Bacteria of the genus Pseudomonas have been associated with the spoilage of raw milk and dairy products due to the production of thermostable proteolytic enzymes. The apr gene encodes for alkaline metalloprotease in Pseudomonas and other related bacteria. Its presence in psychrotrophic proteolytic bacteria isolated from raw milk collected from cooling tanks was verified. A polymerase chain reaction (PCR) technique was used with degenerate primers. Total DNA from 112 isolates was pooled in different groups and then used as template for the amplification reactions. Controls consisted of DNA extracted from 26 cultures. An expected DNA fragment of 194 bp was detected in groups that contained bacteria identified as Pseudomonas. The PCR product was observed only when DNA from control cultures of Pseudomonas aeruginosa, Pseudomonas fluorescens, Serratia marcescens and Aeromonas hydrophila were used. A detection limit assay indicated that the apr gene could be directly amplified from pasteurized milk inoculated with 10(8) CFU/ml of P. fluorescens. With this method it was possible to detect proteolytic bacteria at 10(5) CFU/ml in reconstituted skim milk powder if cells were recovered for DNA extraction before amplification.  相似文献

This study investigates the performance of an adsorption chiller driven by thermal heat collected from solar collectors’ panels with heat storage. The heat is reserved in a storage tank and the reserved heat is used to drive the adsorption chiller. The investigation was carried on the climatic conditions of Dhaka, Bangladesh. Heat transfer fluid goes from the collectors to the adsorption cooling unit, then from the adsorption cooling unit to the storage tank. It is seen that heat storage is more effective than direct solar coupling; however, it requires more collectors, depending on the size of the storage tank. The analysis shows that cycle time is one of the most influential parameters for the solar-driven adsorption cooling system. It is seen that the size of the collector can be reduced if the proper cycle time is adjusted. The analysis also revealed that the system with 22 collectors (each of 2.415 m2) along with 1000 s cycle time provides better performance for the base run conditions. It is also seen that the solar-driven adsorption chiller with heat storage works well beyond the sunset time.  相似文献

Cheeses are able to serve as suitable matrices for supplying probiotics to consumers, enabling appropriate conditions for bacteria to survive gastric transit and reach the gut, where they are assumed to promote beneficial processes. The present study aimed to evaluate the microbiological, immunological, and histological changes in the gut of Salmonella Enteritidis-challenged rats fed goat cheese supplemented with the probiotic strain Lactobacillus rhamnosus EM1107. Thirty male albino Wistar rats were randomly distributed into 5 experimental groups with 6 animals each: negative (NC) and positive (PtC) control groups, control goat cheese (CCh), goat cheese added with L. rhamnosus EM1107 (LrCh), and L. rhamnosus EM1107 only (EM1107). All animals, except NC group were challenged with Salmonella Enteritidis (109 cfu in 1 mL of saline through oral gavage). Microbial composition was assessed with high-throughput 16S rRNA sequencing by means of Illumina MiSeq (Illumina, San Diego, CA). Nuclear factor kappa B (NF-κB) from the animal cecum tissue was determined by real-time PCR and interleukins (TNF-α, IL-1β, IL-10, and IFN-γ) by means of ELISA. Myeloperoxidase and malondialdehyde levels were determined biochemically. The administration of the L. rhamnosus EM1107 probiotic strain, either as a pure culture or added to a cheese matrix, was able to reduce Salmonella colonization in the intestinal lumen and lessen tissue damage compared with rats from PtC group. In addition, the use of cheese for the probiotic strain delivery (LrCh) was associated with a marked shift in the gut microbiota composition toward the increase of beneficial organisms such as Blautia and Lactobacillus and a reduction in NF-κB expression. These findings support our hypothesis that cheeses might be explored as functional matrices for the efficacious delivery of probiotic strains to consumers.  相似文献

Lamiaceae is among the largest families of flowering plants with about 250 genera and over 7,000 species distributed around the world. It is considered as the important source of essential oils, for example, menthol, geraniol, eucalyptol, camphor and thymol. Therefore, it is imperative to study these economically important compounds under in vitro conditions for their sustainable and enhanced production. In addition to proven biological activities, essential oils from this family have recently been evaluated for anticancer activities and considered as a source of anticancer drugs. Mechanisms involved in the essential oils-mediated antiproliferative activity include cell cycle arrest, apoptosis and DNA repair mechanisms. Essential oils also act in the reduction of tumors, inhibiting metastasis and as anti-multidrug resistance molecules. The aim of this review is to assess the anticancer properties of essential oils obtained from different members of family Lamiaceae. The available reports on active components of essential oils and their effect on cancer type and cell line have been discussed. Biotechnological studies to improve the production of essential oils have also been highlighted. Various methods have been adopted to obtain essential oils under in vitro conditions from different plant species of family Lamiaceae, and their production is affected by culture conditions, cultivation mode, utilization of nutrient media and plant growth regulators. The literature survey suggests that essential oils obtained from family Lamiaceae have perspective for the development of new alternatives for disease treatment and prevention.  相似文献

This study shows the influence of the Pt percentage in Pt catalysts (Pt/C) on their application in Direct Glycerol Fuel Cells (DGFC). Catalysts with 20, 30, 40 and 60% Pt were prepared by formic acid reduction. X-Ray Diffraction (XRD) confirmed the formation of Pt fcc nanocrystals (with average sizes between 3 and 4 nm). Thermogravimetric analysis (TGA) corroborated the Pt loadings and the Transmission Electron Microscopy (TEM) images show a homogeneous distribution of the Pt nanoparticles, with larger particles at higher Pt percentages. Electrochemical measurements reveal that higher Pt percentages promote the activity of the glycerol electroxidation reaction, resulting in lower onset potentials, higher current densities and reduced poisoning of the Pt surface. Single cell tests confirm these results, with greater maximum power density at higher Pt percentages, although the glycerol crossover effect starts to become significant in the 60% Pt/C catalyst, owing to the reduced thickness of this catalytic layer. A final product selectivity analysis indicates that tartronate is the preferential glycerol electroxidation product, along with glycerate and oxalate in lower proportions, with minor amounts of glycolate and mesoxalate. In general, a thicker catalytic layer is associated with the formation of more oxidized products.  相似文献

BACKGROUND: Nowadays, organoleptic quality is the primary objective for almost all tomato breeding programmes. In this study, post‐harvest behaviour of a breeding line with genetic resistance to important viruses (tomato mosaic virus, tomato spotted wilt virus and tomato yellow leaf curl virus) has been compared with the original traditional landrace (Muchamiel). The breeding line has been obtained by backcrossing, introgressing three resistance genes but aiming to keep the quality characteristics of the traditional variety. Tomatoes were picked at random and stored at 10 °C for 13 days. Quality analyses were made in both tomato samples: weight loss, colour, respiration rate, ethylene production, maturity index, instrumental hardness and sensory evaluation with trained panel. RESULTS: Fruits of the breeding line were characterized by higher hardness even with a higher maturity index. Results of sensory tests were in agreement with instrumental measurements. Organoleptic quality of Muchamiel virus‐resistant tomatoes was at least as high as that of traditional tomatoes, reaching the best scores in odour and aroma at the 13th storage day. CONCLUSION: Although a long time has been required to develop the breeding line, results indicate that organoleptic fruit quality has been recovered through backcrossing, confirming the success of the breeding programme. Bacteria and other microorganisms have a natural tendency to adhere to surfaces as a survival mechanism. This can occur in many environments, including the living host, industrial systems, and natural waters. The general outcome of bacterial colonization of a surface is biofilm formation, which consists of microorganisms immobilized in a variety of polymeric compounds generally referred to as extracellular polymeric substances. Bacterial adhesion to a solid surface is a crucial step in the biofilm process. This step is dependent upon van der Waals, electrostatic, and acid–base interactions. These interactions are influenced by physicochemical properties of the substratum and the bacterial surface, such as hydrophobicity, surface charge, and electron donor–electron acceptor properties. In addition, the roughness of the substratum and the microbiological characteristics of the cell surface, such as cellular appendages and production of exopolysaccharides, can affect the adherence process. To date, many strategies have been developed to decrease the adherence of bacteria to surfaces. Surface modification with the addition of the suitable compounds makes surfaces less attractive for microorganisms and therefore prevents bacterial adherence and biofilm formation.  相似文献

Abstract: Because sorghum grain has important characteristics (for example, great antioxidant profile) for introduction into the regular human diet, producers and industry aim to develop successful products using sorghum as the base. The 1st step in developing products attractive for consumers is understanding consumer needs and expectations. Conjoint analysis determines attributes which may be important or unimportant for one product. In this study, focus groups and conjoint analysis were used to establish which attributes are helpful for promoting purchase intent of sorghum grain products. Once those characteristics were chosen, a national survey was conducted to confirm the selection. During the focus groups, the heath aspects of grain products seemed to be the most appealing for consumers, whereas conjoint analysis showed that sensory attributes were the principal drivers for purchase intent. Practical Application: The data presented in this study are essential for industry as a starting point for developing sorghum grain based food products. It is necessary to address consumers’ demands to ensure new products’ success in the market, and the present study clearly shows this consumers’ input.  相似文献
