Chromosomal translocations of bcl-3 are associated with chronic B cell lymphocytic leukemias. Previously, we have shown that Bcl-3, a distinct member of the I kappa B family, may function as a positive regulator of NF-kappa B activity, although its physiologic roles remained unknown. To uncover these roles, we generated Bcl-3-deficient mice. Mutant mice, but not their littermate controls, succumb to T. gondii owing to failure to mount a protective T helper 1 immune response. Bcl-3-deficient mice are also impaired in germinal center reactions and T-dependent antibody responses to influenza virus. The results reveal critical roles for Bcl-3 in antigen-specific priming of T and B cells. Altered microarchitecture of secondary lymphoid organs in mutant mice, including partial loss of B cells, may underlie the immunologic defects. The implied role of Bcl-3 in maintaining B cells in wild-type mice may related to its oncogenic potential.  相似文献

The measurement of tumour cell proliferation is becoming increasingly recognised in defining prognostic groups. Proliferating cell nuclear antigen (PCNA) immunolocalisation can be used as an index of cell proliferation and may define the extent of departure from normal growth control. The monoclonal antibody PC10 stains PCNA in archival paraffin-embedded tissue. This study investigates its potential as a prognostic marker in early and advanced ovarian cancer. A three-stage immunoperoxidase technique was developed to detect the monoclonal antibody PC10. Archival paraffin-embedded tissue from 19 stage I ovarian tumours (13 malignant and six borderline) and 79 advanced (stage IIb-IV) ovarian tumours (patients entered into the Third North-West Thames Ovarian Cancer Trial) was immunostained with PC10. PC10 immunostaining was performed successfully in 91.8% of cases. The PC10 labelling index (PC10 LI) ranged from 1.5% to 88% with a mean value of 47.4%. Stage I borderline tumours had significantly lower PCNA labelling indexes than stage I malignant tumours (P < 0.048). In advanced disease there was an inverse correlation between PC10 and overall survival, and in those patients who underwent good debulking surgery (37 patients with disease < 2 cm diameter) a low PC10 value (< 36.5%) correlated with improved survival (log-rank trend test for survival, chi 2 = 5.75, P = 0.017). PCNA immunostaining defines a good prognostic subgroup in adequately debulked patients with ovarian cancer.  相似文献

In this paper, the sedimentology, petrography and biostratigraphy of the Nam Phong Formation and the overlying Mesozoic Khorat Group in NE Thailand are described. Data were gathered during extensive studies in the field (about 100 outcrop localities were investigated) and from laboratory analyses of over 500 surface samples.
The Nam Phong Formation is dated accurately for the first time as late Norian to Rhaetian, while the Phra Wihan, Sao Khua, Phu Phan and Khok Kruat Formations (of the Khorat Group) are reassigned to the Early Cretaceous (Berriasian to Aptian) on the basis of new palynological data. The age of the intervening Phu Kradung Formation, which is conventionally assigned to the Lower Jurassic, is probably Late Jurassic or Early Cretaceous, although this cannot be determined with confidence. Changes in provenance between the Nam Phong Formation and the overlying Phu Kradung Formation suggest a possible sedimentary hiatus. Palaeocurrent data suggest that sediments were mainly sourced from the north and east.

Molded case circuit breakers have proven to be quite versatile, with increasing capabilities being designed into new products. Interrupting capacity and adjustability of tripping times and magnitude are constantly being improved. This proliferation has developed so widely that applications may appear much simpler than actual and may be marginal even if executed in accordance with certified tests and standards. Examples of less publicized facts about the application of molded case circuit breakers are described. The possibility of the user having unsafe results is inescapable. A conclusion is reached that issues of safety should receive more open discussions so that standards can be improved. It also appears that users have not been advised of the risks involved in order to enable an economic justification study to be made. A suggestion is made that the IEEE develop an intensive educational program which would result in certification of engineers involved with protection.  相似文献

Ultrasonic cavitation was employed to enhance sensitivity of bacterial spore immunoassay detection, specifically, enzyme-linked immunosorbent assay (ELISA) and resonant mirror (RM) sensing. Bacillus spore suspensions were exposed to high-power ultrasound in a tubular sonicator operated at 267 kHz in both batch and flow modes. The sonicator was designed to deliver high output power and is in a form that can be cooled efficiently to avoid thermal denaturation of antigen. The 30-s batch and cooled flow (0.3 mL/min) sonication achieved an approximately 20-fold increase in ELISA sensitivity compared to unsonicated spores by ELISA. RM sensing of sonicated spores achieved detection sensitivity of approximately 10(6) spores/mL, whereas unsonicated spores were undetectable at the highest concentration tested. Improvements in detection were associated with antigen released from the spores. Equilibrium temperature increase in the tubular sonicator was limited to 14 K after 30 min and was maintained for 6 h with cooling and flow (0.3 mL/min). The work described here demonstrates the utility of the tubular sonicator for the improvement in the sensitivity of the detection of spores and its suitability as an in-line component of a rapid detection system.  相似文献

We show that gratings can be designed to be symmetry selective, that is, reflecting modes with a particular symmetry. The idea behind a symmetry-selective grating is to replace a grating written over the entire core cross section of a waveguide with a grating that is written only over a part of the core. This new kind of grating exhibits high-order reflectivity and selectivity in comparison with standard gratings and enables the design of more effective and compact wavelength add/drop devices.  相似文献
