Methanol extracts of olive pomace (two‐phase olive oil extraction) and olive pulp were analysed by reverse phase HPLC and the eluted fractions were characterised by electrospray ionisation mass spectrometry. This technique allowed the identification of some common phenolic compounds, namely, verbascoside, rutin, caffeoyl‐quinic acid, luteolin‐4‐glucoside and 11‐methyl‐oleoside. Hydroxytyrosol‐1′‐β‐glucoside, luteolin‐7‐rutinoside and oleoside were also detected. Moreover, this technique enabled the identification, for the first time in Olea europaea tissues, of two oleoside derivatives, 6′‐β‐glucopyranosyl‐oleoside and 6′‐β‐rhamnopyranosyl‐oleoside, and of 10‐hydroxy‐oleuropein. Also, an oleuropein glucoside that had previously been identified in olive leaves was now detected in olive fruit, both in olive pulp and olive pomace. With the exception of oleoside and oleuropein, the majority of phenolic compounds were found to occur in equivalent amounts in olive pulp and olive pomace. Oleoside was the main phenolic compound in olive pulp (31.6 mg g?1) but was reduced to 3.6 mg g?1 in olive pomace, and oleuropein (2.7 mg g?1 in the pulp) almost disappeared (<0.1 mg g?1 in the pomace). The volatile compositions of 192 olive oil samples from five different European countries were investigated by PTR-MS sample headspace analysis. The mass spectra of all samples showed many masses with high abundances, indicating the complex VOC composition of olive oil. Three different PLS-DA models were fitted to the data to classify samples into ‘country’, ‘region’ and ‘district’ of origin, respectively. Correct classification rates were assessed by cross-validation. The first fitted model produced an 86% success rate in classifying the samples into their country of origin. The second model, which was fitted to the Italian oils only, also demonstrated satisfactory results, with 74% of samples successfully classified into region of origin. The third model, classifying the Italian samples into district of origin, yielded a success rate of only 52%. This lower success rate might be due to either the small class set, or to genuine similarities between olive oil VOC compositions on this tight scale.  相似文献

The freeze-dried sprouts’ juice of Raphanus sativus (L.) cv. Sango was prepared and analysed for the first time. HPLC analysis of total isothiocyanates, after protein displacement, resulted in 77.8 ± 3.0 μmol/g of dry juice while GC–MS analysis of hexane and acetone extracts showed E- and Z-raphasatin (8.9 and 0.11 μmol/g, respectively) and sulforaphene (11.7 μmol/g), summing up to 20.7 ± 1.7 μmol/g of free isothiocyanates. Sprouts’ juice contained an unprecedented wealth of anthocyanins and a new fractionation methodology allowed us to isolate 34 mg/g of acylated anthocyanins (28.3 ± 1.9 μmol/g), belonging selectively to the cyanidin family. Analysis was performed by HPLC–PDA–ESI–MSn and extended to deacylated anthocyanins and aglycones, obtained, respectively, by alkaline and acid hydrolysis. This study identified 70 anthocyanins, 19 of which have never been described before and 32 of which are reported here in R. sativus for the first time. Sango radish sprouts are exceptional dietary sources of heath-promoting micronutrients.  相似文献

The objective of this study was the quality loss inhibition of hake (Merluccius merluccius) during the frozen storage. For it, the effect of a previous high-pressure (HP) treatment (150–450 MPa for 2 min) was analysed throughout a 5-month storage at ? 10 °C. Quality changes were monitored by complementary chemical analyses. Inhibition (p < 0.05) of dimethylamine (DMA), free fatty acid (FFA), formaldehyde (FA), trimethylamine, total volatile amine and fluorescent compound (tertiary lipid oxidation compound) formation was concluded by previous pressure treatment according to the one-way ANOVA analysis. On the contrary, no effect (p > 0.05) on the K value, polyene index and formation of peroxides and thiobarbituric acid reactive substances was achieved. Additionally, a multifactor ANOVA test (pressure and frozen storage time effects; i.e. comparison among HP treatments) showed an inhibitory effect (p < 0.015) on DMA and FFA formation, this effect increasing with pressure level applied. This inhibitory effect on the formation of such molecules related to quality loss can be explained on the basis of the damage caused to different kinds of enzymes such as trimethylamine oxide demethylase, lipases and phospholipases, so that their activity during the subsequent frozen storage would decrease. The work here presented provides for the first time information concerning the employment of HP technology to inhibit the DMA, FA and FFA formation during the frozen storage of hake. Further research focussed on commercial frozen conditions (? 18 °C) and including sensory and nutritional aspects is foreseen.  相似文献

Sorghum straw is a renewable, cheap and widespread resource. The acid hydrolysis of sorghum straw to obtain xylose solutions could be a good alternative for this abundant resource. The H2SO4 hydrolysis of sorghum straw at two different temperatures (80 and 100 °C) and three H2SO4 concentrations (2, 4 and 6%) using a solid/liquid ratio of 1:10 (w/w) was studied. Kinetic parameters of mathematical models for predicting the concentrations of xylose, glucose, acetic acid and furfural were determined. The activation energy of the release reaction was 183.3 kJ mol?1 for xylose and 185.8 kJ mol?1 for glucose. The optimal conditions found were 6% H2SO4 at 100 °C for 60 min, which allow one to obtain a solution with 18.27 g xylose l?1, 6.78 g glucose l?1, 0.7 g furfural l?1 and 1.35 g acetic acid l?1. It is concluded that this process has potential for utilisation of this renewable lignocellulosic resource. © 2002 Society of Chemical Industry  相似文献

During the North Atlantic marine boundary layer experiment (NAMBLEX) sampling campaign at Mace Head, Ireland, both continental and maritime air masses were sampled. Aerosol was characterized both with a TSI 3800 time-of-flight mass spectrometer (ATOFMS) and a MOUDI microorifice impactor, and particle number counts were measured independently with an aerodynamic particle sizer. The data have been analyzed in order to elucidate factors determining the particle detection efficiencies of the ATOFMS. These are broken down according to the efficiency of the inlet system, the hit efficiency on particles which enter the sensing zone of the instrument and the sensitivity of the measured ion signal to the chemical species. A substantial matrix effect depending on the chemical composition of the aerosol sampled at the time was found, which is reflected in variations in the hit efficiency of particles entering the sensing zone of the instrument with the main desorption-ionization laser. This is in addition to the strong inverse power-law dependence of inlet transmission efficiency on particle diameter. The variation in hit efficiency with particle type is likely attributable to differences in the energetics of laser energy absorption, ablation, and ion formation. However, once variations in both inlet transmission and hit efficiencies are taken into account, no additional matrix dependence of ATOFMS response is required to obtain a linear relationship between the ion signal and the concentration of a particular chemical species. The observations show that a constant mass of material is ionized from each particle, irrespective of size. Consequently the integrated ion signal for a given chemical component and particle size class needs to be increased by a factor related to the cube of particle diameter in order to correlate with the airborne mass of that component.  相似文献

Consumption of Brassica vegetables containing glucosinolates is associated with decreased risk of cancer at several tissue sites. Studies of the relative potency and metabolism of the major dietary isothiocyanates have been hindered by limited availability of high‐purity isothiocyanates and their glucosinolate precursors. We describe a simple procedure for the preparation of glucosinolates sinigrin, gluconapin, progoitrin, glucoiberin, glucoraphanin, glucoalyssin and gluconasturtiin from vegetable seeds. Glucosinolates were extracted from the seeds of mustard, oilseed, broccoli, cauliflower and winter cress. Purification by preparative anion exchange and reversed‐phase column chromatography gave the pure glucosinolates. Good separation was achieved without using ion‐pair reagents. Related isothiocyanates and amine degradation products were prepared by myrosinase‐catalysed hydrolysis of the corresponding glucosinolate and by organic synthesis, respectively. The compounds were characterized by analytical HPLC, 1H NMR, 13C NMR, elemental analysis and mass spectrometry. Optimized conditions for detection and quantitation of these glucosinolates and metabolites by LC‐MS/MS are also given. Large‐scale glucosinolate preparation could be performed, giving amounts suitable for pharmacological evaluation in vivo.
