Consumption of protein hydrolysates has been proposed to stimulate muscle anabolism more than intact (nonhydrolyzed) proteins via accelerated delivery of amino acids for muscle protein synthesis (MPS). We evaluated whether the rate of amino acid uptake and transport across intestinal cells was enhanced for soy protein hydrolysates versus nonhydrolyzed soy protein. Intact and partially hydrolyzed proteins were subject to simulated gut digestion and applied to the apical surface of Caco‐2 monolayers. Basolateral media was harvested after 3 h and quantitatively analyzed for free amino acids using ion‐exchange chromatography and comparison to an included reference standard. Basolateral concentrations of all amino acids were higher (mean 32%) for hydrolyzed versus nonhydrolyzed protein with the greatest differences in histidine, lysine, and valine. Scale‐up production of the soy protein hydrolysate did not diminish its enhanced absorption properties. These data support the hypothesis that hydrolyzed soy protein may provide dietary amino acids that are more rapidly transported across the intestinal epithelium versus intact soy protein. This would be important under conditions where rapid and increased levels of amino acids are needed such as in the stimulation of MPS.  相似文献

The chemical composition of black teas (Camellia sinensis L) varies when the green leaf from which they are manufactured is withered physically to the same degree but for different periods. Caffeine contents progressively increase with prolonged withering time whereas theaflavins decrease due to prolonged withering. Although the sums of group I and group II volatile flavour compounds (VFC) decrease with prolonged withering time, the flavour index (expressed as group II VFC/group I VFC) increases due to prolonged withering. No significant changes were noted in the thearubigin levels with variation in withering time, but the tasters' evaluations were at a maximum for teas withered for 14 h. PURPOSE: A new dietary zinc assessment tool (ZAT) was evaluated to determine its usefulness in estimating zinc intakes among college students. METHODS: A food frequency questionnaire specifically designed for calculating average zinc intakes was administered to university students who had also completed three-day food records. Zinc intakes from the two instruments were compared. RESULTS: Among 171 participants (38 male, 133 female) aged 20.2 +/- 1.6 years (mean +/- standard deviation), the two dietary assessment methods were positively correlated (r=0.33, p<0.001). The ZAT correctly identified 76% of the women who were obtaining less than the National Academy of Sciences Recommended Dietary Allowance for zinc (8 mg/day for females, 11 mg/day for males). CONCLUSIONS: This convenient tool may assist in the identification of problematic dietary patterns at an early stage. Further design modifications and expanded studies are warranted.  相似文献

In central Florida, alligators (Alligator mississippiensis) inhabiting lakes contaminated with organochlorine pesticides (OCPs) produce eggs that have high OCP concentrations and low clutch viability (proportion of eggs in a clutch that yield a live hatchling) compared to those from less contaminated lakes (reference lakes). However, a clear dose-response relationship has not been established between OCPs and poor clutch viability. In order to better elucidate a cause and effect relationship between OCP exposure and clutch viability, we conducted concurrent field and laboratory studies. Our field study reaffirmed that eggs of wild alligators from OCP-contaminated lakes and wetlands continue to have lower clutch viability and higher OCP burdens than eggs from reference lakes. Our field study also demonstrated that OCP egg burdens were strongly correlated with clutch viability for some of the OCP-contaminated sites, but not all. To better test causal relationships, a parental exposure study was conducted using captive adult alligators. Our laboratory study demonstrated that dietary exposure of captive alligators to an ecologically relevant OCP mixture caused alligators to produce eggs with higher OCP burdens and reduced clutch viability, as compared to the captive-control population. The experimentally induced egg burdens and clutch viability reductions were similar to those of wild alligators from OCP-contaminated sites. Our field and laboratory results suggest parental OCP exposure may be contributing to low clutch viability in wild alligators inhabiting OCP-contaminated habitats, raising some concern for endangered crocodilians living in OCP-contaminated habitats.  相似文献

This study is one of the very few investigating the dioxin body burden of a group of child-bearing-aged women at an electronic waste (e-waste) recycling site (Taizhou, Zhejiang Province) (24 +/- 2.83 years of age, 40% were primiparae) and a reference site (Lin'an city, Zhejiang Province, about 245 km away from Taizhou) (24 +/- 2.35 years of age, 100% were primiparae) in China. Five sets of samples (each set consisted of human milk, placenta, and hair) were collected from each site. Body burdens of people from the e-waste processing site (human milk, 21.02 +/- 13.81 pg WHO-TEQ1998/g fat (World Health Organization toxic equivalency 1998); placenta, 31.15 +/- 15.67 pg WHO-TEQ1998/g fat; hair, 33.82 +/- 17.74 pg WHO-TEQ1998/g dry wt) showed significantly higher levels of polychlorinated dibenzo-p-dioxins and polychlorinated dibenzofurans (PCDD/ Fs) than those from the reference site (human milk, 9.35 +/- 7.39 pg WHO-TEQ1998/g fat; placenta, 11.91 +/- 7.05 pg WHO-TEQ1998/g fat; hair, 5.59 +/- 4.36 pg WHO-TEQ1998/g dry wt) and were comparatively higher than other studies. The difference between the two sites was due to e-waste recycling operations, for example, open burning, which led to high background levels. Moreover, mothers from the e-waste recycling site consumed more foods of animal origin. The estimated daily intake of PCDD/Fs within 6 months by breastfed infants from the e-waste processing site was 2 times higher than that from the reference site. Both values exceeded the WHO tolerable daily intake for adults by at least 25 and 11 times, respectively. Our results implicated that e-waste recycling operations cause prominent PCDD/F levels in the environment and in humans. The elevated body burden may have health implications for the next generation.  相似文献

Oxidation of arsenite, As(III), to arsenate, As(V), is required for the efficient removal of arsenic by many water treatment technologies. The photocatalyzed oxidation of As(III) on titanium dioxide, TiO2, offers an environmentally benign method for this unit operation. In this study, we explore the efficacy and mechanism of TiO2-photocatalyzed As(III) oxidation at circumneutral pH and over a range of As(III) concentrations approaching those typically encountered in water treatment systems. We focus on the effect of As adsorption on observed rates of photooxidation. Adsorption (in the dark) of both As(III) and As(V) on Degussa P25 TiO2 was examined at pH 6.3 over a range in dissolved arsenic concentrations, [As]diss, of 0.10-89 microM and 0.2 or 0.05 g L(-1) TiO2 for As(III) and As(V), respectively. Adsorption isotherms generally followed the Langmuir-Hinshelwood model with As(III) exhibiting an adsorption maxima of 32 micromol g(-1). As(V) adsorption did not reach a plateau under the experimental conditions examined; the maximum adsorbed concentration observed was 130 micromol g(-1). The extent of As(III) and As(V) adsorption observed at the beginning and end of the kinetic studies was consistent with that observed in the adsorption isotherms. Kinetic studies were performed in batch systems at pH 6.3 with 0.8-42 microM As(III) and 0.05 g L(-1) TiO2; complete oxidation of As(III) was observed within 10-60 min of irradiation at 365 nm. The observed effect of As(III) concentration on reaction kinetics was consistent with surface saturation at higher concentrations. Addition of phosphate at 0.5-10 microM had little effect on either As(III) sorption or its photooxidation rate but did inhibit adsorption of the product As(V). The selective use of hydroxyl radical quenchers and superoxide dismutase demonstrated that superoxide, O2-, plays a major role in the oxidation of As(III) to As(V).  相似文献

Seven chemical methods, namely peroxide value (PV), totox value (TV), anisidine value (AV), conjugable oxidation products (COP), oxodiene value (OV), induction period (IP), iodine value (IV) and a sensory analytical procedure (flavour score, FS) have been used in evaluating the oxidation state of groundnut oil heated at 100°C for varying lengths of time up to 20 h. As oxidation progressed, PV, TV, AV, COP and OV increased. IP and IV decreased with oxidation while FS showed a progressive deterioration on a seven-point scale from bland to very rancid. On the basis of sensitivity to oxidative changes, five of the methods (PV, TV, IP, IV and FS) were found to be satisfactory. However, the best correlation with flavour scores were obtained in the case of IP, IV and OV while AV and COP correlated poorly with FS. Three methods (PV, IP and IV) best satisfied the combined criteria of sensitivity to oxidative changes and correlation with flavour.  相似文献

BACKGROUND: Pears are exported in large quantities from South Africa, resulting in large revenues. Minimisation of quality losses once the fruit has reached the export destination is as important as following strict export and distribution protocols. Kafirin can form edible films. In this study an edible coating made from 20 g kg?1 kafirin coating solution was applied as a postharvest treatment to retard quality deterioration of ‘Packham's Triumph’ pears during storage at the typical ripening temperature (20 °C). Changes in physicochemical and sensory quality were monitored over a period of 24 days. RESULTS: The kafirin coating was unable to retard the onset of ripening but decreased the respiration rate and retarded the progression of senescence. However, moisture loss was exacerbated in the kafirin‐coated fruit during ripening at 20 °C, especially towards the end of the shelf‐life. CONCLUSION: The kafirin coating extended the eat‐ripe quality of the pears by 1–2 weeks. However, the appearance of the fruit was unacceptable after 14 days of storage in terms of wrinkled skin. Further work is needed to improve the water barrier properties of the kafirin coating by incorporating a wax or triglyceride into the coating formulation or more simply by applying a kafirin coating to waxed fruit. Soil freezing is a disturbance of the below ground environment, potentially resulting in increased losses of NO3- and surface water acidification. Here, we report the effects of soil freezing on interannual variation in stream chemistry at the Hubbard Brook Experimental Forest, New Hampshire. Data from 1970 to 1997 of soil frost depth, snow cover, precipitation, air temperature, and stream discharge and chemistry were used in a stepwise linear regression model to select the variables that best predicted deviations of annual stream concentrations from 4-year running averages. Variables quantifying soil freezing severity were selected as significant predictors of short-term fluctuations in stream K+, NO3-, Ca2+, and Mg2+ concentrations from 1970 to 1989, explaining 59 and 47% of the short-term variability in K+ and NO3-, respectively. Fine-root mortality and disturbance of root-soil-microbe interactions, with subsequent effects on decomposition and nutrient uptake, likely contributed to the mobilization of K+ and NO3- to streamwater following severe soil freezing events. The relationship between soil freezing and stream chemistry, however, weakened during the period 1990-1997. Because soil freezing has had inconsistent effects on stream chemistry during the period 1970-1997, it is unclear whether future changes in the frequency, duration, and depth of soil freezing events as the result of changes in the snow cover regime under a warmer climate will have significant impacts on the losses of NO3- and nutrient-base cations from temperate northern ecosystems.  相似文献

Nicotine intake via smoking is highly variable. Individualized dosing of nicotine replacement therapy (NRT) may improve product efficacy, but a better understanding of the within-day and within-subject relationships between smoking, NRT use, nicotine and cotinine concentrations in blood, and cravings and withdrawal symptoms is needed to inform dosing algorithms. A pilot study was undertaken to collect data on these relationships and to assess the feasibility of the methods needed for this type of research, including a sophisticated statistical modeling technique (a two-part mixed-effects model with correlated random effects that accounts for clumping at zero). Because nicotine metabolism varies by gender and race, the sample was homogeneous with respect to these characteristics. In a within-subjects study, 27 African American adult male smokers carried a computerized cigarette dispenser for 1 week, capturing the time each cigarette was smoked. Subjects then entered an inpatient setting for 1 day of scheduled smoking (matched to data from the cigarette dispenser to create an ecologically valid schedule) and 4 days of ad libitum nicotine nasal spray use, while tobacco abstinent. Eight times per day, at 2-hour intervals, blood was drawn and ratings of cigarette cravings and withdrawal symptoms were obtained. On average, subjects used less than half of the manufacturer's recommended minimum daily dose of nicotine nasal spray. Large differences in nicotine and cotinine levels were observed between individuals. When predicting nicotine, cotinine, withdrawal, and cravings, we observed significant interactions between route of nicotine intake and a variety of independent variables.  相似文献
