RuII-arene complexes provide a versatile scaffold for novel anticancer drugs. Seven new RuII-arene-thiocarboxylato dimers were synthesized and characterized. Three of the complexes ( 2 a , b and 5 ) showed promising antiproliferative activities in MDA-MB-231 (human invasive breast cancer) cells, and were further tested in a panel of fifteen cancerous and noncancerous cell lines. Complex 5 showed moderate but remarkably selective activity in MDA-MB-231 cells (IC50=39±4 μm Ru). Real-time proliferation studies showed that 5 induced apoptosis in MDA-MB-231 cells but had no effect in A549 (human lung cancer, epithelial) cells. By contrast, 2 a and b showed moderate antiproliferative activity, but no apoptosis, in either cell line. Selective cytotoxicity of 5 in aggressive, mesenchymal-like MDA-MB-231 cells over many common epithelial cancer cell lines (including noninvasive breast cancer MCF-7) makes it an attractive lead compound for the development of specifically antimetastatic Ru complexes with low systemic toxicity. 相似文献

In conventional flash sintering, the current rises nonlinearly to a set current limit, accompanied by a spike in the power density. This sudden power spike may cause hot spot formation, in which current preferentially channels through a small area, causing localized melting while other areas remain unsintered. By using a controlled current ramp early on the sudden power spike can be avoided. In addition, by changing the ramp rate material properties such as porosity, grain size and conductivity can be tuned. 相似文献

A batch processing method is used to fabricate foams comprising of a blend of poly(lactic acid) (PLA) and Novatein, a protein‐based thermoplastic. Various compositions of Novatein/PLA are prepared with and without a compatibilizer, PLA grafted with itaconic anhydride (PLA‐g‐IA). Pure Novatein cannot form a cellular structure at a foaming temperature of 80 °C, however, in a blend with 50 wt % of PLA, microcells form with smaller cell sizes (3.36 µm) and higher cell density (8.44 × 1021 cells cm?3) compared to pure PLA and blends with higher amounts of PLA. The incorporation of 50 wt % of semicrystalline Novatein stiffens the amorphous PLA phase, which restrains cell coalescence and cell collapse in the blends. At a foaming temperature of 140 °C, NTP30–PLA70 shows a unique interconnected porous morphology which can be attributed to the CO2‐induced plasticization effect. © 2017 Wiley Periodicals, Inc. J. Carbon capture and storage (CCS) is a means for reducing carbon dioxide (CO2) emissions from fossil fuel combustion in power generation and industrial processes. It involves the capture of CO2 for subsequent storage in various geological formations. The selection and matching of the power plants and storage sites are often an issue of optimisation due to various constraints, i.e., time of availability, injection rate, and storage capacity limits. In this work, a novel graphical targeting tool based on pinch analysis is proposed to address the planning problem of the storage of captured CO2 from power generating plants into corresponding reservoirs. The main consideration for the problem is the time of availability of the latter, since reservoirs need to be developed prior to CO2 storage. The time limitation is addressed by the graphical technique where time is taken as the governing element in solving the problem. CD26 has been reported as a marker for colorectal cancer stem cells endowed with tumor-initiating properties and capable of colorectal cancer (CRC) metastasis. In this study, we investigated the functional effect of CD26 on CRC angiogenesis and metastasis, and the potential underlying mechanism. The functional effects of CD26 overexpression or repression were determined by a wound healing experiment, and cell migration and invasion assays in vitro and in mouse models. Differentially expressed genes regulated by CD26 were identified by genome-wide mRNA expression array and validated by quantitative PCR. CD26 functionally regulated CRC cell migration and invasion in vitro and angiogenesis and metastasis in vivo. Genome-wide mRNA expression array and qPCR showed that MMP1 was up-regulated in CD26+ subpopulation, and a subsequent experiment demonstrated the regulatory effect of CD26 on MMP1 in CRC cell lines with CD26 repression or overexpression. Furthermore, overexpression of CAV1 abrogated the CD26-regulated MMP1 induction in CRC cell lines. This study demonstrated the functional roles of CD26 in inducing CRC migration, invasion, angiogenesis and metastasis and identified the potential involvement of MMP1 and CAV1 in such process. CD26 is an attractive therapeutic target for combating tumor progression to improve the prognosis of CRC patients. 相似文献

Proton exchange membranes were prepared by radiation‐induced grafting of styrene onto commercial poly(tetrafluoroethylene‐co‐hexafluoropropylene) films using a simultaneous irradiation technique followed by a sulfonation reaction. The resulting membranes were characterized by measuring their physicochemical properties such as water uptake, ion exchange capacity, hydration number, and proton conductivity as a function of the degree of grafting. The thermal properties (melting and glass transition temperatures) and thermal stability of the membrane were also investigated using differential scanning calorimetry and thermal gravimetric analysis, respectively. Membranes having degrees of grafting of 16% and above showed proton conductivity of the magnitude of 10−2 Ω−1 cm−1 at room temperature, as well as thermal stability at up to 290°C under an oxygen atmosphere. © 2000 John Wiley & Sons, Inc. The physical and chemical properties of polystyrene grafted and sulfonated polytetrafluoroethylene (PTFE‐graft‐PSSA) membranes prepared by radiation‐induced grafting of styrene onto commercial PTFE films using simultaneous irradiation technique followed by a sulfonation reaction are evaluated. The investigated properties include water uptake, ion exchange capacity, hydration number and ionic conductivity. All properties are correlated with the amount of grafted polystyrene (degree of grafting). The thermal stability of the membrane evaluated by thermal gravimetric analysis (TGA) is compared with that of original and grafted PTFE films. The membrane surface structural properties are analysed by electron spectroscopy for chemical analysis (ESCA). Membranes having degrees of grafting of 18 % and above show a good combination of physical and chemical properties that allow them to be proposed for use as proton conducting membranes, provided that they have sufficient chemical and mechanical stability. © 2000 Society of Chemical Industry 相似文献

This work addresses the problem of synthesising cost-effective batch water networks where a number of process sources along with fresh water are mixed, stored, and assigned to process sinks. In order to address the complexity of the problem, a three-stage hierarchical approach is proposed. In the first stage, global targets are identified by formulating and solving a linear transportation problem for minimum water usage, maximum water recycle, and minimum wastewater discharge. These targets are determined a priori and without commitment to the network configuration. Next, a network with minimum number of tanks is synthesised by solving a mixed-integer non-linear program. The bilinear constraints are relaxed to transform the program into a mixed-integer linear program that is globally solvable. The third stage is aimed at simplifying the network configuration by minimising the number of network inter-connections. Insights gained from the water pinch analysis are also incorporated into the approach to further reduce the water flows via the placement of water regeneration unit. A case study is solved to illustrate the effectiveness of the proposed procedure. 相似文献

α/γ phase transformations occurring in Fe–10Cu–xNi alloys (0?≤?x?≤?15 in mass%) were studied using X-ray diffraction, scanning electron microscopy, electron back scattered diffraction, transmission electron microscopy and chemical analysis, combining X-ray microanalysis with energy dispersive spectrometry in the scanning electron microscope and electron microprobe analysis with wavelength dispersive spectrometry. The influence of cooling rate on the microstructure was investigated using ice-brine quenching and 2 °C/min slow cooling rate performed with dilatometry. Ni addition induces metastable transformations on cooling: massive and bainitic ferrite are formed depending on the alloy composition and cooling rate. Moreover, most of the Cu phase precipitates on cooling giving rise to a fine distribution of Cu particles in the ferrite grains. For both cooling conditions, the hardness increases with increasing Ni content and a higher hardness is obtained in the quenched alloy for each composition. The change in hardness is correlated to the effect of Ni solid solution, transformation structure and size of Cu particles. 相似文献

Midazolam (MDZ) could affect lymphocyte immune functions. However, the influence of MDZ on cell’s K+ currents has never been investigated. Thus, in the present study, the effects of MDZ on Jurkat T lymphocytes were studied using the patch-clamp technique. Results showed that MDZ suppressed the amplitude of delayed-rectifier K+ current (IK(DR)) in concentration-, time-, and state-dependent manners. The IC50 for MDZ-mediated reduction of IK(DR) density was 5.87 μM. Increasing MDZ concentration raised the rate of current-density inactivation and its inhibitory action on IK(DR) density was estimated with a dissociation constant of 5.14 μM. In addition, the inactivation curve of IK(DR) associated with MDZ was shifted to a hyperpolarized potential with no change on the slope factor. MDZ-induced inhibition of IK(DR) was not reversed by flumazenil. In addition, the activity of intermediate-conductance Ca2+-activated K+ (IKCa) channels was suppressed by MDZ. Furthermore, inhibition by MDZ on both IK(DR) and IKCa-channel activity appeared to be independent from GABAA receptors and affected immune-regulating cytokine expression in LPS/PMA-treated human T lymphocytes. In conclusion, MDZ suppressed current density of IK(DR) in concentration-, time-, and state-dependent manners in Jurkat T-lymphocytes and affected immune-regulating cytokine expression in LPS/PMA-treated human T lymphocytes. 相似文献
