N-acetyl-p-aminophenol (APAP)-induced liver damage is associated with upregulation of Interleukin-11 (IL11), which is thought to stimulate IL6ST (gp130)-mediated STAT3 activity in hepatocytes, as a compensatory response. However, recent studies have found IL11/IL11RA/gp130 signaling to be hepatotoxic. To investigate further the role of IL11 and gp130 in APAP liver injury, we generated two new mouse strains with conditional knockout (CKO) of either Il11 (CKOIl11) or gp130 (CKOgp130) in adult hepatocytes. Following APAP, as compared to controls, CKOgp130 mice had lesser liver damage with lower serum Alanine Transaminase (ALT) and Aspartate Aminotransferase (AST), greatly reduced serum IL11 levels (90% lower), and lesser centrilobular necrosis. Livers from APAP-injured CKOgp130 mice had lesser ERK, JNK, NOX4 activation and increased markers of regeneration (PCNA, Cyclin D1, Ki67). Experiments were repeated in CKOIl11 mice that, as compared to wild-type mice, had lower APAP-induced ALT/AST, reduced centrilobular necrosis and undetectable IL11 in serum. As seen with CKOgp130 mice, APAP-treated CKOIl11 mice had lesser ERK/JNK/NOX4 activation and greater features of regeneration. Both CKOgp130 and CKOIl11 mice had normal APAP metabolism. After APAP, CKOgp130 and CKOIl11 mice had reduced Il6, Ccl2, Ccl5, Il1β, and Tnfα expression. These studies exclude IL11 upregulation as compensatory and establish autocrine, self-amplifying, gp130-dependent IL11 secretion from damaged hepatocytes as toxic and anti-regenerative.  相似文献

This research focuses on Nafion modification using plasma techniques for direct methanol fuel cell applications. The results indicated the both argon (Ar) and carbon tetrafluoride (CF4) plasma treatments modified the Nafion surface substantially without altering the bulk properties. The Nafion surface exposed to CF4 plasma resulted in a more hydrophobic layer and an even lower MeOH permeability than the Ar-treated membrane. The plasma operating conditions using CF4 were optimized by utilizing an experimental design. The minimum MeOH permeability was reduced by 74%. The conductivity was 1–2×10-3 S/cm throughout the entire experimental range. Suppressed MeOH permeability can be achieved while maintaining the proton conductivity at a satisfactory level by adjusting the plasma operating conditions.  相似文献

We present a phase plate loading system developed for a commercial transmission electron microscope (TEM). Our system can be installed without modifying the optical design of the TEM. This system is equipped with a loading monitoring set that allows users to easily and safely locate the phase plate between the pole pieces, and also comes with an airlock that permits quick loading of a phase plate without the need to re-vent the TEM column. The system uses a home-made three-axis nano-positioner to precisely position the phase plate hole at the desired location. Our system has a precision of ∼10 nm, an improvement of one order of magnitude compared with the precision of a phase plate holder modified from an objective aperture. We demonstrate the successful installation and the use of the loading system to place a phase plate at the desired position. Our phase plate loading system can be used to accommodate various types of phase plates and thus provides a good way to greatly speed up the development of TEM phase plates.  相似文献

In this study, a novel polymer electrolyte composite membrane is successfully fabricated using electrospinning and solution casting. The composite membrane comprises two microporous poly(vinylidene fluoride-hexafluoropropylene) (PVDF-HFP) layers plus an intermediate quaternary ammonium-containing SiO2 nanoparticles modified polyethylene terephthalate (PET) nanofibrous nonwoven to form a sandwiched PVDF-HFP/PET/PVDF-HFP composite, which is employed as a separator for lithium-ion batteries (LIBs). The properties of the PET composite membrane are compared with those of commercial PE separator, such as the morphologies, physical properties, and electrochemical performances. According to our results, the composite membrane demonstrates superior thermal stability (thermal shrinkage ~8%), electrolyte-philicity (contact angle ~2.9°), electrolyte uptake and retention (282%, 74%), and ionic conductivity (~10?3 S cm?1). The separators are assembled into Li/LiFePO4 cells for electrochemical tests, showing that the PET composite membrane cells exhibit higher capacities than those with the PE separator at 0.2–10C both at 25 °C and 55 °C. The discharge capacity retention and coulombic efficiency of the PET composite membrane cells at 1C/1C for 200 cycles can be respectively enhanced about 20% and 2% at 55 °C as compared to the PE separator cells. These results demonstrate that our prepared PET composite membrane is highly promising for LIB applications.  相似文献

A gel-based proteomics approach was used to screen for proteins of differential abundance between the saliva of smokers and those who had never smoked. Subjecting precipitated proteins from whole human saliva of healthy non-smokers to two-dimensional electrophoresis (2-DE) generated typical profiles comprising more than 50 proteins. While 35 of the proteins were previously established by other researchers, an additional 22 proteins were detected in the 2-DE saliva protein profiles generated in the present study. When the 2-DE profiles were compared to those obtained from subjects considered to be heavy cigarette smokers, three saliva proteins, including interleukin-1 receptor antagonist, thioredoxin and lipocalin-1, showed significant enhanced expression. The distribution patterns of lipocalin-1 isoforms were also different between cigarette smokers and non-smokers. The three saliva proteins have good potential to be used as biomarkers for the adverse effects of smoking and the risk for inflammatory and chronic diseases that are associated with it.  相似文献
