Background: If menopause is really independent risk factor for cardiovascular disease is still under debate. We studied if ovariectomy in the model of insulin resistance causes cardiovascular changes, to what extent are these changes reversible by estradiol substitution and if they are accompanied by changes in other organs and tissues. Methods: Hereditary hypertriglyceridemic female rats were divided into three groups: ovariectomized at 8th week (n = 6), ovariectomized with 17-β estradiol substitution (n = 6), and the sham group (n = 5). The strain of abdominal aorta measured by ultrasound, expression of vascular genes, weight and content of myocardium and also non-cardiac parameters were analyzed. Results: After ovariectomy, the strain of abdominal aorta, expression of nitric oxide synthase in abdominal aorta, relative weight of myocardium and of the left ventricle and circulating interleukin-6 decreased; these changes were reversed by estradiol substitution. Interestingly, the content of triglycerides in myocardium did not change after ovariectomy, but significantly increased after estradiol substitution while adiposity index did not change after ovariectomy, but significantly decreased after estradiol substitution. Conclusion: Vascular and cardiac parameters under study differed in their response to ovariectomy and estradiol substitution. This indicates different effects of ovariectomy and estradiol on different cardiovascular but also extracardiac structures.  相似文献

A process of plasma‐initiated grafting of acrylic acid on commercial porous polypropylene membrane was studied. The influence of parameters of the plasma (power, gas pressure, time plasma‐sample distance, sample arrangement) and grafting (solvent composition, monomer concentration, time, inhibitor presence) on the degree of grafting, amount of homopolymer produced and surface electrical resistance was determined. A degree of grafting up to 18 mmol/g was obtained, which resulted in sample resistance as low as 30 mΩ cm2. The molecular weight of AAc homopolymer that can be assumed as equal to the MW of grafted chains, ranged from 25,000 to 50,000,000 da. SEM and water permeability measurements show that grafting causes filling of the pores, which, however, does not stop K+ ions from penetrating the membrane. We demonstrate that oral delivery of self-assembled nanostructured nanoparticles consisting of 5-fluorouracil (5-FU) lipid prodrugs results in a highly effective, target-activated, chemotherapeutic agent, and offers significantly enhanced efficacy over a commercially available alternative that does not self-assemble. The lipid prodrug nanoparticles have been found to significantly slow the growth of a highly aggressive mouse 4T1 breast tumour, and essentially halt the growth of a human MDA-MB-231 breast tumour in mouse xenografts. Systemic toxicity is avoided as prodrug activation requires a three-step, enzymatic conversion to 5-FU, with the third step occurring preferentially at the tumour site. Additionally, differences in the lipid prodrug chemical structure and internal nanostructure of the nanoparticle dictate the enzymatic conversion rate and can be used to control sustained release profiles. Thus, we have developed novel oral nanomedicines that combine sustained release properties with target-selective activation.  相似文献

The blends of pectin (PEC) and poly(vinyl alcohol) (PVA) at different components ratios were prepared by mixing in water. Thin polymeric films of PEC/PVA blends and pure polymers were obtained by casting method. All samples were then artificially aged using Suntest apparatus (Atlas) up to 780 h. The changes in chemical structure during sample ageing have been monitored by infrared and ultraviolet‐visible absorption spectroscopies. The first stage of weathering (up to ~ 300 h) was very slow and alteration of chemical structure was negligible in all samples. Prolonged ageing (>300 h) caused more significant degradation processes. FTIR spectra exhibited the highest changes in hydroxyl and carbonyl band ranges indicating the efficient photooxidation of macromolecules. The mechanisms of the observed processes have been discussed. It was found that PVA undergoes faster photoxidative degradation than pectin aged at the same conditions. The PEC/PVA blends exhibited the improved resistance to weathering comparing with both polymers aged individually. Mutual stabilization effect can be explained by intermolecular interactions between PEC and PVA confirmed by spectroscopic methods. © 2011 Wiley Periodicals, Inc. J Appl Polym Sci, 2011  相似文献

Micro-Raman light scattering experiments on PbZrO3 (PZO) single crystal doped with Nb5+ have been investigated. Special attention was paid to the paraelectric (PE) phase in which nominally forbidden first-order Raman spectra were detected at temperatures far above the phase transition TC. Complex Raman spectra were observed in the vicinity of three structural phase transitions. These results mainly from the coexistence of phases with different symmetries in a wide temperature range below TC. The Raman measurements have been compared with dielectric and optical observations and proved that polar nanoregions in a centrosymmetric lattice appear well above TC. It was shown that doping ABO3 perovskites with heterovalent ions like Nb5+ unbalances charge neutrality of the lattice and strongly extends the temperature range of polar regions. The investigations performed point out that in the PE matrix the interaction between electrons and lattice vibrations, as recently suggested for pure PZO, plays an important role.  相似文献

Even though immunotherapy has radically changed the search for anticancer therapies, there are still many different pathways that are open to intervention with traditional small molecules. To expand our investigation in the anticancer field, we report here a new series of compounds in which our previous pyrazole and imidazopyrazole scaffolds are linked to a differently decorated phenyl ring through an acylhydrazone linker. Preliminary tests on the library were performed at the National Cancer Institute (USA) against the full NCI 60 cell panel. The best compounds among the imidazopyrazole series were then tested by immunofluorescence staining for their inhibition of cell proliferation, apoptosis induction, and their effect on the cell cycle and on microtubules. Two compounds, in particular 4-benzyloxy-3-methoxybenzyliden imidazopyrazole-7-carbohydrazide showed good growth inhibition, with IC50 values in the low-micromolar range, and induced apoptosis. Both compounds altered the cell-cycle phases with the appearance of polyploid cells. Immunofluorescence analysis evidenced microtubules alterations; tubulin polymerization assays and docking studies suggested the tubulin system to be the possible, although not exclusive, target of the new acylhydrazone series reported here.  相似文献

A two‐step procedure was applied to obtain antimicrobial films; this procedure involved a corona treatment of the polyethylene (PE) surface and its chemical activation with 1‐ethyl‐3‐[3‐dimethylaminopropyl] carbodiimide hydrochloride and N‐hydroxysuccinimide, and this led to the covalent bonding of chitosan on the PE surface. Electrochemical methods were used to investigate the stability of the deposited chitosan layer. The potentiometric and polyelectrolyte titrations showed that some amount of chitosan desorbed faster from the surface until equilibrium was reached and also that the grafted chitosan layer was more stable than the physically adsorbed one. The chitosan immobilized on the PE surface exhibited the expected antibacterial activity when tested against three bacteria, which included two Gram‐negative bacteria, Salmonella enteritidis and Escherichia coli, and one Gram‐positive bacterium, Listeria monocytogenes. © 2013 Wiley Periodicals, Inc.
