The limited options of suitable elastomers with adequate cost-performance balance drive the necessity to introduce new materials in the oil & gas (O&G) application space. The relevance of a recently developed copolymer of chloroprene and acrylonitrile (referred to as acrylonitrile–chloroprene rubber or NCR) to O&G applications is described in this technical contribution. The new elastomer demonstrates adequate physical properties and reasonably good high and low-temperature capabilities. It offers good resistance to several aqueous and non-aqueous fluids with low volume swelling and retains its physical properties to reasonable extents while exposed to hot test oils. Acrylonitrile–chloroprene rubber has been found to sustain “sour gas” exposure. Good abrasion resistance, high tear strength, and remarkably high flex-fatigue resistance coupled with low heat build-up reflect its durability under dynamic conditions. In addition, acrylonitrile–chloroprene rubber can very well withstand the rapid gas decompression test at 25°C. This unique combination of attributes may allow acrylonitrile–chloroprene rubber to be considered as a candidate material for high-performance O&G applications.  相似文献

New biomarkers are needed to further stratify the risk of malignancy in intraductal papillary mucinous neoplasm (IPMN). Although microRNAs (miRNAs) are expected to be stable biomarkers, they can vary owing to a lack of definite internal controls. To identify universal biomarkers for invasive IPMN, we performed miRNA sequencing using tumor-normal paired samples. A total of 19 resected tissues and 13 pancreatic juice samples from 32 IPMN patients were analyzed for miRNA expression by next-generation sequencing with a two-step normalization of miRNA sequence data. The miRNAs involved in IPMN associated with invasive carcinoma were identified from this tissue analysis and further verified with the pancreatic juice samples. From the tumor-normal paired tissue analysis of the expression levels of 2792 miRNAs, 20 upregulated and 17 downregulated miRNAs were identified. In IPMN associated with invasive carcinoma (INV), miR-10a-5p and miR-221-3p were upregulated and miR-148a-3p was downregulated when compared with noninvasive IPMN. When these findings were further validated with pancreatic juice samples, miR-10a-5p was found to be elevated in INV (p = 0.002). Therefore, three differentially expressed miRNAs were identified in tissues with INV, and the expression of miR-10a-5p was also elevated in pancreatic juice samples with INV. MiR-10a-5p is a promising additional biomarker for invasive IPMN.  相似文献

Three beta-glucosidase- and two endoglucanase-encoding genes were cloned from Aspergillus oryzae, and their gene products were displayed on the cell surface of the sake yeast, Saccharomyces cerevisiae GRI-117-UK. GRI-117-UK/pUDB7 displaying beta-glucosidase AO090009000356 showed the highest activity against various substrates and efficiently produced ethanol from cellobiose. On the other hand, GRI-117-UK/pUDCB displaying endoglucanase AO090010000314 efficiently degraded barley beta-glucan to glucose and smaller cellooligosaccharides. GRI-117-UK/pUDB7CB codisplaying both beta-glucosidase AO090009000356 and endoglucanase AO090010000314 was constructed. When direct ethanol fermentation from 20 g/l barley beta-glucan as a model substrate was performed with the codisplaying strain, the ethanol concentration reached 7.94 g/l after 24 h of fermentation. The conversion ratio of ethanol from beta-glucan was 69.6% of the theoretical ethanol concentration produced from 20 g/l barley beta-glucan. These results showed that sake yeast displaying A. oryzae cellulolytic enzymes can be used to produce ethanol from cellulosic materials. Our constructs have higher ethanol production potential than the laboratory constructs previously reported.  相似文献

Improved agar diffusion method for detecting residual antimicrobial agents   总被引:4,自引:0,他引:4  
The improved agar diffusion method for determination of residual antimicrobial agents was investigated, and the sensitivities of various combinations of test organisms and assay media were determined using 7 organisms, 5 media, and 31 antimicrobial agents. Bacillus stearothermophilus and synthetic assay medium (SAM) showed the greatest sensitivity for screening penicillins (penicillin G and ampicillin). The combination of Bacillus subtilis and minimum medium (MM) was the most sensitive for tetracyclines (oxytetracycline and chlortetracycline), B. stearothermophilus and SAM or Micrococcus luteus and Mueller-Hinton agar (MHA) for detecting tylosin and erythromycin, B. subtilis and MHA for aminoglycosides (streptomycin, kanamycin, gentamicin, and dihydrostreptomycin), B. stearothermophilus and SAM for polyethers (salinomycin and lasalocid), and B. subtilis and MM or Clostridium perfringens and GAM for polypeptides (thiopeptin, enramycin, virginiamycin, and bacitracin). However, gram-negative bacterium Escherichia coli ATCC 27166 and MM were better for screening for colistin and polymixin-B. For detecting the synthetic drugs tested, the best combination was B. subtilis and MM for sulfonamides, E. coli 27166 and MM for quinolones (oxolinic acid and nalidixic acid), B. subtilis and MM for furans (furazolidone), and the bioluminescent bacterium Photobacterium phosphoreum and luminescence assay medium for chloramphenicol and oxolinic acid. The results showed that the use of four assay plates, B. stearothermophilus and SAM, B. subtilis and MM, M. luteus and MHA, and E. coli 27166 and MM, was superior to the currently available techniques for screening for residual antimicrobial agents in edible animal tissues.  相似文献

Making use of the fine resolution of satellite SAR imagery, we observe small eddies during the spring and summer months in several locations in Lake Superior. During these months there is a thermal gradient between warmer nearshore waters and colder offshore waters which enhances cyclonic coastal currents. Using spaceborne SAR imagery from the European Space Agency's ERS-1 and ERS-2 missions from 1992 to 1998, we observe small eddies, identifying and mapping basic eddy characteristics including diameter, location, and rotational sense. In total, 45 eddies were located, of which 41 were cyclonic and 4 anticyclonic. Average diameter was 9.8 km and average distance to shore was 8.1 km. Based on sea surface temperature data from AVHRR, the eddies are located within the region of sharp thermal gradients of order 3–5 °C per 3 km. Spatial and temporal coverage was uneven, however, more eddies were seen in SAR images taken in late summer along the southern and eastern shores as well as areas where the boundary current interacts with topographic features including islands and promontories.  相似文献

There are two predominant palm tree species producing edible fruit known as “açaí” found widely dispersed through the Amazon: Euterpe oleracea Mart. and Euterpe precatoria Mart. They differ from each other in terms of how the plants grow and their phytochemical composition. E. oleracea (EO) has received considerable attention as a “super fruit” because of its high antioxidant capacity, while studies on E. precatoria (EP) remain rare. In this study, the antioxidant and anti-inflammatory activities of EP fruit pulps were evaluated by different assays including a series of oxygen radical absorbance capacity (ORAC) based assays, the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay, the cell-based antioxidant protection in erythrocyte (CAP-e) assay, as well as the nuclear factor-kappa B (NF-κB) secreted embryonic alkaline phosphatase (SEAP) assay. Total phenolics were also measured as an indication of the total phenol content. For comparative purposes, the EO fruit pulp was included. The antioxidant capacity of the EP fruit pulp was determined to be superior to the EO fruit pulp in every chemical based assay. In the cell-based CAP-e assay, the EP fruit pulp showed a dose-dependent inhibition against oxidative damage with an IC50 of 0.167 g/l. In the SEAP reporter assay, the EP fruit pulp polyphenol-rich extracts inhibited lipopolysaccharide (LPS)-induced NF-κB activation by 23% (p < 0.05) at 20 μg/ml, whereas the extract of the EO fruit pulp did not show a significant inhibitory effect at comparable doses. In addition, carotenoids were quantified for the first time in EP, since EP has high scavenging capacity against singlet oxygen.  相似文献

The effect of the disinfectant sodium hypochlorite (NaClO), with or without mild heat (50 degrees C) and fumaric acid, on native bacteria and the foodborne pathogens Staphylococcus aureus, Escherichia coli O157:H7, and Salmonella Typhimurium DT104 attached to iceberg lettuce leaves was examined. The retail lettuce examined consistently harbored 6 to 7 log CFU/g of native bacteria throughout the study period. Inner leaves supported 1 to 2 log CFU/g fewer bacteria than outer leaves. About 70% of the native bacterial flora was removed by washing five times with 0.85% NaCl. S. aureus, E. coli, and Salmonella allowed to attach to lettuce leaves for 5 min were more easily removed by washing than when allowed to attach for 1 h or 2 days, with more S. aureus being removed than E. coli or Salmonella Typhimurium. An increase of time for attachment of pathogens from 5 min to 2 days leads to decreased efficiency of the washing and sanitizing treatment. Treatment with fumaric acid (50 mM for 10 min at room temperature) was the most effective, although it caused browning of the lettuce, with up to a 2-log reduction observed. The combination of 200 ppm of sodium hypochlorite and mild heat treatment at 50 degrees C for 1 min reduced the pathogen populations by 94 to 98% (1.2- to 1.7-log reduction) without increasing browning.  相似文献

Coexpression of two classes of folding accessory proteins, molecular chaperones and foldases, can be expected to improve the productivity of soluble and active recombinant proteins. In this study, horseradish peroxidase (HRP), which has four disulfide bonds, was selected as a model enzyme and overexpressed in Escherichia coli. The effects of coexpression of a series of folding accessory proteins (DnaK, DnaJ, GrpE, GroEL/ES, trigger factor (TF), DsbA, DsbB, DsbC, DsbD, and thioredoxin (Trx)) on the productivity of active HRP in E. coli were examined. Active HRP was produced by very mild induction with 1 microM isopropyl-beta-D-thiogalactopyranoside (IPTG) at 37 degrees C, whereas the amount of active HRP produced by the induction with 1 mM IPTG was negligibly small. Active HRP production was increased significantly by coexpression of DsbA-DsbB (DsbAB) or DsbC-DsbD (DsbCD), while coexpression of molecular chaperones did not improve active HRP production. The growth of E. coli cells was inhibited significantly by the induction with 1 mM IPTG in a HRP single expression system. In contrast, when HRP was coexpressed with DsbCD, the growth inhibition of E. coli was not observed. Therefore, coexpression of Dsb proteins improves both the cell growth and the productivity of HRP.  相似文献

Faecal soluble lignin (FSL), extracted from the faeces of sheep that received orchardgrass and timothy, were compared with 90% dioxane-soluble lignins released by ball milling (MHL) and by subsequent enzymatic hydrolysis (EHL) from the same grasses. FSL contained much less carbohydrate and esterified p-coumaric and ferulic acids than MHL and EHL. Although no considerable differences were found in the yields of vanillin and syringaldehyde produced by nitrobenzene oxidation, the molar ratio of syringaldehyde to vanillin was higher for FSL than for MHL and EHL. Such differences in chemical properties among the lignin preparations were confirmed by 13C-NMR spectroscopy. FSL had a lower molecular size compared with MHL and EHL. Results indicated that syringyl-rich lignin fragments with less phenolic acid esters, probably depolymerisation products of the grass lignins, were released by digestion in sheep from the forage grasses.  相似文献

This paper reports on the impingement cooling characteristics of a heat sink with thin longitudinal fins of 0.2 mm thickness, which are spaced with a fin-pitch in the range 0.5 mm to 2.0 mm. The air cooling of the heat sink comes from a slot-shaped orifice positioned above the heat-sink center. The breadth of the gap between the fin tops and the inlet orifice is in the range 0 mm to 10 mm. The thermal resistance of the thin longitudinal fins used is about 50% to 57% that of the thick longitudinal fins now in commercial use. The cooling performance of the thin-plate fins is almost the same as that of optimally arranged pin-fins with the same total surface area. A maximum value of six times the heat transfer rate of a single flat plate having the same base area was observed for the thin-plate fins. A comparison of cooling performance between impingement and channel flow systems was conducted. The performance of impingement cooling systems is almost unaffected by the breadth of the gap between the fin tops and the inlet orifice (or, for channel cooling, the upper wall).
