Alloreactive T lymphocytes that respond directly to foreign major histocompatibility complex (MHC) molecules and bound peptide are known to be central mediators of graft-versus-host disease (GVHD) and allograft rejection. We have recently identified a peptide from the human protein, cytochrome P450 (isotypes IIC9, 10, or 18), that is recognized in association with human leukocyte antigen (HLA) B*3501 by alloreactive cytotoxic T lymphocytes (CTLs). These CTLs with this specificity were isolated from several unrelated individuals and were found to express a common T-cell receptor (TCR). Synthetic analogs of the cytochrome P450 peptide were generated by introducing single amino acid substitutions at putative TCR contact positions. Four altered peptide ligands were powerful competitive antagonists of these CTL clones, reducing lysis levels of target cells expressing the alloantigen HLA B*3501 by over 80%. This first demonstration that it is possible to suppress CTL alloreactivity with structural variants of allodeterminants raises the prospect that such TCR antagonists could be exploited within the clinical arena to specifically modulate GVHD and allograft rejection.  相似文献

Secondary electron imaging is not possible in the variable pressure scanning electron microscope because the mean free path of the secondaries in the gas is too short to permit them to reach the detector. This paper therefore investigates an alternative strategy for producing an image containing significant amounts of secondary electron contrast. This involves modifying the microscope by the addition of a biased electrode above the sample and then collecting a specimen current signal. This system, originally described by Farley and Shah (1988), is found to produce true secondary electron detail over a wide range of conditions.  相似文献

In this work, an amplify‐and‐forward variable‐gain relayed mixed RF‐FSO system is studied. The considered dual‐hop system consists of a radio frequency (RF) link followed by a free space optical (FSO) channel. The RF link is affected by short‐term multipath fading and long‐term shadowing effects and is assumed to follow the generalized‐K fading distribution that approximates accurately several important distributions often used to model communication channels. The FSO channel experiences fading caused by atmospheric turbulence that is modeled by the gamma‐gamma distribution characterizing moderate and strong turbulence conditions. The FSO channel also suffers path loss and pointing error induced misalignment fading. The performance of the considered system is analyzed under the collective influence of distribution shaping parameters, pointing errors that result in misalignment fading, atmospheric turbulence, and path loss. The moment‐generating function of the Signal power to noise power ratio measured end‐to‐end for this system is derived. The cumulative distribution function for the Signal power to noise power ratio present between the source and destination receiver is also evaluated. Further, we investigate the error and outage performance and the average channel capacity for this system. The analytical expressions in closed form for the outage probability, symbol and bit error rate considering different modulation schemes and channel capacity are also derived. The mathematical expressions obtained are also demonstrated by numerical plots.  相似文献

Automatic annotation of images is one of the fundamental problems in computer vision applications. With the increasing amount of freely available images, it is quite possible that the training data used to learn a classifier has different distribution from the data which is used for testing. This results in degradation of the classifier performance and highlights the problem known as domain adaptation. Framework for domain adaptation typically requires a classification model which can utilize several classifiers by combining their results to get the desired accuracy. This work proposes depth-based and iterative depth-based fusion methods which are basically rank-based fusion methods and utilize rank of the predicted labels from different classifiers. Two frameworks are also proposed for domain adaptation. The first framework uses traditional machine learning algorithms, while the other works with metric learning as well as transfer learning algorithm. Motivated from ImageCLEF’s 2014 domain adaptation task, these frameworks with the proposed fusion methods are validated and verified by conducting experiments on the images from five domains having varied distributions. Bing, Caltech, ImageNet, and PASCAL are used as source domains and the target domain is SUN. Twelve object categories are chosen from these domains. The experimental results show the performance improvement not only over the baseline system, but also over the winner of the ImageCLEF’s 2014 domain adaptation challenge.  相似文献

In this study, fixed bed model and reduced lumped diffusion model were used to explain biosorption behaviour of Cu (II) in a continuous column of Aspergillus niger biomass. The breakthrough time was evaluated for both models as a function of influent flow rate and bed height for a feed solution at 10 mg/L metal ion concentration and compared with experimental results. MATLAB was used to solve the partial differential equations and breakthrough curve was plotted. The breakthrough time obtained experimentally was comparable with data evaluated by these models. It was observed that fixed bed model and reduced lumped diffusion model can predict column dynamics more accurately at high flow rate, that is, at 9.8 mL/min for all (2.1, 3.1, and 4.1 cm) bed heights. © 2011 Canadian Society for Chemical Engineering  相似文献

The paper reports the energization of Hydroxyl‐Terminated Polybutadiene (HTPB) by functionalizing explosophore  NO2 over the HTPB backbone, resulting in the formation of conjugated nitro‐alkene derivative of HTPB. A convenient, inexpensive and efficient “one pot” procedure of synthesizing Nitro‐Functionalized Hydroxyl‐Terminated Polybutadiene (Nitro‐HTPB) is reported. The reaction was carried out with sodium nitrite and iodine. To retain the unique physico‐chemical properties of HTPB, functionalization by  NO2 group was restricted to 10 to 15 % of double bonds. The Nitro‐HTPB was characterized by FTIR, 1H NMR, VPO, DSC, TGA etc. The polymer has shown good thermal stability for practical applications. The kinetic parameters for the decomposition of Nitro‐HTPB at 150–300 °C were obtained from non‐isothermal DSC data.  相似文献

Curcumin, the principal curcuminoid of turmeric, is responsible for its yellow colour and serves as a measure of turmeric quality. The Prevention of Food Adulteration (PFA) Act of India allows only Curcuma longa L. for the production of turmeric powder and prohibits addition of any foreign matter/artificial colour, but it does not specify a minimum curcumin content. The present surveillance was undertaken to study the quality of loose versus branded turmeric powders vis-à-vis curcumin content and the presence of unwarranted extraneous colours from city markets in India using a newly developed two-dimensional high-performance thin-layer chromatography (2D-HPTLC) method. The results show that curcumin content in branded samples ranged from 2.2% to 3.7%, while non-branded samples had from 0.3% to 2.6%. Though none of the branded turmeric powders contained artificial colours, 17% of loose powders showed the presence of extraneous colour metanil yellow, in the range 1.0–8.5 mg g?1, which may pose health threats. Low curcumin content in the analysed samples may be due to mixing of other curcuma species or their curcumin-depleted matrices and foreign starches as cheaper alternatives. This is supported by the fact that major Indian turmeric trade types are known to possess curcumin contents ranging from 2.1% to 8.6%, with an average of 4.8%. There is thus an urgent need to prescribe realistic curcumin limits for turmeric powder, otherwise there is no obligation on the part of traders to stick to any minimum levels and consumers will keep on getting this nutrient-depleted household spice.  相似文献

Novolak resins provide the best overall performance for “g” and “i”-line photoresists. There is a continuing need for advanced novolak designs that will provide improved lithographic, thermal, and etch characteristics that may be reproducibly synthesized. A novolak synthesis process was developed using the solution condensation technique. Cresol mixtures with m-cresol and 3,5-xylenol at specific ratios provide reproducible novolaks with controlled molecular weights. In order to achieve high thermal and etch performance, while retaining photospeed and resolution characteristics, three basic approaches were investigated: (1) increase in molecular weight, which produces novolaks with Tg ranging from 120 to 130°C with relatively slow dissolution rates; (2) incorporation of multi-hydroxyphenols such as resorcinol to tailor the dissolution rate, resolution, thermal, etch, and adhesion characteristics; (3) partial esterification of multi-hydroxy novolaks giving a Tg range of 140 to 150°C. Lithographic evaluation of the novolak resins was performed by formulating with a 2,1,4-diazonaphthoquinone (DNQ) sensitizer. Results on resin synthesis, molecular weights, lithographic, thermal, and etch characteristics are discussed.  相似文献
