Four experiments explored the task span procedure: Subjects received lists of 1-10 task names to remember and then lists of 1-10 stimuli on which to perform the tasks. Task span is the number of tasks performed in order perfectly. Experiment 1 compared the task span with the traditional memory span in 6 practiced subjects and found little difference. Experiment 2 compared the task span and the memory span in 64 unpracticed subjects and also found little difference. Experiment 3 compared practice with consistent and varied lists to address retrieval from long-term memory. Experiment 4 manipulated the number of task switches and found that it had little effect on task spans. The results suggest there is no trade-off between storage and task switching, which supports some theories of executive control and challenges others. In a previous study, we found that administration of ILB®, a new low molecular weight dextran sulphate, significantly improved mitochondrial functions and energy metabolism, as well as decreased oxidative/nitrosative stress, of brain tissue of rats exposed to severe traumatic brain injury (sTBI), induced by the closed-head weight-drop model of diffused TBI. Using aliquots of deproteinized brain tissue of the same animals of this former study, we here determined the concentrations of 24 amino acids of control rats, untreated sTBI rats (sacrificed at 2 and 7 days post-injury) and sTBI rats receiving a subcutaneous ILB® administration (at the dose levels of 1, 5 and 15 mg/kg b.w.) 30 min post-impact (sacrificed at 2 and 7 days post-injury). Additionally, in a different set of experiments, new groups of control rats, untreated sTBI rats and ILB®-treated rats (administered 30 min after sTBI at the dose levels of 1 or 5 mg/kg b.w.) were studied for their neurocognitive functions (anxiety, locomotor capacities, short- and long-term memory) at 7 days after the induction of sTBI. Compared to untreated sTBI animals, ILB® significantly decreased whole brain glutamate (normalizing the glutamate/glutamine ratio), glycine, serine and γ-aminobutyric acid. Furthermore, ILB® administration restored arginine metabolism (preventing nitrosative stress), levels of amino acids involved in methylation reactions (methionine, L-cystathionine, S-adenosylhomocysteine), and N-acetylaspartate homeostasis. The macroscopic evidences of the beneficial effects on brain metabolism induced by ILB® were the relevant improvement in neurocognitive functions of the group of animals treated with ILB® 5 mg/kg b.w., compared to the marked cognitive decline measured in untreated sTBI animals. These results demonstrate that ILB® administration 30 min after sTBI prevents glutamate excitotoxicity and normalizes levels of amino acids involved in crucial brain metabolic functions. The ameliorations of amino acid metabolism, mitochondrial functions and energy metabolism in ILB®-treated rats exposed to sTBI produced significant improvement in neurocognitive functions, reinforcing the concept that ILB® is a new effective therapeutic tool for the treatment of sTBI, worth being tested in the clinical setting.  相似文献

Obstructive sleep apnea (OSA), a disease associated with excessive sleepiness and increased cardiovascular risk, affects an estimated 1 billion people worldwide. The present study examined proteomic biomarkers indicative of presence, severity, and treatment response in OSA. Participants (n = 1391) of the Stanford Technology Analytics and Genomics in Sleep study had blood collected and completed an overnight polysomnography for scoring the apnea–hypopnea index (AHI). A highly multiplexed aptamer-based array (SomaScan) was used to quantify 5000 proteins in all plasma samples. Two separate intervention-based cohorts with sleep apnea (n = 41) provided samples pre- and post-continuous/positive airway pressure (CPAP/PAP). Multivariate analyses identified 84 proteins (47 positively, 37 negatively) associated with AHI after correction for multiple testing. Of the top 15 features from a machine learning classifier for AHI ≥ 15 vs. AHI < 15 (Area Under the Curve (AUC) = 0.74), 8 were significant markers of both AHI and OSA from multivariate analyses. Exploration of pre- and post-intervention analysis identified 5 of the 84 proteins to be significantly decreased following CPAP/PAP treatment, with pathways involving endothelial function, blood coagulation, and inflammatory response. The present study identified PAI-1, tPA, and sE-Selectin as key biomarkers and suggests that endothelial dysfunction and increased coagulopathy are important consequences of OSA, which may explain the association with cardiovascular disease and stroke.  相似文献

We describe methods for continual prediction of manufactured product quality prior to final testing. In our most expansive modeling approach, an estimated final characteristic of a product is updated after each manufacturing operation. Our initial application is for the manufacture of microprocessors, and we predict final microprocessor speed. Using these predictions, early corrective manufacturing actions may be taken to increase the speed of expected slow wafers (a collection of microprocessors) or reduce the speed of fast wafers. Such predictions may also be used to initiate corrective supply chain management actions. Developing statistical learning models for this task has many complicating factors: (a) a temporally unstable population (b) missing data that is a result of sparsely sampled measurements and (c) relatively few available measurements prior to corrective action opportunities. In a real manufacturing pilot application, our automated models selected 125 fast wafers in real-time. As predicted, those wafers were significantly faster than average. During manufacture, downstream corrective processing restored 25 nominally unacceptable wafers to normal operation.  相似文献

A facility for testing SiC fiber tows in static fatigue and creep at elevated temperatures in air and steam was developed. Static fatigue of Hi-Nicalon™-S fibers was investigated at 800°C-1100°C at applied stresses between 115 and 1250 MPa in air, in Si(OH)4(g)-saturated steam, and in unsaturated steam. Fibers tested in Si(OH)4(g)-saturated steam and in air had silica scales throughout the test sections, but those tested in unsaturated steam did not develop scales near the steam injection point. Fiber lifetimes in static fatigue were shortest in unsaturated steam, intermediate in Si(OH)4(g)-saturated steam, and longest in air. Failure strains did not exceed 0.3%. Steady-state strain rates and static fatigue lifetimes are modelled empirically by the Monkman-Grant relationship. Failure mechanisms are discussed.  相似文献

A microbial fuel cell (MFC) is a device that converts organic matter to electricity using microorganisms as the biocatalyst. Most MFCs contain two electrodes separated into one or two chambers that are operated as a completely mixed reactor. In this study, a flat plate MFC (FPMFC) was designed to operate as a plug flow reactor (no mixing) using a combined electrode/proton exchange membrane (PEM) system. The reactor consisted of a single channel formed between two nonconductive plates that were separated into two halves by the electrode/PEM assembly. Each electrode was placed on an opposite side of the PEM, with the anode facing the chamber containing the liquid phase and the cathode facing a chamber containing only air. Electricity generation using the FPMFC was examined by continuously feeding a solution containing wastewater, or a specific substrate, into the anode chamber. The system was initially acclimated for 1 month using domestic wastewater orwastewater enriched with a specific substrate such as acetate. Average power density using only domestic wastewater was 72+/-1 mW/m2 at a liquid flow rate of 0.39 mL/min [42% COD (chemical oxygen demand) removal, 1.1 h HRT (hydraulic retention time)]. At a longer HRT = 4.0 h, there was 79% COD removal and an average power density of 43+/-1 mW/m2. Power output was found to be a function of wastewater strength according to a Monod-type relationship, with a half-saturation constant of Ks = 461 or 719 mg COD/L. Power generation was sustained at high rates with several organic substrates (all at approximately 1000 mg COD/L), including glucose (212+/-2 mW/ m2), acetate (286+/-3 mW/m2), butyrate (220+/-1 mW/ m2), dextran (150+/-1 mW/m2), and starch (242+/-3 mW/ m2). These results demonstrate the versatility of power generation in a MFC with a variety of organic substrates and show that power can be generated at a high rate in a continuous flow reactor system.  相似文献

There is great interest in hydrogen evolution in bioelectrochemical systems, such as microbial electrolysis cells (MECs), but these systems require non-optimal near-neutral pH conditions and the use of low-cost, non-precious metal catalysts. Here we show that molybdenum disulfide (MoS2) composite cathodes have electrochemical performance superior to stainless steel (SS) (currently the most promising low-cost, non-precious metal MEC catalyst) or Pt-based cathodes in phosphate or perchlorate electrolytes, yet they cost ∼4.5 times less than Pt-based composite cathodes. At current densities typical of many MECs (2-5 A/m2), the optimal surface density with MoS2 particles on carbon cloth was 25 g/m2, achieving 31 mV less hydrogen evolution overpotential than similarly constructed Pt cathodes in galvanostatic tests with a phosphate buffer. At higher current densities (8-10 A/m2) the MoS2 catalyst had 82 mV less hydrogen evolution overpotential than the Pt-based catalyst. MoS2 composite cathodes performed similarly to Pt cathodes in terms of current densities, hydrogen production rates and COD removal over several batch cycles in MEC reactors. These results show that MoS2 can be used to substantially reduce the cost of cathodes used in MECs for hydrogen gas production.  相似文献

Proactive adjustments of response strategies in the stop-signal paradigm.   总被引:2,自引:0,他引:2  
In the stop-signal paradigm, fast responses are harder to inhibit than slow responses, so subjects must balance speed in the go task with successful stopping in the stop task. In theory, subjects achieve this balance by adjusting response thresholds for the go task, making proactive adjustments in response to instructions that indicate that relevant stop signals are likely to occur. The 5 experiments reported here tested this theoretical claim, presenting cues that indicated whether or not stop signals were relevant for the next few trials. Subjects made proactive response-strategy adjustments in each experiment: Diffusion-model fits showed that response threshold increased when participants expected stop signals to occur, slowing go responses and increasing accuracy. Furthermore, the results show that subjects can make proactive response-strategy adjustments on a trial-by-trial basis, suggesting a flexible cognitive system that can proactively adjust itself in changing environments.
