Tissue dynamics spectroscopy combines dynamic light scattering with short-coherence digital holography to capture intracellular motion inside multicellular tumor spheroid tissue models. The cellular mechanical activity becomes an endogenous imaging contrast agent for motility contrast imaging. Fluctuation spectroscopy is performed on dynamic speckle from the proliferating shell and hypoxic core to generate drug-response spectrograms that are frequency versus time representations of the changes in spectral content induced by an applied compound or an environmental perturbation. A combination of 28 reference compounds and conditions applied to rat osteogenic UMR-106 spheroids generated spectrograms that were crosscorrelated in a similarity matrix used for unsupervised hierarchical clustering of similar compound responses. This work establishes the feasibility of tissue dynamics spectroscopy for three-dimensional tissue-based phenotypic profiling of drug response as a fully endogenous probe of the response of tissue to reference compounds.  相似文献

This paper presents a quantitative approach to multimodal discourse analysis for analyzing online collaborative learning. The coding framework draws together the fields of systemic functional linguistics and Activity Theory to analyze interactions between collaborative-, content- and technology-related discourse. The approach is used to examine how the task subject matter, the activity design, and the choice of interface affected interaction and collaboration for a computing course conducted in a web-conferencing environment. The analysis revealed the critical impact of activity design on the amount and type of discourse that transpired. Student-centred designs resulted in over six times more student discourse as compared to teacher-centred designs and created a learning environment where students took greater ownership over the tasks and contributed more to the content-based discussion. The paper also incorporates a rationale for the approach to coding and a reflection on its efficacy for discourse analysis in technology-based learning environments.  相似文献

Instructors in higher education are disseminating instructional content via podcasting, as many rally behind the technology’s potential benefits. Others have expressed concern about the risks of deleterious effects that might accompany the adoption of podcasting, such as lower class attendance. Yet, relatively few studies have investigated students’ perceptions of podcasting for educational purposes, especially in relation to different podcasting forms: repetitive and supplemental. The present study explored students’ readiness and attitudes towards these two forms of podcasting to provide fundamental information for future researchers and educators. The results indicated that students may not be as ready or eager to use podcasting for repetitive or supplemental educational purposes as much as we think they are, but they could be persuaded.  相似文献

The software project management body of knowledge is gradually being renewed across the entire lifecycle. In the conception phase, the focus is on fostering innovation through new approaches such as business analysis and crowdsourcing techniques. In the construction phase, the rise of global software development has shrunk the world and led to new approaches to risk management that take into account factors such as cultural diversity. In the project conclusion phase, new approaches to the evaluation of project success are being introduced, such as project retrospectives and intellectual capital reporting. Conversely, long accepted approaches to measurement and evaluation such as those promoted in the Chaos reports of the Standish Group are being called into question.  相似文献

This paper introduces two new one-dimensional cutting stock models: the generalized assortment problem (GAP) and the best cutting stock length (BSL) problem. These new models provide the potential to reduce waste to values lower than the optimum of current models, under the right management circumstances. In the GAP, management has a standard length and can select one or more of any additional custom stock lengths, and management wishes to minimize cutting stock waste. This model is different from existing models that assume that the selection is from a small fixed set of stock lengths. In the BSL problem, management chooses any number of custom stock lengths, but wishes to find the fewest custom stock lengths in order to have zero waste. Results show waste reductions of 80% with just one custom stock length compared with solutions from standard cutting stock formulations, when item lengths are long relative to the stock length. The models are most effective when the item lengths are nearly as long as the stock length. Solutions from the model have been implemented for a manufacturer. The model is easily generalized to allow multiple existing stock lengths and different costs.  相似文献

In a number of prior papers we described and explored a new pattern recognition method called Margin-Setting that accomplishes excellent generalization using very few training samples. The result was a multi-round classifier with each round consisting of a set of hyperspheres such that if a datum fell within a certain hypersphere, it was labeled with one particular class. Margin-Setting achieves concurrent low Vapnik-Chervonenkis (VC) dimension and high accuracy which is a consequence of partitioning the training set into smaller sets that make this possible. This paper extends Margin-Setting from hyperspheres to hyperellipsoids resulting in improved performance.  相似文献

Genetic programming has now been used to produce at least 76 instances of results that are competitive with human-produced results. These human-competitive results come from a wide variety of fields, including quantum computing circuits, analog electrical circuits, antennas, mechanical systems, controllers, game playing, finite algebras, photonic systems, image recognition, optical lens systems, mathematical algorithms, cellular automata rules, bioinformatics, sorting networks, robotics, assembly code generation, software repair, scheduling, communication protocols, symbolic regression, reverse engineering, and empirical model discovery. This paper observes that, despite considerable variation in the techniques employed by the various researchers and research groups that produced these human-competitive results, many of the results share several common features. Many of the results were achieved by using a developmental process and by using native representations regularly used by engineers in the fields involved. The best individual in the initial generation of the run of genetic programming often contains only a small number of operative parts. Most of the results that duplicated the functionality of previously issued patents were novel solutions, not infringing solutions. In addition, the production of human-competitive results, as well as the increased intricacy of the results, are broadly correlated to increased availability of computing power tracked by Moore’s law. The paper ends by predicting that the increased availability of computing power (through both parallel computing and Moore’s law) should result in the production, in the future, of an increasing flow of human-competitive results, as well as more intricate and impressive results.  相似文献
