Within the reactive oxygen species (ROS) generated by cellular metabolisms, hydroxyl radicals (HO) play an important role, being the most aggressive towards biomolecules. The reactions of HO with methionine residues (Met) in peptides and proteins have been intensively studied, but some fundamental aspects remain unsolved. In the present study we examined the biomimetic model made of Ac-Met-OMe, as the simplest model peptide backbone, and of HO generated by ionizing radiation in aqueous solutions under anoxic conditions. We performed the identification and quantification of transient species by pulse radiolysis and of final products by LC-MS and high-resolution MS/MS after γ-radiolysis. By parallel photochemical experiments, using 3-carboxybenzophenone (CB) triplet with the model peptide, we compared the outcomes in terms of short-lived intermediates and stable product identification. The result is a detailed mechanistic scheme of Met oxidation by HO, and by CB triplets allowed for assigning transient species to the pathways of products formation.  相似文献

The authors show that a narrowband noise (NBN) is perceived as longer when presented immediately after a wideband noise (WBN), compared to when the WBN is absent. This effect depended on the WBN’s frequency spectrum overlapping that of the NBN, and it increased as the duration of the WBN increased up to 300 ms. It decreased when a silent gap was introduced between the WBN and NBN, but remained significant for an easily detectable gap of 40 ms. A correlate of the effect was observed in the mismatch negativity (MMN) to a deviant stimulus, consisting of a WBN + NBN, presented in a sequence of more common isolated WBNs. The MMN latency was longer for an on-frequency than for an off-frequency WBN; and, more importantly, the size of this difference correlated across participants with the difference in perceived duration. A rhythm-adjustment experiment showed that the presence of an on-frequency WBN immediately preceding a tone caused that tone to be heard as starting earlier than when the WBN was absent. Operational remote monitoring of snowpack stratigraphy, melt water intrusions and their evolution with time for forecasting snowpack stability is not possible to date. Determination of the spatial variability of snowpack conditions on various scales requires a number of point measurements with various methods. These methods are either destructive or do not provide information about the internal structure of the snowpack. The application of a remotely controlled non-destructive sensor system would help to gain a higher spatio-temporal resolution about information of the snowpack. In this study we present results from upward-looking ground-penetrating radar (GPR) surveys from horizontal caves dug in the front wall of snow pits at the bottom of the snowpack. GPR data are compared with vertical profiles of snow hardness and density, obtained in the snow pit. Data were acquired in different areas with varying snow conditions with various GPR impulse systems, frequencies and polarizations. Radar experiments with high frequencies (> 1 GHz) detect internal layers in the snowpack in dry snow, but fail to provide clear reflections at the upper snow-air transition because of attenuation. In wet snow, the radar signals < 1 GHz are capable to penetrate a meter-thick snowpack and detect the snow surface, although the signal is strongly attenuated. Analysis of reflection phases and magnitudes allows interpretation of their physical origin in terms of changes in electrical permittivity. Varying antenna polarization causes a strongly different signal response, likely caused by the snow-pit wall present in our set-up. Forward calculation of density-based reflection coefficients between neighboring layers of varying hardness yields ambiguous results in terms of correspondence with observed radar reflections apart except for interferences of neighboring reflections. Moreover, we identify several pitfalls for future applications. The system set-up used here represents a basis for further developments towards a system, which is capable of improving information on the spatial and temporal snowpack characteristics.  相似文献

Cryogenic probe technology can significantly compensate for the inherently low sensitivity of natural abundance 13C NMR spectroscopy. This now permits its routine use in NMR spectroscopy of biofluids, such as urine or plasma, with acquisition times that enable a high throughput of samples. Metabonomic studies often generate numerous samples in order to characterize fully the time-dependent biochemical response to stimuli, but until now, they have been largely conducted using 1H NMR spectroscopy because of its high sensitivity and hence efficient data acquisition. Here, we demonstrate that information-rich 13C NMR spectra of rat urine can be obtained using appropriately short acquisition times suitable for biochemical samples when using a cryogenic probe. Furthermore, these data were amenable to automated pattern recognition analysis, which produced a profile of the metabolic response to the model hepatotoxin hydrazine that was consistent with earlier studies. Thus, a new source of detailed and complementary information is available to metabonomics using cryogenic probe 13C NMR spectroscopy.  相似文献

Pipe fabrication for the repair sector. The paper discusses the changes in pipe fabrication that have taken place in recent years. These enable repair work to be carried out more efficiently, without thereby impairing the flexibility and speed of operation that are essential to maintenance work. A prerequisite for this is that work on the pipes be transferred to workshops. Only here is it possible for manual operations, such as manual welding, to be reduced to a minimum and be replaced by mechanized or semi-mechanized welding methods. The use of modern machines for bending the pipes is more economic than the welding in of pipe bends. Necking out of the pipes by mechanical means or the use of mechanically pre-fabricated connections permit optimum preparation of the weld seam for branch fittings. Any additional work on the pipes, where this is necessary, is carried out at workplaces specially designed for the purpose.  相似文献

Two phenolic acids were identified in the collembolan Ceratophysella denticulata: 3-hydroxy-4,5 dimethoxy benzoic acid and 4-hydroxy-3,5-dimethoxybenzoic acid (syringic acid). These are localized on or in the integument of the springtail, in field-collected animals, in a ratio of 47:100 (v/v). Springtails kept under different temperature regimes showed differences in production and ratio of the benzoic acid derivatives. At 20 degrees C, C. denticulata produced only syringic acid, whereas at 10 degrees C both isomers in a ratio of 100:61 (v/v) were detected. Bioassays with C. denticulata as well as with the specialized collembolan predator Stenus comma (Staphylinidae) were carried out. Staphylinid beetles topically treated with the acids try to clean their mouthparts by rubbing them on the ground significantly more often than do control beetles. Both compounds individually and as a natural mixture have deterrent effects towards the predator S. comma.  相似文献

Systems biology is a reemerging paradigm which, among other things, focuses on mathematical modeling and simulation of biochemical reaction networks in intracellular processes. For most simulation tools and publications, they are usually characterized by either preferring stochastic simulation or rate equation models. The use of stochastic simulation is occasionally accompanied with arguments against rate equations. Motivated by these arguments, we discuss in this paper the relationship between these two forms of representation. Toward this end, we provide a novel compact derivation for the stochastic rate constant that forms the basis of the popular Gillespie algorithm. Comparing the mathematical basis of the two popular conceptual frameworks of generalized mass action models and the chemical master equation, we argue that some of the arguments that have been put forward are ignoring subtle differences and similarities that are important for answering the question in which conceptual framework one should investigate intracellular dynamics.  相似文献

International surveys of occupational injuries among seafarers have so far been missing. It was the aim to test the method of self-report of injuries and length of time at risk during the latest duty period and second to study the injury incidence rate among seafarers by use of the method. A pilot study was conducted (n = 1068) in Finland, Denmark, the Philippines, Croatia and Spain using self-completed questionnaires with questions about the person, the ship, the duration of latest duty period and injuries. The duration of the self-reporting duty period was in the Danish part compared with information from the crew register of the Maritime Authority. For seafarers from merchant ships in the Danish sub-study there was acceptable correspondence between the information from the seafarers and the Maritime Authority, but not when referring to ferries and non-specified types of ship. Unadjusted and adjusted injury incidence rates-ratios (IRRs) based on number of injuries per number of work hours were calculated. Adjusted IRRs for ordinary seamen/officers: IRR = 2.43 (95% CI: 1.25-4.72); for age < 35/35+ years: IRR = 1.97 (1.02-3.81); length of tour: 117 days or longer compared with < 117 days: IRR = 0.46 (95% CI: 0.22-0.95); 57-70 working hours per week compared with < 57 h: IRR = 1.26 (0.48-3.29), 71+h compared with < 57 h: IRR = 2.12 (0.84-5.36). Non-significant IRRs >1.00 were found for ships under 10,000 GT compared with larger ships and for own flagged ships compared with ships under flag of convenience. In conclusion, more than 70 h of work per week was related to a higher rate of injuries for seafarers on merchant ships, but the result was not statistically significant. Self-report of the duration of the latest tour of duty is useful for seafarers from merchant ships with short-term employments, but not for ferries and other, non-specified types of ship with other or permanent employment.  相似文献
