A new type of tracking analog-to-digital converter, which is capable of responding quickly to step changes in the input voltage, has been developed. The device produces an error signal which indicates the difference between the converter's state and the input signal level. This error signal is used to modify the converter's slew rate in such a way as to optimize convergence back to the tracking condition (i.e., the condition at which the converter's state accurately tracks the input signal). This error signal is also used to generate an accuracy code which indicates if the converter is accurately tracking the input signal, or if the converter is out of track, how many bits of the encoded data are actually significant. An eight-bit transient recorder was developed using the converter described above. The instrument was found to be capable of recording data at rates up to 10(6) points/s, and was observed to respond to step voltage changes within eight datum points.  相似文献

Magnetic resonance (MR) imaging is attractive for a noninvasive and radiation-free assessment of in vivo trabecular bone architecture. However the quantitative evaluation of architectural parameters could be biased by the limited sensitivity of MR. The aim of this study was to determine the accuracy of trabecular bone architectural parameters obtained from 3D high-resolution MR images, by comparison to reference images obtained by high-resolution X-ray microtomography using synchrotron radiation, from 29 samples of human calcaneus. MR images were obtained with a 66 m×66 m×66 m voxel size, using a 8.5 T MR microscope. Microtomography images were acquired with a 10 m×10 m×10 m voxel size, from the same samples. 3D architectural parameters characterizing the morphometry, topology, anisotropy, and orientation were computed from both modalities and carefully compared. To avoid errors, an identical region of interest was selected in the two corresponding images, and the same algorithms were run at identical spatial resolution. Our results establish that network connectivity, orientation and anisotropy are reliable from the MR data. The bone volume fraction, and morphometric parameters measured from the MR data, were found to be biased with respect to their values from the microtomography data, although there was a significant correlation between the two modalities.An erratum to this article can be found at  相似文献

To date, there are a few technologies for the development of noncompetitive immunoassays for small molecules, the most common of which relies on the use of anti-immunocomplex antibodies. This approach is laborious, case specific, and relies upon monoclonal antibody technology for its implementation. We recently demonstrated that, in the case of monoclonal antibody-based immunoassays, short peptide loops isolated from phage display libraries can be used as substitutes of the anti-immunocomplex antibodies for noncompetitive immunodetection of small molecules. The aim of this work was to demonstrate that such phage ligands can be isolated even when the selector antibodies are polyclonal in nature. Using phenoxybenzoic acid (PBA), a major pyrethroid metabolite, as a model system, we isolated the CFNGKDWLYC peptide after panning a cyclic peptide library on the PBA/anti-PBA immunocomplex. The sensitivity of the noncompetitive enzyme-linked immunosorbent assay (ELISA) setup with this peptide was 5-fold (heterologous) or 400-fold (homologous) higher than that of the competitive assay setup with the same antibody. Phage anti-immunocomplex assay (PHAIA) was also easily adapted into a rapid and highly sensitive dipstick assay. The method not only provides a positive readout but also constitutes a major shortcut in the development of sensitive polyclonal-based assays, avoiding the need of synthesizing heterologous competing haptens.  相似文献

Due to their size, small molecules cannot be simultaneously bound by two antibodies, precluding their detection by noncompetitive two-site immunoassays, which are superior to competitive ones in terms of sensitivity, kinetics, and working range. This has prompted the development of anti-immune complex antibodies, but these are difficult to produce, and often exhibit high cross-reactivity with the unliganded primary antibody. This work demonstrates that anti-immune complex antibodies can be substituted by phage particles isolated from phage display peptide libraries. Phages bearing specific small peptide loops allowed to focus the recognition to changes in the binding area of the immune complex. The concept was tested using environmental and drug analytes; with improved sensitivity and ready adaptation into on-site formats. Peptides specific for different immune complexes can be isolated from different peptide libraries in a simple and systematic fashion allowing the rapid development of noncompetitive assays for small molecules.  相似文献

Cloning and characterization of a Choristoneura fumiferana ultraspiracle (Cfusp) cDNA are described. First, a PCR fragment and then a cDNA clone (4.4 kb) were isolated from spruce budworm cDNA libraries. Comparison of the deduced amino acid sequence of this cDNA with the sequences in Genbank showed that this sequence had high homology with the ultraspiracle cDNAs cloned from Drosophila melanogaster (Dmusp), Bombyx mori (Bmusp), Manduca sexta (Msusp), and Aedes aegypti (Aausp). The Cfusp cDNA contained all the regions that are typical for a steroid/thyroid hormone receptor superfamily member. The DNA binding domain or C region was the most conserved sequence among all the usps. The A/B, D, and E regions also showed high amino acid identity with the amino acid sequences of Dmusp, Msusp, Bmusp, and Aausp. The Cfusp 4.5-kb mRNA was present in the embryos, in all larval stages, and in the pupae. The Cfusp mRNA levels in the midgut increased during the sixth-instar larval development and reached peak levels during the ecdysteroid raises for the pupal molt. However, Cfusp mRNA levels remained unchanged in the midgut of fifth-instar larvae, and in the epidermis and fat body of sixth-instar larvae indicating both a tissue- and stage-specific regulation of Cfusp mRNA expression.  相似文献

We present a numerical study of dephasing of electron spin ensembles in a diffusive quasi-one-dimensional GaAs wire due to the D’yakonov–Perel’ spin-dephasing mechanism. For widths of the wire below the spin precession length and for equal strength of Rashba and linear Dresselhaus spin–orbit fields a strong suppression of spin-dephasing is found. This suppression of spin-dephasing shows a strong dependence on the wire orientation with respect to the crystal lattice. The relevance for realistic cases is evaluated by studying how this effect degrades for deviating strength of Rashba and linear Dresselhaus fields, and with the inclusion of the cubic Dresselhaus term.  相似文献

The in-vessel control coil (IVCC) system, which has been designed for dedication of various active feedback plasma control functions, successfully fabricated and installed in the vacuum vessel of the Korea Superconducting Tokamak Advanced Research (KSTAR). The IVCC system consists of sixteen segmented coils that were independently fabricated outside the vacuum vessel and installed without any inside welding or brazing joints. The segmented coil system has several advantages such as eliminating possibility of cooling water leakage at the welded or brazed joints, simplification in fabrication and installation, and easy repair and maintenance of the coil system. Each segment contains eight oxygen-free high conductive coppers, which are grouped to four pairs, called as sections. Consequently, a segmented coil forms four sections for position control, field error correction (FEC), and resistive wall mode (RWM) control in accordance with electrical connection outside the cryostat. The eight conductors (or four sections) with internal coolant holes were enclosed in a rectangular welded jacket made of stainless steel 316LN and electrically insulated from the conductors by epoxy/glass composite layers. This coil system was commissioned up to 5 kA (30 kA-turns) for 5 s to achieve tentative use for the fast vertical plasma position control in the 2010 campaign of the KSTAR. This paper describes the several remarkable results in the fabrication and installation of the IVCC as well as commissioning results.  相似文献

Most data-mining algorithms assume static behavior of the incoming data. In the real world, the situation is different and most continuously collected data streams are generated by dynamic processes, which may change over time, in some cases even drastically. The change in the underlying concept, also known as concept drift, causes the data-mining model generated from past examples to become less accurate and relevant for classifying the current data. Most online learning algorithms deal with concept drift by generating a new model every time a concept drift is detected. On one hand, this solution ensures accurate and relevant models at all times, thus implying an increase in the classification accuracy. On the other hand, this approach suffers from a major drawback, which is the high computational cost of generating new models. The problem is getting worse when a concept drift is detected more frequently and, hence, a compromise in terms of computational effort and accuracy is needed. This work describes a series of incremental algorithms that are shown empirically to produce more accurate classification models than the batch algorithms in the presence of a concept drift while being computationally cheaper than existing incremental methods. The proposed incremental algorithms are based on an advanced decision-tree learning methodology called “Info-Fuzzy Network” (IFN), which is capable to induce compact and accurate classification models. The algorithms are evaluated on real-world streams of traffic and intrusion-detection data.  相似文献

The reflectivity of grazing angle X-ray mirrors, used for X-ray deep lithography, is tested by means of a calorimetric method. A deviation in the reflectivity of a used mirror compared with the reflectivity of a clean surface is observed. This deviation is caused by an oxide layer on the mirrors surfaces. The density, thickness and roughness of the assumed oxide layers are determined experimentally.  相似文献

E-beam lithography is a well-known technology used in the structuring of resist for mask fabrication. In the LIGA process E-beam lithography is used to fabricate the first X-ray mask. Due to the high precision of X-ray lithography patterning errors and defects are transformed into the several hundred micrometers thick resist structures. The side walls of these high-aspect-ratio structures are often used as optical mirrors, for which very good surface properties are essential. Deviations in the shape of the side walls even far below the wavelength of the used light lead to spurious strayed or misguided light. For grating microspectrometers the requirements are particularly stringent. The width of the grating teeth does not only have to be precise, but also the grating teeth have to be positioned accurately along the entire grating width. Using LIGA microspectrometers as an example, deviations in masks and LIGA-structured side walls were studied and subsequent correlations made with the corresponding e-beam writing pattern.  相似文献
