, and the results were validated at the protein level using immunohistochemical (IHC) stains. For IHC expression, the encoded protein expressions were checked on 96 consecutive SKMs and 30 nevi. The UALCAN database showed no prognostic value for the mRNA expression level of KRAS and BRAF and demonstrated a longer survival for patients with low mRNA expression of KIT in SKMs. IHC examinations of SKMs confirmed the UALCAN data and showed that KIT expression was inversely correlated with ulceration, Breslow index, mitotic rate, and pT stage. KRAS expression was also found to be inversely correlated with ulceration and perineural invasion. When the subcellular expression of BRAF protein was recorded (nuclear vs. cytoplasmatic vs. mixed nucleus + cytoplasm), a direct correlation was emphasized between nuclear positivity and lymphovascular or perineural invasion. The independent prognostic value was demonstrated for mixed expression of the BRAF protein in SKM. BRAF cytoplasmic predominance, in association with KIT’s IHC positivity, was more frequently observed in early-stage nonulcerated SKMs, which displayed a low mitotic rate and a late death event. The present study firstly verified the possible prognostic value of BRAF subcellular localization in SKMs. A low mRNA expression or IHC cytoplasmic positivity for KIT and BRAF might be used as a positive prognostic parameter of SKM. SKM’s BRAF nuclear positivity needs to be evaluated in further studies as a possible indicator of perineural and lymphovascular invasion.  相似文献

We initiate a deep study of Riesz MV-algebras which are MV-algebras endowed with a scalar multiplication with scalars from \([0,1]\) . Extending Mundici’s equivalence between MV-algebras and \(\ell \) -groups, we prove that Riesz MV-algebras are categorically equivalent to unit intervals in Riesz spaces with strong unit. Moreover, the subclass of norm-complete Riesz MV-algebras is equivalent to the class of commutative unital C \(^*\) -algebras. The propositional calculus \({\mathbb R}{\mathcal L}\) that has Riesz MV-algebras as models is a conservative extension of ?ukasiewicz \(\infty \) -valued propositional calculus and is complete with respect to evaluations in the standard model \([0,1]\) . We prove a normal form theorem for this logic, extending McNaughton theorem for ? ukasiewicz logic. We define the notions of quasi-linear combination and quasi-linear span for formulas in \({\mathbb R}{\mathcal L},\) and relate them with the analogue of de Finetti’s coherence criterion for \({\mathbb R}{\mathcal L}\) .  相似文献

A theoretical DFT study was employed to confirm the Kolbe-Schmitt reaction mechanism and investigate solvent effects on this reaction. The use of a solvent in the Kolbe-Schmitt reaction is desirable to facilitate a homogeneous reaction mixture and potentially improve the reaction rate. The candidate solvents were designed using computer aided molecular design (CAMD) and tested using DFT solvation calculations. The results from the quantum mechanical calculations were then used to determine the rate constants for each elementary step, the overall reaction yields and the corresponding residence time. The methodology was tested on the reaction without solvent, with solvents reported in the literature, and with the designed solvents. The study revealed that in the presence of solvents with high dielectric constant the reaction becomes reversible, leading to low product yields.  相似文献

Abstract

The enantioselective, potentiometric membrane electrode (EPME) based on maltodextrin with dextrose equivalence (DE) 16.5–19.5 is proposed for the assay of R‐deprenyl. The linear concentration range for the proposed electrode is 10?10?10?3 mol L?1. The slope of the electrode is 53.1 mV per decade of concentration. The detection limit is 3.6×10?11 mol L?1. The proposed electrode could be reliably employed for the assay of R‐deprenyl raw material and its pharmaceutical formulation, Lentogesic tablets.  相似文献

In collaboration with the United States Department of Agriculture and a number of major milling companies, the “Insect-Detect” immunoassay for analyzing insect contamination in grains has been compared with three more traditional methods, X-ray analysis, cracking and flotation, and the insect fragment test (IFT). Testing was carried out in blind fashion using clean wheat samples that were spiked with differing numbers of grain kernels infested with late instar larvae of the granary weevil (Sitophilus granarius (L.)). Three different laboratories analyzed the samples for each of the four methods. The collaborative trials showed that the insect immunoassay clearly provided the most accurate measurement of actual insect infestation, followed by X-ray analysis. While both cracking and flotation and IFT procedures provided a general measure of contamination, they showed much greater variability.  相似文献
