Glioblastoma (GBM) is the most common tumor in the central nervous system in adults. This neoplasia shows a high capacity of growth and spreading to the surrounding brain tissue, hindering its complete surgical resection. Therefore, the finding of new antitumor therapies for GBM treatment is a priority. We have previously described that cyclin D1-CDK4 promotes GBM dissemination through the activation of the small GTPases RalA and RalB. In this paper, we show that RalB GTPase is upregulated in primary GBM cells. We found that the downregulation of Ral GTPases, mainly RalB, prevents the proliferation of primary GBM cells and triggers a senescence-like response. Moreover, downregulation of RalA and RalB reduces the viability of GBM cells growing as tumorspheres, suggesting a possible role of these GTPases in the survival of GBM stem cells. By using mouse subcutaneous xenografts, we have corroborated the role of RalB in GBM growth in vivo. Finally, we have observed that the knockdown of RalB also inhibits cell growth in temozolomide-resistant GBM cells. Overall, our work shows that GBM cells are especially sensitive to Ral-GTPase availability. Therefore, we propose that the inactivation of Ral-GTPases may be a reliable therapeutic approach to prevent GBM progression and recurrence.  相似文献

Soil and water conservation ecological compensation can be simply defined as a monetary payment to offset the environmental loss. An illustration is given in this study describing the payment compensation to water resource conservation and soil losses in Erlongshan reservoir catchment, China. A semi-distributed hydrological SWAT model was applied to establish compensation standard considering six scenarios of land use changes by combined application of remote sensing and geographic information systems. Cost-benefit analysis (CBA) method is applied to evaluate the function of soil and water conservation, of which marginal opportunity cost and market value methods have been explored calculate the cost and benefit of water and soil conservation ecological function from provider and beneficiaries. Finally the ecological compensation of soil and water conservation for different land-use scenarios is calculated incorporating benefit apportion coefficient. The results provide an economically evaluated and market-oriented standard for the study of eco-compensation of environmental services and will be of great benefit to the implementation of soil and water conservation at a mesoscale catchment scale.  相似文献

A wide variety of privacy metrics have been proposed in the literature to evaluate the level of protection offered by privacy-enhancing technologies. Most of these metrics are specific to concrete systems and adversarial models and are difficult to generalize or translate to other contexts. Furthermore, a better understanding of the relationships between the different privacy metrics is needed to enable more grounded and systematic approach to measuring privacy, as well as to assist system designers in selecting the most appropriate metric for a given application. In this work, we propose a theoretical framework for privacy-preserving systems, endowed with a general definition of privacy in terms of the estimation error incurred by an attacker who aims to disclose the private information that the system is designed to conceal. We show that our framework permits interpreting and comparing a number of well-known metrics under a common perspective. The arguments behind these interpretations are based on fundamental results related to the theories of information, probability, and Bayes decision.  相似文献

Seventy samples of rice purchased from local markets in six cities from Morocco (Rabat, Casablanca, Kénitra, Mohammadia, Tanger and Errachidia) were analyzed for the presence of six emerging mycotoxins: four enniatins ENs (ENA, ENA1, ENB and ENB1), beauvericin (BEA) and fusaproliferin (FUS). Samples were extracted with a mixture of acetonitrile/water (85/15, v/v) by using an ultra-turrax homogenizer. Mycotoxins were then identified and quantified with liquid chromatography (LC) coupled to diode array detector (DAD). Positive samples were confirmed with an LC-MS/MS. Analytical results showed that BEA was present in 75.7% of total analyzed samples. BEA levels varied between 3.8 and 26.3 mg/kg. The frequencies of contamination of samples with total ENs and FUS were 50% and 4.3%, respectively. Among the ENs, ENB was the mycotoxin much more found (30% of total samples), while ENB1, ENA and ENA1 were found in 24.6%, 22.8% and 5.7% of total samples, respectively. The high ENs value was registered in a rice sample from kénitra (448.7 mg/kg of ENA1). This is the first study that describes the presence of emerging Fusarium mycotoxins in rice available in Morocco.  相似文献

Traditional kernel classifiers assume independence among the classification outputs. As a consequence, each misclassification receives the same weight in the loss function. Moreover, the kernel function only takes into account the similarity between input values and ignores possible relationships between the classes to be predicted. These assumptions are not consistent for most of real-life problems. In the particular case of remote sensing data, this is not a good assumption either. Segmentation of images acquired by airborne or satellite sensors is a very active field of research in which one tries to classify a pixel into a predefined set of classes of interest (e.g. water, grass, trees, etc.). In this situation, the classes share strong relationships, e.g. a tree is naturally (and spectrally) more similar to grass than to water. In this paper, we propose a first approach to remote sensing image classification using structured output learning. In our approach, the output space structure is encoded using a hierarchical tree, and these relations are added to the model in both the kernel and the loss function. The methodology gives rise to a set of new tools for structured classification, and generalizes the traditional non-structured classification methods. Comparison to standard SVM is done numerically, statistically and by visual inspection of the obtained classification maps. Good results are obtained in the challenging case of a multispectral image of very high spatial resolution acquired with QuickBird over a urban area.  相似文献

In recent years, some beneficial effects on health of melanins that are formed as a result of polyphenol oxidase action have been discovered. However, it is also known that similar polymers have a detrimental effect on proteases activity. Thus, the aim of this work was to assay whether melanins from two different substrates act as carboxypeptidase A and carboxypeptidase B inhibitors. It was found that melanin synthesized from l-tyrosine decreases both the apparent affinity between carboxypeptidases and their substrates and the maximum reaction rate. Consequently, it is possible to conclude that these melanins inhibit CPA and CPB activity in a mixed way. On the contrary, no inhibition was found in the case of melanin synthesized from 4-methylcatechol.  相似文献

A recent computational methodology is applied here in order to analyze the impact of free surfaces on the measurement of the interdiffusion coefficients if shear cells are used. Calculations indicate that the above-mentioned impact depends on the intensity of the solutal Marangoni convection but mainly on the number of active Marangoni segments, that is to say, on the extent of the free surface considered. Bulk solutal convection due to reduced gravity levels is not relevant if the gravity vector acts orthogonally to the gradient of concentration.  相似文献

The preparation, characterization, and controlled release of hydroxyapatite (HAp) nanoparticles loaded with streptomycin (STR) was studied. These nanoparticles are highly appropriate for the treatment of bacterial infections and are also promising for the treatment of cancer cells. The analyses involved scanning electron microscopy, dynamic light scattering (DLS) and Z-potential measurements, as well as infrared spectroscopy and X-ray diffraction. Both amorphous (ACP) and crystalline (cHAp) hydroxyapatite nanoparticles were considered since they differ in their release behavior (faster and slower for amorphous and crystalline particles, respectively). The encapsulated nanoparticles were finally incorporated into biodegradable and biocompatible polylactide (PLA) scaffolds. The STR load was carried out following different pathways during the synthesis/precipitation of the nanoparticles (i.e., nucleation steps) and also by simple adsorption once the nanoparticles were formed. The loaded nanoparticles were biocompatible according to the study of the cytotoxicity of extracts using different cell lines. FTIR microspectroscopy was also employed to evaluate the cytotoxic effect on cancer cell lines of nanoparticles internalized by endocytosis. The results were promising when amorphous nanoparticles were employed. The nanoparticles loaded with STR increased their size and changed their superficial negative charge to positive. The nanoparticles’ crystallinity decreased, with the consequence that their crystal sizes reduced, when STR was incorporated into their structure. STR maintained its antibacterial activity, although it was reduced during the adsorption into the nanoparticles formed. The STR release was faster from the amorphous ACP nanoparticles and slower from the crystalline cHAp nanoparticles. However, in both cases, the STR release was slower when incorporated in calcium and phosphate during the synthesis. The biocompatibility of these nanoparticles was assayed by two approximations. When extracts from the nanoparticles were evaluated in cultures of cell lines, no cytotoxic damage was observed at concentrations of less than 10 mg/mL. This demonstrated their biocompatibility. Another experiment using FTIR microspectroscopy evaluated the cytotoxic effect of nanoparticles internalized by endocytosis in cancer cells. The results demonstrated slight damage to the biomacromolecules when the cells were treated with ACP nanoparticles. Both ACP and cHAp nanoparticles were efficiently encapsulated in PLA electrospun matrices, providing functionality and bioactive properties.  相似文献

Morcilla de Burgos is the most famous blood sausage in Spain. However, while producers are interested in extending its shelf life, the consumer is increasingly demanding more natural food. This situation has led to the current search for new and mild preservation technologies. Two batches of four different products: control without any treatment, control with organic acid salts (CnOAS; a 3% mixture of potassium/sodium l-lactate), control with high hydrostatic pressure processing (CnHPP; 600MPa-10min), and a combination of both treatments (OAS+HPP), were carried out to evaluate any synergistic effect that occurs when combining OAS and HPP, and the influence of different preservative treatments on the spoilage bacterial population and their evolution. HPP (with or without addition of OAS) can be considered the most suitable method for preserving morcilla de Burgos as it does not produce negative changes in sensory attributes. No clear selective effect of different treatments on the composition of the spoilage bacteria was seen and similar spoilage patterns were observed independently of the preservation treatment used.  相似文献
