Methods of inhibiting the hyperacute antibody-mediated rejection that occurs when pig organs are transplanted into primates have been investigated using the baboon as a potential recipient. Baboons were treated with different regimens that included combinations of (1) splenectomy, (2) pharmacologic immunosuppression (CsA, cyclophosphamide, corticosteroids +/- methotrexate), and (3) intravenous infusion of oligosaccharides. The cytotoxicity of the serum was then assessed on cultures of pig kidney cells (PK15). Unmodified serum caused approximate 65-100% pig cell destruction. Splenectomy and/or pharmacologic immunosuppression, and infusions of dextran, dextrose or mannitol, did not result in any reduction of cytotoxicity. Infusions of melibiose and/or arabinogalactan, both of which have terminal non-reducing alpha-galactose, however, decreased relative PK15 cell damage significantly in a dose-dependent manner. At high concentrations (< or = 50 g/hr), complete inhibition of cytotoxicity was achieved in 4 of 15 baboons. The extracorporeal immunoadsorption of baboon serum utilizing immunoaffinity columns of melibiose also resulted in a significant reduction (of approximately 80%) in cytotoxic effect. In 1 baboon, melibiose and arabinogalactan infusion delayed vascular rejection of a pig cardiac xenograft from 10 min to about 12 hr, at which time the baboon died from the toxic effects of the carbohydrate infusion. These observations (1) add further support to the role that anti-alpha-galactosyl antibodies play in the hyperacute rejection of pig tissues transplanted into primates, and (2) demonstrate that serum cytotoxicity can be reduced by the intravenous infusion of alpha-galactosyl oligosaccharides or by extracorporeal immunoadsorption using these carbohydrates.  相似文献

A 1.5-μm multiquantum well amplifier divided into three sections with short contacts at the input and output facets is described. A simple derivation shows that the amplifier optical gain is proportional to the ratio of voltage changes at the facet contacts induced by the optical signal. We demonstrate greater than 90% quantum efficiency in an In0.53Ga0.47As photodetector with a thin (900 Å) absorbing layer. This was achieved by inserting the In0.53 Ga0.47As/InP epitaxial layer into a microcavity composed of a GaAs/AlAs quarter-wavelength stack (QWS) and a Si/SiO2 dielectric mirror. The 900-Å-thick In0.53 Ga0.47As layer was wafer fused to a GaAs/AlAs mirror, having nearly 100% power reflectivity. A Si/SiO2 dielectric mirror was subsequently deposited onto the wafer-fused photodiode to form an asymmetric Fabry-Perot cavity. The external quantum efficiency and absorption bandwidth for the wafer-fused RCE photodiodes were measured to be 94±3% and 14 nm, respectively. To our knowledge, these wafer-fused RCE photodetectors have the highest external quantum efficiency and narrowest absorption bandwidth ever reported on the long-wavelength resonant-cavity-enhanced photodetectors  相似文献

Emergencies, and efforts to address them, create disruptions to local and global supply chains and surges in demand of emergency resources, which substantially affect global production. Reconfigurable manufacturing systems are promising solutions to improve flexibility and to reduce the effort needed to adapt supply chains and production networks to fit a perturbed environment. This paper proposes a method for coordination of reconfigurable manufacturing resources from multiple enterprises to structure ad-hoc production networks for critical products required in emergencies. Network optimization models and interaction algorithms are integrated to evolve the production network through synchronous machine-level and network-level reconfiguration driven by data.  相似文献

Noncommensurate 2D interfaces hold great promise toward low friction and nanoelectromechanical applications. For identical constituents, the crystals interlock at specific rotational configurations leading to high barriers for slide. In contrast, nonidentical constituents comprising different lattice parameters should enable robust superlubricity for all rotational configurations. This is however not the case for gold–graphite interfaces, as both theory and experiments show scaling behavior of the sliding force as a function of the interface contact area. By simulating the sliding force for gold–graphite interfaces, this work shows that the origin for high force barriers at special angular configurations is a result of commensurability between the moiré structure and the contact geometry. Consequently, this paper suggests new geometries that can potentially overcome such commensurability effects to enable robust superlubricity.  相似文献

Conclusions A method for producing roof products using magnesite-chromite clinker from caustic magnesite and Kimpersai chromite was developed and tested on an industrial scale.
