The effect of cooking on proteins from acha and durum wheat was assessed from an analysis of protein extractability, gel electrophoretic profiles, in-vitro protein digestibility (IVPD) and the amino acid compositions of wholemeal flour and residue proteins. Heating wholemeal flour samples at 100–140°C (t = 10–40 min) resulted in 0–30% and 45–55% decreases in acha and durum protein solubility, respectively. In general, high molecular weight (30–70 k Da) protein subunits were more susceptible to heat damage. For both cereals, sodium dodecyl sulphate (SDS; 10 g litre?1) and/or dithiothrcitol (DTT; 10 mM ) increased protein solubility in unheated and heated samples. The IVPD index was 90–91% and was not significantly altered by cooking (100–120°C, t = 40 min). Cooking at extreme temperatures (140°C, t = 40 min) reduced the IVPD by 8% (P = 0.05). Osborne fractionation resulted in a durum or acha residue level of 7.8% or 55.2%. Treatment with solvent containing propanol, SDS and/or DTT at room temperature followed by SDS-polyacrylamide gel electrophoresis of non-solubilised proteins showed that the glutelin fraction of acha, with the exception of a 65 kDa subunit, was insoluble owing to strong inter-subunit hydrophobic and disulphide interactions. Wholemeal acha flour and residue protein showed a significantly greater level of hydrophobic and sulphur amino acids as well as glutamine which is associated with H-bonding. The possibility that cereal protein solubility is also dependent on protein-carbohydrate links is discussed.  相似文献

Literature with data from dairy cows on pasture was reviewed to evaluate the effects of supplementation on intake, milk production and composition, and ruminal and postruminal digestion. Low dry matter intake (DMI) of pasture has been identified as a major factor limiting milk production by high producing dairy cows. Pasture DMI in grazing cows is a function of grazing time, biting rate, and bite mass. Concentrate supplementation did not affect biting rate (58 bites/min) or bite mass (0.47 g of DM/bite) but reduced grazing time 12 min/d per kilogram of concentrate compared with unsupplemented cows (574 min/d). Substitution rate, or the reduction in pasture DMI per kilogram of concentrate, is a factor which may explain the variation in milk response to supplementation. A negative relationship exists between substitution rate and milk response; the lower the substitution rate the higher the milk response to supplements. Milk production increases linearly as the amount of concentrate increases from 1.2 to 10 kg DM/d, with an overall milk response of 1 kg milk/kg concentrate. Compared with pasture-only diets, increasing the amount of concentrate supplementation up to 10 kg DM/d increased total DMI 24%, milk production 22%, and milk protein percentage 4%, but reduced milk fat percentage 6%. Compared with dry ground corn, supplementation with nonforage fiber sources or processed corn did not affect total DMI, milk production, or milk composition. Replacing ruminal degradable protein sources with ruminal undegradable protein sources in concentrates did not consistently affect milk production or composition. Forage supplementation did not affect production when substitution rate was high. Fat supplementation increased milk production by 6%, without affecting milk fat and protein content. Increasing concentrate from 1.1 to 10 kg DM/d reduced ruminal pH 0.08 and NH3-N concentration 6.59 mg/dl, compared with pasture-only diets. Replacing dry corn by high moisture corn, steam-flaked or steam-rolled corn, barley, or fiber-based concentrates reduced ruminal NH3-N concentration 4.36 mg/dl. Supplementation did not affect in situ pasture digestion, except for a reduction in rate of degradation when high amounts of concentrate were supplemented. Supplementation with energy concentrates reduced digestibility of neutral detergent fiber and intake of N but did not affect digestibility of organic matter or flow of microbial N.  相似文献

Two experiments were conducted to evaluate steam-flaked corn and nonforage fiber sources as supplemental carbohydrates for lactating dairy cows on pasture. Cows were allotted to a new paddock of an orchardgrass (Dactylis glomerata L.) pasture twice daily in one group in both trials. In experiment 1, 28 Holstein cows, averaging 216 d in milk, were randomly assigned to either a cracked-corn (CC) or a steam-flaked (SFC) supplement in a split plot design. The supplement contained 66.7% of corn and a protein/mineral pellet. In experiment 2, 28 Holstein cows, averaging 182 d in milk, were randomly assigned to either a ground corn (GC) or a nonforage fiber (NFF)-based supplemented in a single reversal design. The GC supplement contained 85% ground corn plus protein, mineral, and vitamins. The NFF supplement contained 35% ground corn, 18% beet pulp, 18% soyhulls, 8% wheat middlings plus protein, mineral, and vitamins. In both experiments, cows were fed the grain supplement twice daily after each milking at 1 kg/4 kg milk. In experiment 1, milk production (24.3 kg/d) and composition did not differ between treatments; however, plasma and milk urea N were lower with the SFC supplement. In experiment 2, milk production (27.5 kg/d) was not affected by treatments, which may be related to the medium quality of pasture grazed. The GC supplement tended to reduce plasma and milk urea N and increased milk protein percentage (3.23 vs. 3.19%). Pasture dry matter intake, measured using Cr2O3, did not differ between treatments in either experiment 1 (15.1 kg/d) or experiment 2 (12.2 kg/d). Milk production did not differ when mid-late lactation cows on pasture were supplemented with SFC or NFF instead of dry corn.  相似文献

Three configurations have been developed to improve the understanding of structural element interactions in food material during deformation. The three configurations combine an inverted confocal scanning laser microscope (CSLM) and a cell that can apply to the sample a specific deformation: continuous shear, linear oscillatory shear and biaxial extension (compression).In the continuous shear and oscillatory shear configurations (OSCs), a zero-velocity plane is created in the sample by moving two plates in opposite direction, maintaining stable observation conditions of the structural behaviour under deformation. The OSC allows simultaneous application of CSLM and diffusing wave spectroscopy, a multiple light scattering technique. The third configuration (compression configuration) allows observation at a stagnation point during rheometric measurements. The configurations accept semi-liquid products (dressing, sauces, dairy products, etc.) for investigations in area such as aggregation, gelation, interactions at interface, coalescence, break-up.  相似文献

Two phenolic compounds, p-coumaric acid and feruloyl-arabinose, were localised by immunocytochemistry in the cell walls of the apical internode of two lines of maize (Co125 and W401) of different digestibility. The compounds were detected at two stages of cell maturity in the lignified tissues (sclerenchyma, fibres and xylem) and in the medullary parenchyma, which, in the samples studied, was not lignified. p-Coumaric acid is a phenolic acid associated with lignins, which confer resistance on plant cell walls to microbial degradation in the rumen. Feruloyl-arabinose is a compound associated with xylans, the principal hemicelluloses in Gramineae, which are potentially degradable. Labelling of p-coumaric acid decreased in both maize lines with cell age and as the cell walls became lignified. The mass of lignin deposited in the cell walls masked p-coumaric acid, thereby making it less accessible to the antibodies. There was an inverse relationship in the labelling of p-coumaric acid and feruloyl-arabinose. Feruloyl-arabinose was more heavily labelled as the plant cell walls matured in all the lignified tissues of both maize lines and in the parenchyma of the less digestible line. To probe the reactivity of 2,4,6-trimethylphenol with humic triplet excited states, we investigated its influence on the humic substances-mediated photooxygenation offurfuryl alcohol. Elliott soil humic and fulvic acids were employed for these experiments. When added in the concentration range of 10(-4) - 10(-3) M, 2,4,6-trimethylphenol inhibited furfuryl alcohol photooxygenation to an extent depending on its concentration. The inhibiting effect decreased as the oxygen concentration was increased. By postulating that 2,4,6-trimethylphenol competes with oxygen for reaction with humic triplet excited states and with furfuryl alcohol for reaction with singlet oxygen, we obtained kinetic laws describing the consumption profiles of furfuryl alcohol and 2,4,6-trimethylphenol. Experimental rates of 2,4,6-trimethylphenol and furfuryl alcohol loss could be satisfactorily fitted with 1.09-1.16 for the ratio k2/k3, where k2 and k3 are the reaction rate constants of humic triplet excited states with oxygen and 2,4,6-trimethylphenol, respectively. These types of experiments could be extended to a variety of substrates to measure their reaction rate constants with humic triplet excited states.  相似文献

The objective of this study was to determine i) if Camembert cheese micro-organisms could be detected in fecal samples after regular consumption by human subjects and ii) the consequence of this consumption on global metabolic activities of the host colonic microbiota. An open human protocol was designed where 12 healthy volunteers were included: a 2-week period of fermented products exclusion followed by a 4-weeks Camembert ingestion period where 2x40 g/day of Camembert cheese was consumed. Stools were collected from the volunteers before consumption, twice during the ingestion period (2nd and 4th week) and once after a wash out period of 2 weeks. During the consumption of Camembert cheese, high levels of Lactococcus lactis and Leuconostoc mesenteroides were measured in fecal samples using real-time quantitative PCR, reaching median values of 8.2 and 7.5 Log(10) genome equivalents/g of stool. For Ln. mesenteroides, persistence was observed 15 days after the end of Camembert consumption. The survival of Geotrichum candidum was also assessed and the fecal concentration reached a median level of 7.1 Log(10) CFU/g in stools. Except a decreasing trend of the nitrate reductase activity, no significant modification was shown in the metabolic activities during this study.  相似文献

Current temperature tracking systems lack the convenience and accuracy demanded by the real conditions of a fast-paced produce supply chain. In recent years RFID technology has been suggested to be an enhanced method for temperature tracking because of its many benefits, such as using little instrumentation, offering the quick readings necessary for real-time decision making, and allowing the capture of long-duration temperature profiles. However its limitation lies in that probeless systems fail to provide accurate temperature readings in some of the critical points of the pallet and the load. The objective of this work was to study the use of RFID in temperature monitoring by comparing the performance of RFID temperature tags versus conventional temperature tracking methods, as well as RFID temperature tags with probe versus RFID temperature tags without probes and their utilization along the supply chain. Therefore, the temperature mapping of a shipping trial comprising pallets of crownless pineapples instrumented using different RFID temperature dataloggers and traditional temperature dataloggers and packed in two kinds of packages (corrugated boxes and reusable plastic containers) inside a container was performed. The results showed that RFID temperature tags are analogous with regards to accuracy to the conventional methods, but have a superior performance because they allow quick instrumentation and data recovery, and the possibility of accessing the sensor program and data at any point of the supply chain without line of sight. In addition, the use of RFID tags with probe was justified by its role in determining the efficiency of the pre-cooling operations and low temperature abuse tracking during transportation and refrigerated storage; while the RFID tags without probe proved useful for high temperature abuse monitoring during transportation and refrigerated storage. The creation of a RFID sensor with a probe, able to record both ambient and pulp temperatures simultaneously is suggested.
