Multiple sclerosis (MS) is an autoimmune inflammatory disease of the central nervous system that leads to the progressive disability of patients. A characteristic feature of the disease is the presence of focal demyelinating lesions accompanied by an inflammatory reaction. Interactions between autoreactive immune cells and glia cells are considered as a central mechanism underlying the pathology of MS. A glia-mediated inflammatory reaction followed by overproduction of free radicals and generation of glutamate-induced excitotoxicity promotes oligodendrocyte injury, contributing to demyelination and subsequent neurodegeneration. Activation of purinergic signaling, in particular P2X7 receptor-mediated signaling, in astrocytes and microglia is an important causative factor in these pathological processes. This review discusses the role of astroglial and microglial cells, and in particular glial P2X7 receptors, in inducing MS-related neuroinflammatory events, highlighting the importance of P2X7R-mediated molecular pathways in MS pathology and identifying these receptors as a potential therapeutic target. 相似文献

The aim of this work was to study the feasibility of detection of Trichinella spiralis in swine meat using Middle Infrared Spectroscopy Fourier Transform with Attenuated Total Reflectance (ATR) and Soft Independent Modeling of Class Analogy (MID-FTIR-ATR-SIMCA). Five male Pigs were orally infected at different larvae concentrations (13,000, 6500, 3500, 1625, 812 larvae/pig) and after 24 weeks the animals were euthanized. Five types of muscles were studied (leg, loin, rib, masseter, and diaphragm). Results showed that MID-FTIR-ATR-SIMCA was useful to determine the presence of T. spiralis in the samples, as the interclass distance between infected and non infected muscles varied from 13.5 to 36.8. This technique was also useful to discriminate among pig muscles, where masseter showed the largest interclass distance, while rib presented the smallest one. In all cases the recognition and rejection rates were 100%, which means that the methodology is capable of accurately separating T. spiralis infected from non infected swine meat. 相似文献

The turbine synchronization phenomenon is of great interest in order to estimate the flicker produced by a wind farm. This paper proposes an initial approach to analyze the appearance of this phenomenon by the use of various image processing techniques: a method to automatically calculate the angular frequency of an unknown number of wind turbines from a video. Prostaglandins are a group of lipids that produce diverse physiological and pathological effects. Among them, prostaglandin E2 (PGE2) stands out for the wide variety of functions in which it participates. To date, there is little information about the influence of PGE2 on gap junctional intercellular communication (GJIC) in any type of tissue, including epithelia. In this work, we set out to determine whether PGE2 influences GJIC in epithelial cells (MDCK cells). To this end, we performed dye (Lucifer yellow) transfer assays to compare GJIC of MDCK cells treated with PGE2 and untreated cells. Our results indicated that (1) PGE2 induces a statistically significant increase in GJIC from 100 nM and from 15 min after its addition to the medium, (2) such effect does not require the synthesis of new mRNA or proteins subunits but rather trafficking of subunits already synthesized, and (3) such effect is mediated by the E2 receptor, which, in turn, triggers a signaling pathway that includes activation of adenylyl cyclase and protein kinase A (PKA). β-Glucosidases are used in the food industry to hydrolyse glycosidic bonds in complex sugars, with enzymes sourced from extremophiles better able to tolerate the process conditions. In this work, a novel β-glycosidase from the acidophilic organism Alicyclobacillus herbarius was cloned and heterologously expressed in Escherichia coli BL21(DE3). AheGH1 was stable over a broad range of pH values (5–11) and temperatures (4–55 °C). The enzyme exhibited excellent tolerance to fructose and good tolerance to glucose, retaining 65 % activity in the presence of 10 % (w/v) glucose. It also tolerated organic solvents, some of which appeared to have a stimulating effect, in particular ethanol with a 1.7-fold increase in activity at 10 % (v/v). The enzyme was then applied for the cleavage of isoflavone from isoflavone glucosides in an ethanolic extract of soy flour, to produce soy isoflavones, which constitute a valuable food supplement, full conversion was achieved within 15 min at 30 °C. 相似文献

The morphology of a series of poly(butylene terephthalate) (PBT)/fatty acid dimer diol (FADD)-based copolyesters prepared by solid-state modification (SSM) was studied. It was shown that in copolyesters containing less than 10 wt% FADD two different phases, i.e. a PBT crystalline phase and a PBT-rich amorphous phase, are present. The FADD residues were more or less homogeneously distributed throughout the interlamellar regions. For copolymers containing more than 10 wt% of FADD, a three-phase morphological model has to be used due to phase separation of a FADD-rich amorphous phase from the PBT-rich matrix, as confirmed by solid-state nuclear magnetic resonance spectroscopy and transmission electron microscopy. The final morphology was dependent on the morphology of the PBT/FADD-based physical mixtures prior to SSM. In addition, it was shown that FADD diffusion during SSM influences the final morphology. 相似文献

The morphological and rheological properties of a commercial propylene‐ethylene copolymer (PEC) and a series of blends with different concentrations of poly (ethylene‐co‐propylene) are investigated. The blends are prepared mixing PEC with fractions obtained from it by solvent extraction. The phase structure of samples exposed to different thermal and mechanical histories was analyzed using scanning electron microscopy. The linear viscoelastic properties of the molten polymers were measured using different test sequences that include dynamic frequency and time sweeps. The phase structure of most blends changes dramatically with time when the polymers are kept in the molten state due to the coalescence of the domains. For example, the initial morphology of PEC which presents domains of ~1 μm diameter changes to regions of more than 10 μm of average diameter after 90 min at 178°C at rest. Coincidentally, the dynamic moduli of the blends change during annealing reaching values that depend on the mechanical history. For example, the elastic modulus of PEC increases ~32% during a dynamic time sweep of 45 min using a frequency of 0.1 s?1, while it decreases ~18% when a frequency of 1 s?1 is applied. Moreover, the modulus measured at 0.1 s?1 of samples annealed at rest during 45 min is ~58% larger than that of the fresh material. POLYM. ENG. Curcumin is a natural product possessing therapeutic properties but the low water solubility of this compound limits its use. We have successfully incorporated curcumin into a bilayer of dodecanoic acid attached to magnetite nanoparticles in an effort to maximize solubility and delivery efficiency. Curcumin/magnetite nanoparticles were characterized using diffused reflectance infra-red fourier transform spectroscopy (DRIFTS) and X-ray powder diffraction (XRD). Moreover curcumin associated magnetite nanoparticles inhibited in vitro melanoma cell growth. An inhibitory concentration (IC50) of 66.0 +/- 3.0 microM (48 +/- 2.2 microg-iron/mL) was observed for the curcumin/magnetite nanoparticles. Fluorescent microscopy revealed that curcumin associated magnetite nanoparticles were internalized by the melanoma cells and remained in the cytoplasm. The curcumin/magnetic nanoparticles synthesized in this study possess magnetic and water solubility properties making this a novel curcumin formulation with therapeutic potential. 相似文献

This paper describes the performance of a full‐scale common effluent plant treating tannery wastewater, based on a single‐sludge nitrification/denitrification process, with the objective of investigating the reasons for its instability. The plant was monitored over a 1.5‐year period; operational parameters were evaluated daily and their impact on process rates assessed together with the characteristics of the wastewater sampled at different points in the treatment scheme. The results showed that instability problems and a periodic failure of nitrification/denitrification occurred due to a series of reasons, often persisting simultaneously and having an additive synergetic effect. Factors whose magnitude would be insufficient to affect the performance of a plant treating domestic wastewater, eg; small deviations from the optimal pH in nitrification or denitrification basins, a temperature decrease to 17 °C or an increase of the influent nitrogen content, caused the plant's efficiency to drop from 100% to 40% in some periods. To ensure stable performance, the dilution rate, D, along with the sludge retention time were shown to be key parameters for the control of the process. Maintaining values of D considerably lower than critical theoretical values calculated from the specific growth rate of nitrifiers at a given temperature and sludge recycle ratio may prevent problems in the plant's performance, particularly when high fluctuations of the concentration of inflow solids make calculation of the sludge age uncertain and proper sludge management difficult.
