Corynebacterium glutamicum is a biotin auxotrophic bacterium in which glutamate production is induced under biotin-limited conditions. During glutamate production, anaplerotic reactions catalyzed by phosphoenolpyruvate carboxylase (PEPC) and a biotin-containing enzyme pyruvate carboxylase (PC) are believed to play an important role in supplying oxaloacetate in the tricarboxylic acid cycle. To understand the distinct roles of PEPC and PC on glutamate production by C. glutamicum, we observed glutamate production induced under biotin-limited conditions in the disruptants of the genes encoding PEPC (ppc) and PC (pyc), respectively. The pyc disruptant retained the ability to produce high amounts of glutamate, and lactate was simultaneously produced probably due to the increased intracellular pyruvate levels. On the other hand, the ppc knockout mutant could not produce glutamate. Additionally, glutamate production in the pyc disruptant was enhanced by overexpression of ppc rather than disruption of the lactate dehydrogenase gene (ldh), which is involved in lactate production. Metabolic flux analysis based on the 13C-labeling experiment and measurement of 13C-enrichment in glutamate using nuclear magnetic resonance spectroscopy revealed that the flux for anaplerotic reactions in the pyc disruptant was lower than that in the wild type, concomitantly increasing the flux for lactate formation. Moreover, overexpression of ppc increased this flux in both the pyc disruptant and the wild type. Our results suggest that the PEPC-catalyzed anaplerotic reaction is necessary for glutamate production induced under biotin-limited conditions, because PC is not active during glutamate production, and overexpression of ppc effectively enhances glutamate production under biotin-limited conditions. 相似文献
Cancer stem cells (CSCs) are in general characterized by higher resistance to cell death and cancer therapies than non-stem differentiated cancer cells. However, we and others have recently revealed using glioma stem cells (GSCs) as a model that, unexpectedly, CSCs have specific vulnerabilities that make them more sensitive to certain drugs compared with their differentiated counterparts. We aimed in this study to discover novel drugs targeting such Achilles’ heels of GSCs as anti-GSC drug candidates to be used for the treatment of glioblastoma, the most therapy-resistant form of brain tumors. Here we report that domatinostat (4SC-202), a class I HDAC inhibitor, is one such candidate. At concentrations where it showed no or minimal growth inhibitory effect on differentiated GSCs and normal cells, domatinostat effectively inhibited the growth of GSCs mainly by inducing apoptosis. Furthermore, GSCs that survived domatinostat treatment lost their self-renewal capacity. These results suggested that domatinostat is a unique drug that selectively eliminates GSCs not only physically by inducing cell death but also functionally by inhibiting their self-renewal. Our findings also imply that class I HDACs and/or LSD1, another target of domatinostat, may possibly have a specific role in the maintenance of GSCs and therefore could be an attractive target in the development of anti-GSC therapies. 相似文献
Cholecystokinin (CCK) is a peptide hormone secreted from enteroendocrine cells and regulates the exocrine pancreas, gastric motility, and appetite. Dietary triacylglycerols are hydrolyzed to fatty acids (FA) and 2-monoacylglycerols (2-MAG) in the small intestine. Although it is well known that FA stimulate CCK secretion, whether 2-MAG have the CCK-releasing activity remains unclear. We examined the CCK-releasing activity of four commercially available 2-MAG in a murine CCK-producing cell line, STC-1, and the molecular mechanism underlying 2-MAG-induced CCK secretion. CCK released from the cells was measured using ELISA. Among four 2-MAG (2-palmitoyl, 2-oleoyl, 2-linoleoyl, and 2-arachidonoyl monoacylglycerols) examined, 2-arachidonoyl glycerol (2-AG) potently stimulated CCK secretion in a dose-dependent manner. Structurally related compounds, such as 2-arachidonoyl glycerol ether and 1-arachidonoyl glycerol, did not stimulate CCK secretion. Both arachidonic acid and 2-AG stimulated CCK secretion at 100 μM, but only 2-AG did at 50 μM. 2-AG-induced CCK secretion but not arachidonic acid-induced CCK secretion was attenuated by treatment with a cannabinoid receptor 1 (CB1) antagonist. These results indicate that a specific 2-MAG, 2-AG, directly stimulates CCK secretion via CB1. 相似文献
This article proposes a novel method of improving the overall coupling coefficient of a wireless power transfer system by using a four‐winding transformer of which two pairs of windings each are connected in parallel in the primary side and secondary side. By weakening the coupling coefficients kc between the parallel windings, the overall magnetic coupling between the primary side and secondary side of the transformer is improved. In order to confirm the validity of the proposed method, a conventional high‐kc four‐winding transmission coil and the proposed low‐kc four‐winding transmission coil are experimentally assessed. Then, the coils are introduced into a nonresonant inductive power transfer system utilizing a dual active bridge converter in order to confirm the correction of the input power factor. As a result, the overall magnetic coupling of the proposed four‐winding coils was found to be improved by 17% compared with the overall magnetic coupling of the conventional coil. Moreover, the input power factor for the entire load is corrected with the proposed four‐winding coil. Therefore, a low kc is effective in improving the overall magnetic coupling. 相似文献
A novel needle‐free injection system by electrically induced microbubbles has been developed. First of all, we evaluated the depth of injected reagent for evaluating injection performance. Then, the cavitation‐induced shock wave has been focused to magnify the injection performance by semispherical reflector. Sodium concentration is responsible for (at least part of) the stiffness of articular cartilage due to the osmotic pressure it generates. Therefore, we hypothesized that we could use sodium MRI to approximate the stiffness of cartilage to assess early cartilage degeneration.
Methods
Four human tibial plateaus were retrieved from patients undergoing total knee replacement (TKR), and their cartilage stiffness mapped with indentation testing, after which samples were scanned in a 7 T MRI to determine sodium concentration. The relation of biomechanical parameters to MRI sodium and glycosaminoglycan (GAG) concentration was explored by a linear mixed model.
Results
Weak correlations of GAG concentration with apparent peak modulus (p?=?0.0057) and apparent equilibrium modulus (p?=?0.0181) were observed and lack of correlation of GAG concentration versus MRI sodium concentration was observed. MRI sodium concentration was not correlated with apparent peak modulus, though a moderate correlation of MRI sodium concentration with permeability was shown (p?=?0.0014).
Discussion and conclusion
Although there was correlation between GAG concentration and cartilage stiffness, this was not similar with sodium concentration as measured by MRI. Thus, if the correlation between MRI sodium imaging and GAG concentration could be resolved, this strategy for assessing cartilage functional quality still holds promise.
Dynamic centrifuge model tests and associated analyses are carried out to develop a procedure for simulating the slope failure process during earthquakes and to evaluate the force of a sliding soil mass impacting a structure. This impact force is successfully measured in the dynamic centrifuge model tests using a slope height of 50 m in the prototype scale. The results of a pseudo-static limit equilibrium analysis, assuming a circular failure plane, show that a stability analysis can be an effective tool for evaluating the critical acceleration under which the slope failure starts, although it is not applicable for evaluating the runout distance or the impact force of the sliding soil mass to the structure. Therefore, an attempt is also made in this study to examine the applicability of empirical equations for evaluating the impact force by comparing the evaluated impact force using Hertz’s equation and an empirical equation based on fluid mechanics with the measured ones. The comparison reveals that these simple equations would not be able to estimate the impact force well, even though parameter setting is required for applying the equations. On the other hand, it is also found from a relevant analysis that the material point method (MPM) is an effective tool for simulating the failure behavior of a slope and for evaluating the impact force of a sliding soil mass induced by a slope failure. 相似文献
