The fabrication of one-dimensional (1D) nanostructures and microstructures inside the pores of porous templates is intensively investigated. The release of these structures is commonly accomplished by etching and destroying the templates. The 1D nanostructures and microstructures tend to condense because of the occurrence of capillary forces during drying of the specimens. It is shown that highly ordered arrays of polymer microfibers can be easily detached from silanized porous templates by mechanical lift-off. This procedure leaves the templates intact, thus allowing their recycling, and does not involve the use of solutions or solvents, thus circumventing condensation. Therefore, mechanical lift-off may enable the up-scaling of template-based approaches to the fabrication of highly ordered assemblies of 1D nanostructures and microstructures. 相似文献

From an airship dockyard to a tropical rainforest ndash; fire protection concept for the the largest hall construction of the world. Tropical Islands is a project with enormous dimensions and very specific utilizations. It is used as a dockyard for large cargo airships as well as for recreational and event presentation purposes with a tropical rainforest in it. This article does not show the complete fire protection concept but has represented certain aspects and focal points that transcend the project and could be of general interest. The results of smoke tests and their specific implementation concerning the particular conditions of the dockyard were shown to point out the very special question of smoke exhaust venting. For the intended new usage of the building the effectiveness of the measures was tested systematically under various conditions and trough successive mathematical approximation. In this case it was pointed out that it was more important to see the superior context than to simulate single scenarios. This cognition may also be useful for projects which are less spectacular. This project is an ideal example to show how largely fire protection measures must and can be adapted to the specific usage of the building. This aspect becomes more important especially in times where buildings were used in other ways as they were intended originally because of economical decisions or simply to conserve resources. 相似文献

The use of multiblock copolymers for the compatibilization of immiscible polymer blends is controversially discussed in the literature. Investigations have been carried out to estimate the effect of multiblock copolymers containing segments of a liquid crystalline polyester (LCP) and polysulfone (PSU) segments in blends of the based homopolymers. One goal was to determine whether multiblock copolymers provide an opportunity for compatibilizing PSU/LCP blends. By using PSU/LCP multiblock copolymers with different molecular weights of the blocks in the appropriate binary, solution-casted blends, it was shown that the interpenetration of the polysulfone phase of the block copolymer and the PSU matrix leads to an improved miscibility of the blend. This effect is retained in ternary blends of PSU, LCP, and the multiblock copolymer, assuming a certain critical molecular weight of the multiblock copolymer segments. In addition, some mechanical characteristics of PSU/LCP melt blends such as the E-modulus and fracture strength are improved by adding long-segmented multiblock copolymers. © 1997 John Wiley & Sons, Inc. Subarachnoid hemorrhage (SAH) is associated with acute and delayed cerebral ischemia. We suggested spasms of pial arterioles as a possible mechanism; however, it remained unclear whether and how pial microvasospasms (MVSs) induce cerebral ischemia. Therefore, we used in vivo deep tissue imaging by two-photon microscopy to investigate MVSs together with the intraparenchymal microcirculation in a clinically relevant murine SAH model. Male C57BL/6 mice received a cranial window. Cerebral vessels and leukocytes were labelled with fluorescent dyes and imaged by in vivo two-photon microscopy before and three hours after SAH induced by filament perforation. After SAH, a large clot formed around the perforation site at the skull base, and blood distributed along the perivascular space of the middle cerebral artery up to the cerebral cortex. Comparing the cerebral microvasculature before and after SAH, we identified three different patterns of constrictions: pearl string, global, and bottleneck. At the same time, the volume of perfused intraparenchymal vessels and blood flow velocity in individual arterioles were significantly reduced by more than 60%. Plugging of capillaries by leukocytes was observed but infrequent. The current study demonstrates that perivascular blood is associated with spasms of pial arterioles and that these spasms result in a significant reduction in cortical perfusion after SAH. Thus, the pial microvasospasm seems to be an important mechanism by which blood in the subarachnoid space triggers cerebral ischemia after SAH. Identifying the mechanisms of pial vasospasm may therefore result in novel therapeutic options for SAH patients. 相似文献

We propose a multimedia model-based methodology to evaluate whether a chemical substance qualifies as POP-like based on overall persistence (Pov) and potential for long-range transport (LRTP). It relies upon screening chemicals against the Pov and LRTP characteristics of selected reference chemicals with well-established environmental fates. Results indicate that chemicals of high and low concern in terms of persistence and long-range transport can be consistently identified by eight contemporary multimedia models using the proposed methodology. Model results for three hypothetical chemicals illustrate that the model-based classification of chemicals according to Pov and LRTP is not always consistent with the single-media half-life approach proposed by the UNEP Stockholm Convention and thatthe models provide additional insight into the likely long-term hazards associated with chemicals in the environment. We suggest this model-based classification method be adopted as a complement to screening against defined half-life criteria at the initial stages of tiered assessments designed to identify POP-like chemicals and to prioritize further environmental fate studies for new and existing chemicals. 相似文献

A triple mutant of an esterase from Pseudomonas fluorescens (PFE) that was created by directed evolution exhibited high enantioselectivity (E=89) in a kinetic resolution and yielded the building block (S)-but-3-yn-2-ol. Surprisingly, a mutation close to the active site caused the formation of inclusion bodies, but remote mutations were found to be responsible for the high selectivity. Back mutations gave a variant (double mutant PFE Ile76Val/Val175Ala) that showed excellent selectivity (E=96) and activity (20 min for 50% conversion, which corresponds to 1.25 U per mg of protein). 相似文献

The objective of this study was to determine the prevalence and numbers of Campylobacter on the skin and in the muscle of chicken legs at retail to examine the external and internal contamination for an exposure assessment. Furthermore, the study assessed seasonal influence on Campylobacter contamination in chicken legs. Of the 140 examined skin samples, 66% were positive, and the internal contamination of 115 sampled chicken legs was 27%. The enumeration of Campylobacter on the surface of positive chicken legs revealed a median of 2.4 log CFU/g of skin, and the quantification of Campylobacter in the muscle gave results mainly under the detection limit of the most-probable-number method (<0.3 MPN Campylobacter per g). The external contamination was significantly higher than the internal. In both skin and muscle samples, Campylobacter jejuni had a much higher incidence than Campylobacter coli. However, with regard to the specification of Campylobacter on the surface of chicken legs, C. coli was isolated at higher colony counts than C. jejuni. During the 1-year study, two peaks of Campylobacter contamination occurred, one in the early springtime (February and March, 100 and 90%, respectively) and the second during the warmer months in the summer (July and August, both 90%). Carbohydrate-based sensors, that specifically detect sugar binding molecules or cells, are increasingly important in medical diagnostic and drug screening. Here we demonstrate that cantilever arrays functionalized with different mannosides allow the real-time detection of several Escherichia coli strains in solution. Cantilever deflection is thereby dependent on the bacterial strain studied and the glycan used as the sensing molecule. The cantilevers exhibit specific and reproducible deflection with a sensitivity range over four orders of magnitude. 相似文献

A combined experimental and theoretical study is presented revealing the influence of metal–molecule coupling on electronic transport through single‐molecule junctions. Transport experiments through tolane molecules attached to gold electrodes via thiol, nitro, and cyano anchoring groups are performed. By fitting the experimental current–voltage characteristics to a single‐level tunneling model, we extract both the position of the molecular orbital closest to the Fermi energy and the strength of the metal–molecule coupling. The values found for these parameters are rationalized with the help of density‐functional‐theory‐based transport calculations. In particular, these calculations show that the anchoring groups determine the junction conductance by controlling not only the strength of the coupling to the metal but also the position of the relevant molecular energy levels. 相似文献
