This study describes the extraction and characterisation of cashew apple polyphenol oxidase (PPO) and the effect of wounding on cashew apple phenolic acid composition, PPO activity and fruit browning. Purification factor was 59 at 95% (NH4)2SO4 saturation. For PPO activity, the optimal substrate was catechol and the optimum pH was 6.5. PPO Km and Vmax values were 18.8 mM and 13.6 U min−1 ml−1, respectively. Ascorbic acid, citric acid, sodium sulphite and sodium metabisulphite decreased PPO activity, while sodium chloride increased PPO activity. Wounding at 2 °C and 27 °C for 24 h increased PPO activity but storage at 40 °C reduced PPO activity. Gallic acid, protocatechuic acid and cinnamic acid (free and conjugate) were identified in cashew apple juice. Cutting and subsequent storage at 40 °C hydrolysed cinnamic acid. 5-Hydroxymethylfurfural content in cashew apple juice increased after injury and storage at higher temperatures, indicating non-enzymatic browning.  相似文献

Through honey's fermentation, diverse beverages can be obtained, among which is mead, an alcoholic drink with 8 to 18% of ethanol (v/v). Since honey is a matrix with a low nutrient concentration and other unfavourable growth conditions, several problems are usually encountered, namely delayed or arrested fermentations, unsatisfactory quality parameters and lack of uniformity of the final product, as well as unpleasant sensory properties. In this context, the aim of this work was to optimize mead production through honey‐must supplementation with (a) salts, (b) vitamins or (c) salts + vitamins. The effects of the honey‐must formulation on the fermentation kinetics, growth profile and physicochemical characteristics of final meads were evaluated. The results showed minor differences in the fermentation profile and time between fermentations with the different formulations. The growth profile was influenced more by the yeast strain than by the supplements added to the honey‐must. In general, the honey‐must composition did not influence meads’ final characteristics, except regarding the SO2 concentration of the meads produced using the strain QA23. In summary, the addition of salts and/or vitamins to honey‐must had no positive effects on the fermentation, growth profile or characteristics of the final products. Mead is an alcoholic beverage, produced since ancient times, resulting from an alcoholic fermentation of diluted honey by yeasts. When it is produced in a traditional manner, mead producers can encounter several problems related to a lack of essential nutrients, such as available nitrogen. Thus, the aim of this study was to evaluate the effect of nitrogen addition to honey‐must on the fermentation performance of two Saccharomyces cerevisiae wine yeasts, QA23 and ICV D47, as well as on the mead composition and production of volatile aroma compounds. A portion of honey‐must was supplemented with diammonium phosphate (DAP) to achieve the nitrogen concentration required by yeast to complete alcoholic fermentation. The supplementation with DAP reduced the fermentation length to around 7 days, but not all sugars were fully consumed, suggesting that other factors could be interfering with yeast growth. For both yeasts the specific growth rate and final biomass were higher in musts supplemented with DAP. Mead final composition was similar under the two experimental conditions. Analysis of the volatile profile revealed that the concentrations of the volatile fatty acids and volatile phenols were higher in meads supplemented with DAP. The concentrations of ethyl hexanoate, ethyl octanoate and isoamyl acetate were above their perception threshold and were higher in meads supplemented with DAP, which could contribute to the enhancement of the fruity character. Poly(vinylidene fluoride)/Pb(Zr0.53Ti0.47)O3([PVDF]1 ? x/[PZT]x ) composites of volume fractions x and (0?C3) type connectivity were prepared in the form of thin films. The films were prepared by solvent casting with PZT powder of 0.84, 1.86, and 2.35???m average size with filler contents up to 40?% volume. The crystalline phase of the polymer matrix was the nonpolar ??-phase and the polar ??-phase. Dielectric measurements were performed in order to evaluate the influence of the filler size and content as well as the effect of the polymer matrix in the overall response of the material. No nucleation effect of any of the phases was observed for the used fillers. The spherulitic structure of the pure ??-PVDF and the characteristic porosity of the ??-phase material are destroyed for high PZT volume fractions. The inclusion of ceramic particles in the PVDF polymer matrix increases the complex dielectric constant of the composites independently of the PVDF polymer matrix. The dielectric properties of the composites are mainly affected by the amount of the ceramic particles. With respect to the relaxation processes of the polymer, the activation energy of the ??a-relaxation increases and the glass transition temperature decreases with increasing particle size and content. The high-temperature conductivity decreases with increasing filler content and there is an important contribution of the Maxwell-Wagner-Sillars effect to the overall dielectric response.  相似文献

In an attempt to enrich the world of dentistry through the development of new materials, this study proposes to synthesize and incorporate a monomer containing the N-hydroxysuccinimide (NHS) ester reactive group to a dental adhesive. As such, this study developed a simple method to obtain NHS esters by employing acrylic acid (AA) and NHS in the presence of EDC (N-ethyl-N′-(3-dimethylaminopropyl)carbodiimide), from an aprotic medium. The experimental N-acryloxysuccinimide (NSA) monomer was analyzed by infrared spectroscopy (FTIR) and nuclear magnetic resonance of hydrogen (1H NMR). This monomer was then incorporated within a prepared dental adhesive, in a 5% proportion. The modified adhesive, containing NSA, was applied to the dental surface of bovine teeth to obtain an adhesive/dentin interface in vitro. The characterization of this interface by Raman spectroscopy presented the formation of new amide bonds. Moreover, through scanning electron microscopy (SEM), it became possible to observe an intense penetration of this modified adhesive on the dental surface. Considering the outcome, it can be concluded that the synthesized NSA monomer provided a favorable condition for the dental adhesives to interact chemically with the dentin collagen fibers.  相似文献

Quadruplex nucleic acids are promising targets for cancer therapy. In this study we used a fragment-based approach to create new flexible G-quadruplex (G4) DNA-interactive small molecules with good calculated oral drug-like properties, based on quinoline and triazole heterocycles. G4 melting temperature and polymerase chain reaction (PCR)-stop assays showed that two of these compounds are selective G4 ligands, as they were able to induce and stabilize G4s in a dose- and DNA sequence-dependent manner. Molecular docking studies have suggested plausible quadruplex binding to both the G-quartet and groove, with the quinoline module playing the major role. Compounds were screened for cytotoxicity against four cancer cell lines, where 4,4′-(4,4′-(1,3-phenylene)bis(1H-1,2,3-triazole-4,1-diyl))bis(1-methylquinolin-1-ium) ( 1 d ) showed the greater activity. Importantly, dose–response curves show that 1 d is cytotoxic in the human colon cancer HT-29 cell line enriched in cancer stem-like cells, a subpopulation of cells implicated in chemoresistance. Overall, this study identified a new small molecule as a promising lead for the development of drugs targeting G4 in cancer stem cells.  相似文献

Many scale-up criteria for supercritical fluid extraction (SFE) can be found in literature. However, the studies are often divergent and inconclusive; therefore, more studies on this field are needed. The objective of the present work was to study the scale-up of SFE process focusing application to Brazilian raw materials. A laboratory scale equipment (290 mL extraction vessel) and a pilot scale equipment (5.15 L extraction vessel) were used to study scale-up of SFE for clove and sugarcane residue. The scale-up criterion adopted consisted in maintaining solvent mass to feed mass ratio constant. The criterion was successfully used for a 15-fold scale-up of overall extraction curves for both raw materials studied; yields in pilot scale were slightly higher than in laboratory scale. The criterion studied allows a rapid and simple scale-up procedure, which can be very useful for the purpose of developing SFE technology at industrial scale in developing countries where such technology is still not available at industrial level.  相似文献

The reaction of methanol steam reforming was studied in a carbon membrane reactor over a commercial CuO/ZnO/Al2O3 catalyst (Süd-Chemie, G66 MR). Carbon molecular sieve membranes supplied by Carbon Membranes Ltd. were tested at 150 °C and 200 °C. The carbon membrane reactor was operated at atmospheric pressure and with vacuum at the permeate side, at 200 °C. High methanol conversion and hydrogen recovery were obtained with low carbon monoxide permeate concentrations. A sweep gas configuration was simulated with a one-dimensional model. The experimental mixed-gas permeance values at 200 °C were used in a mathematical model that showed a good agreement with the experimental data. The advantages of using water as sweep gas were investigated in what concerns methanol conversion and hydrogen recovery. The concentration of carbon monoxide at the permeate side was under 20 ppm in all simulation runs. These results indicate that the permeate stream can be used to feed a polymer electrolyte membrane fuel cell.  相似文献

Acrylic‐based nanocomposite resins have been investigated in view of future application in water borne automotive coatings by the aid of postemulsification processes. Mechanical, flow and leveling properties of the nanocomposite resins, containing various concentrations of silicate, have been investigated. The results are related to morphological information obtained from TEM and WAXS measurements in the liquid suspension as well as from the cured film. At low silicate loadings, when flow properties are still acceptable for typical emulsification processing, a strong increase in modulus of the cured coating films is observed because of the mainly exfoliated silicate platelets. The rate of increase in modulus of the cured films decreases at higher silicate loadings. Analyzing the mechanical data, using Halpin–Tsai theory indicates that this is due to less perfect exfoliation at higher silicate loading, which is confirmed by TEM analysis. In addition at higher silicate loading, the flow properties of the resin as analyzed by DMA show solid‐like behavior.
