The association between inflammatory cell influx, cell activation status and change of airway responsiveness to acetylcholine (ACh) after daily inhalation of ovalbumin (OA) in sensitized guinea-pigs was investigated. Starting 3 weeks after sensitization (OA at 50 mg/kg s.c.+i.p.) guinea-pigs were exposed daily to 2% OA (10 min; under cover of 0.5 mg/kg mepyramine i.p. 15 min before OA) for 2 weeks. Concentration-response curves (CRCs) for inhaled ACh were performed 24 h after the last OA-challenge and 24 h after another single OA-inhalation 1 week later. CRCs for inhaled ACh were neither affected 24 h after the last OA challenge (daily for two weeks) nor 24 h after another OA-inhalation one week later. In contrast, bronchoalveolar lavage (BAL) from repeatedly OA- sensitized/-challenged guinea-pigs immediately after the last CRC showed a significant increase of total cell count by about tenfold and increases in eosinophils by about 20-fold, neutrophils by 30-fold, macrophages by about fivefold and lymphocytes by about tenfold (P < 0.05, multiple Wilcoxon-test). In contrast, markers of cell activation (EPO, MPO) were significantly decreased (P < 0.05). Methylprednisolone almost completely prevented these changes in increased cell numbers and decreased cell activation (vs OA contr., P < 0.05). The lack of increased airway hyperresponsiveness despite a massive inflammatory cell influx suggests other factors controlling airway responsiveness than inflammation.  相似文献

The cerebral cortex of anaesthetised 2- to 12-day-old rats was superfused with artificial cerebrospinal fluid containing 100 mM acetate substituted for chloride to condition the brain for spreading depression (SD). After such superfusion, the earliest SD-like events were found at day 9 and full blown SD at day 10, whereas in the unconditioned brain the first SD occurred between days 12 and 15. Acetate conditioning of the cerebral cortex may be used to unmask neuronal and glial properties that are hidden in early stages of development.  相似文献

Tumor necrosis factor-alpha (TNF-alpha) is thought to be an important mediator in the pathogenesis of neonatal septicemia. To assess the role of TNF-alpha in neonatal sepsis, serum levels of TNF-alpha were measured in a group of neonates with septicemia and compared with the levels of gestational-postnatal, age-matched healthy controls. The relationships between severity of infection, the nature of causative microorganisms, and TNF-alpha levels were also investigated in this prospective study. A total of 49 infants (25 full-term, 24 preterm) with proven sepsis and 40 healthy infants (20 full-term, 20 preterm) were included. Serum TNF-alpha levels were measured using the TNF-alpha immunoradiometric assay. The median level of TNF-alpha was found to be significantly higher in infants suffering from sepsis (154 pg/mL) particularly in those with septic shock (242.5 pg/mL) as compared to healthy controls (61.5 pg/mL) (p < 0.001). No correlation was found between TNF-alpha and postnatal ages, gestational ages or birth weights of the infants. TNF-alpha levels were not different in surviving and terminal neonates. Although serum, TNF-alpha levels were found to be slightly higher in gram-negative septicemia, the difference was not significant. These findings suggest that TNF-alpha plays an important role in the pathophysiology of neonatal septicemia, but its importance as a prognostic factor is not yet clear.  相似文献

A solid solution of vanadium (V)-doped tin oxide (SnO2) particles of average diameter 2 nm and V content of 19 at.% was prepared at normal pressure and low temperature (100°C) by mixing wet SnO2 gel SnO2· x H2O with a boiling solution of vanadium oxide (V2O5). Experimental characterizations using X-ray, electron diffraction, and transmission electron microscope show evidence of a pure single phase. The nanoparticles exhibit a mixed magnetic behavior, namely paramagnetic and ferromagnetic. Their thermal stability is also investigated. At higher temperature, 850°C, some amount of Fe precipitates from the solid solution.  相似文献

Multiwalled carbon nanotube/polycaprolactone nanocomposites (MWNT/PCL) were prepared by in situ polymerization, whereby as‐received MWNTs (P‐MWNTs) and purified MWNTs (A‐MWNTs) were used as reinforcing materials. The A‐MWNTs were purified by nitric acid treatment, which introduced the carboxyl groups (COOH) on the MWNT. The micrographs of the fractured surfaces of the nanocomposites showed that the A‐MWNTs in A‐MWNT/PCL were better dispersed than P‐MWNTs in PCL matrix (P‐MWNT/PCL). Percolation thresholds of the P‐MWNT/PCL and A‐MWNT/PCL, which were studied by rheological properties, were found at ~2 wt % of the MWNT. The conductivity of the P‐MWNT/PCL was between 10?1 and 10?2 S/cm by loading of 2 wt % of MWNT although that of the A‐MWNT/PCL reached ~10?2 S/cm by loading of 7 wt % of MWNT. The conductivity of the P‐MWNT/PCL was higher than that of the A‐MWNT/PCL at the entire range of the studied MWNT loading, which might be due to the destruction of π‐network of the MWNT by acid treatment, although the A‐MWNT/PCL was better dispersed than the P‐MWNT/PCL. The amount of the MWNT at which the conductivity of the nanocomposite started to increase was strongly correlated with the percolation threshold. © 2007 Wiley Periodicals, Inc. Heats of solution at infinite dilution of solutes in poly(ethyl methacrylate) were estimated using gas–liquid chromatography over a temperature range of 417.74 K–427.55 K. The heat of solution is related to solute polarizability and dipole moment. Contributions of specific interactions such as hydrogen bonding and charge-transfer complexing to the enthalpy of solution were also determined.  相似文献

We characterize fracture and effective stress–strain graphs in 2D random composites subjected to a uniaxial in-plane uniform strain. The fibers are arranged randomly in the matrix. Both fibers and matrix are isotropic and elastic–brittle. We conduct this analysis numerically using a very fine two-dimensional triangular spring network and simulate the crack initiation and propagation by sequentially removing bonds which exceed a local fracture criterion. In particular, we focus on effect of geometric randomness on crack path of random composites. Based on that two stochastic micro-mechanic models are presented that can predict with confidence the failure probability of random matrix–inclusion composites.  相似文献

The use of eco-freindly polymers as antimicrobial materials is in growth due to the need to reduce the negative impact of conventional treatments on the environment and the human health. The purpose of the present study was to assess the antifungal properties of films and solutions based on chitosan with different molecular weight at different concentrations. Surfactants were added to the formulation to assess their impact on treatment efficiency. The antifungal activity was conducted against tree fungi, Aspergillus niger, Alternaria alternata and Rhizopus oryzae. Results indicated important and significant differences of the antifungal activity between chitosan based solutions and chitosan based films. Furthermore, the antifungal activity of the different treatment depended on the type of fungus treated. Thus, chitosan film treatments were significantly more effective on A. niger than solution treatments. On the other hand, solution treatments resulted in higher radial inhibition when applied against A. alternata or R. oryzae. The highest radial inhibition was observed against A. alternata (97%) using a chitosan solution. The influence of the other parameters (concentration, molecular weight and surfactant type) on treatment efficiency was not as important and their significance depended on treatment type and fungus nature.  相似文献

Abstract— In an LCOS projection system, a wire‐grid PBS may be used to improve contrast and a field lens may be employed to simplify the projection lens. However, the combination of the two leads to a reduced contrast, which decreases with increasing field angle in one direction. In a representative arrangement, measured contrast with a mirror varied from over 10,000 in the center to 450 at the left and right edges. Contrast loss can be improved with a weaker field lens. The problem arises due to a phase delay between polarization states introduced by the wire grid. Polarization measurements show a +53‐nm phase delay with an axis parallel to the wire‐grid surface normal. Various compensation schemes are proposed in order to reduce the aforementioned loss of contrast.  相似文献
