Pactamycin is a bacteria‐derived aminocyclitol antibiotic with a wide‐range of biological activity. Its chemical structure and potent biological activities have made it an interesting lead compound for drug discovery and development. Despite its unusual chemical structure, many aspects of its formation in nature remain elusive. Using a combination of genetic inactivation and metabolic analysis, we investigated the tailoring processes of pactamycin biosynthesis in Streptomyces pactum. The results provide insights into the sequence of events during the tailoring steps of pactamycin biosynthesis and explain the unusual production of various pactamycin analogues by S. pactum mutants. We also identified two new pactamycin analogues that have better selectivity indexes than pactamycin against malarial parasites.  相似文献

Pt-decorated CdSe tetrapods with different arm lengths were tested for the photocatalytic hydrogen generation reaction. Well-defined CdSe tetrapods with controlled wurtzite arm lengths were synthesized by the continuous precursor injection (CPI) approach. Pt nanocrystals with an extremely small size of ~1 nm were directly decorated on the overall surfaces of CdSe tetrapods. Ligand-exchanged Pt-decorated CdSe tetrapods with different arm lengths were employed as photocatalysts for photocatalytic hydrogen generation reaction in the presence of hole scavengers. Pt-decorated CdSe tetrapods with shorter arm length showed the highest photocatalytic efficiency, which is due to higher probability of charge separation.  相似文献

With the advent of modern bioengineering tools, photosynthetic organisms are increasingly being engineered to produce chemicals from CO2 sources, thereby creating a potential route of sustainable chemical production. Cyanobacteria have evolved a carbon‐concentrating mechanism (CCM) that enables growth at low‐environmental carbon concentrations. However at high‐carbon concentrations these benefits may not outweigh synthesis costs. Here, mass transport and kinetic modeling analyses were performed on two species of cyanobacteria as well as a hypothetical no‐CCM mutant. Modeling results correlated with published experimental data. Three conclusions were drawn from the analysis. Carboxysome geometry was unimportant due to the fast relative rate of diffusion of carbon species. Interspecies variations were largely due to active transporters. The no‐carboxysome cell approaches the wild‐type at 10% CO2. Therefore, in high CO2 environments the carboxysome and active bicarbonate transporters provide no benefit and a metabolic advantage could be achieved by eliminating the energy‐intensive CCM proteins. © 2014 American Institute of Chemical Engineers AIChE J, 60: 1269–1277, 2014  相似文献

The axial distribution of large biomass particles in bubbling fluidized beds comprised of sand and biomass is investigated in this study. The global and local pressure drop profiles are analyzed in mixtures fluidized at superficial gas velocities ranging from 0.2 to 1 m/s. In addition, the radioactive particle tracking technique is used to track the trajectory of a tracer mimicking the behavior of biomass particles in systems consisting of 2, 8, and 16% of biomass mass ratio. The effects of superficial gas velocity and the mixture composition on the mixing/segregation of the bed components are explored by analyzing the circulatory motion of the active tracer. Contrary to low fluidization velocity (U = 0.36 m/s), biomass circulation and distribution are enhanced at U = 0.64 m/s with increasing the load of biomass particles. Foraging success of parasitoids depends on the utilization of reliable information on the presence of their often, inconspicuous hosts. These parasitic wasps use herbivore-induced plant volatiles (HIPVs) that provide reliable cues on host presence. However, host searching of hyperparasitoids, a group of parasitoids that parasitize the larvae and pupae of other parasitoids, is more constrained. Their hosts do not feed on plants, and often are even concealed inside the body of the herbivore host. Hyperparasitoids recently have been found to use HIPVs of plants damaged by herbivore hosts in which the parasitoid larvae develop. However, hyperparasitoids that search for these parasitoid larvae may be confronted with healthy and parasitized caterpillars on the same plant, further complicating their host location. In this study, we addressed whether the primary hyperparasitoid Baryscapus galactopus uses caterpillar body odors to discriminate between unparasitized herbivores and herbivores carrying larvae of parasitoid hosts. We show that the hyperparasitoids made faster first contact and spent a longer mounting time with parasitized caterpillars. Moreover, although the three parasitoid hosts conferred different fitness values for the development of B. galactopus, the hyperparasitoids showed similar behavioral responses to caterpillar hosts carrying different primary parasitoid hosts. In addition, a two-chamber olfactometer assay revealed that volatiles emitted by parasitized caterpillars were more attractive to the hyperparasitoids than those emitted by unparasitized caterpillars. Analysis of volatiles revealed that body odors of parasitized caterpillars differ from unparasitized caterpillars, allowing the hyperparasitoids to detect their parasitoid host.  相似文献

The crystallization kinetics of poly(L -lactide-co-meso-lactide) were determined over a range of 0% to 9% mesolactide. The kinetics were fit to the nonlinear Avrami equation and then to the Hoffman–Lauritzen equation modified for optical copolymers. The theory was found to fit the data well. The crystallization half-time was found to increase about 40% for every 1 wt % meso-lactide in the polymerization mixture. The change in crystallization rate is driven mainly by the reduction in melting point for the copolymers. The copolymer crystallization kinetics were also determined in the presence of talc, a nucleating agent for polylactide. The theory again fit the data well, using the same growth parameters and accounting for the talc only through the nucleation density term. © 1996 John Wiley & Sons, Inc.  相似文献

A novel Saccharomyces cerevisiae gene, HST1, was identified from among anonymous cDNAs and the complete corresponding genomic clone was isolated and sequenced. HST1 is very closely related to SIR2, showing 71% sequence identity over 84% of its length. Polymerase chain reaction with degenerate primers on S. cerevisiae DNA identified three additional SIR2-related genes designated HST2, HST3 and HST4. The sequences of HST2, HST3 and HST4 correspond to sequences previously released by the S. cerevisiae genome sequencing project as U33335, NCBI gi:965078; X87331, NCBI gi:829135; and Z48784, YD9346.03, respectively. Disruption of HST1 has shown no phenotype with respect to mechanisms in which SIR2 has a role, namely, regional silencing of HMLα, or in rDNA recombination. The sequence of HST1 has been deposited in the DDBJ, EMBL and GenBank at NCBI database under Accession Number L47120.  相似文献

We have applied the novel method of hot filament-activated chemical vapour deposition (HFCVD) for low-temperature deposition of a variety of nitride thin films. In this paper the results from our recent work on aluminium, silicon and titanium nitride have been reviewed. In the HFCVD method a hot tungsten filament (1500–1850°C) was utilised to decompose ammonia in order to deposit nitride films at low substrate temperatures and high rates. The substrate temperatures ranged from 245 to 600°C. The film properties were characterised by a number of analytical and optical methods. The effect of various deposition conditions on film properties was studied. All the films obtained were of high chemical purity and had very low or no detectable tungsten contamination from the filament metal.  相似文献

Resolvin (Rv) D2 and RvD1 are biosynthesized from docosahexaenoic acid (DHA) and promote resolution of inflammation in multiple organs and tissues, including the conjunctiva. Histamine is a mediator produced by mast cells in the conjunctiva during the allergic response. We determined the interaction of RvD2 with histamine and its receptor subtypes in cultured conjunctival goblet cells and compared them with RvD1 by measuring intracellular [Ca2+] and mucous secretion. Treatment with RvD2 significantly blocked the histamine-induced [Ca2+]i increase as well as secretion. RvD2 and RvD1 counter-regulate different histamine receptor subtypes. RvD2 inhibited the increase in [Ca2+]i induced by the activation of H1, H3, or H4 receptors, whereas RvD1 inhibited H1 and H3 receptors. RvD2 and RvD1 also activate distinct receptor-specific protein kinases to counter-regulate the histamine receptors, probably by phosphorylation. Thus, our data suggest that the counter-regulation of H receptor subtypes by RvD2 and RvD1 to inhibit mucin secretion are separately regulated.  相似文献

The reactions of combustion products (CO, H2O) from the insensitive high explosive LX-17 (92.5 wt% 1,3,5-triamino-2,4,6-trinitrobenzene (TATB), 7.5 wt% Kel-F 800 binder) with molten metal (75 a%Nd, 25 a% Fe) were studied, to determine their contribution to the energy release rate of deflagrating LX-17, Kinetic data taken with gas mixtures of CO and H2O, and larger-scale tests with LX-17, showed that the reactions were slow and incomplete and that there was no significant contribution to the energy release rate. From the tests with JX-17, the effective flame temperature and burn velocity were dependent on confinement of gas products. LX-17 preheated to 500 K - 510 K showed effective flame temperatures of 1100 K - 1400 K and burn rates of 0.08–0.11 mm/s, depending on the extent of gas confinement by the molten metal.  相似文献
