China's continuous and rapid economic growth has led to the reclamation of large sections of the intertidal mud coast in combination with port construction,such as that of the proposed Tongzhou Bay port on the Jiangsu coast.These reclamations threaten the local ecosystem services.An ecotope distribution map was created and a hydrodynamic numerical model of Tongzhou Bay was set up to quantify the impacts of reclamation on the ecosystem.Based on the field data and model results,several abiotic features were classified into 11 ecotopes and visualized in an ecotope map of the Tongzhou Bay ecosystem.Validation with spatial distributions of two threatened shorebird species(bar-tailed godwit and great knot) showed confirmation with the mid-range and low-range littoral zones(inundated from 40% to 100% of a tidal cycle),indicating the importance of the areas with these conditions to these populations.Overlaying the ecotope map with recent and proposed land reclamation schemes revealed a loss of ecotopes,composed of the high-range(42%),mid-range(48%),and low-range(38%) littoral habitats,corresponding to a 44%-45% loss of the most important ecotopes for bar-tailed godwit and great knot(mid-range and low-range littoral zones).These results confirm the applicability of the novel ecotope assessment approach in practice.  相似文献

Numerous studies in both rats and humans indicate the importance of the amygdala in the acquisition and expression of learned fear. The identification of the amygdala as an essential neural substrate for fear conditioning has permitted neurophysiological examinations of synaptic processes in the amygdala that may mediate fear conditioning. One candidate cellular mechanism for fear conditioning is long-term potentiation (LTP), an enduring increase in synaptic transmission induced by high-frequency stimulation of excitatory afferents. At present, the mechanisms underlying the induction and expression of amygdaloid LTP are only beginning to be understood, and probably involve both the N-methyl-D-aspartate (NMDA) and alpha-amino-3-hydroxy-5-methyl-4-isoxazolepropionate (AMPA) subclasses of glutamate receptors. This article will examine recent studies of synaptic transmission and plasticity in the amygdala in an effort to understand the relationships of these processes to aversive learning and memory.  相似文献

Cell cultures of Arabidopsis thaliana (A.t.) respond to changes in the gravitational field strength with fluctuations of the amount of cytosolic calcium (Ca2+). In parabolic flight experiments, where hyper- and μg phases follow each other, μg clearly increased Ca2+, while hyper-g caused a slight reduction. Since the latter observation had not been reported before, we studied this effect in more detail. Using a special centrifuge for heavy items (ZARM, Bremen, Germany), we determined the hyper-g-dependent intracellular Ca2+ level with transgenic cell lines expressing the Ca2+ sensor, cameleon. This sensor exhibits a shift in fluorescence from 480 to 530 nm in response to Ca2+ binding. The data show a drop in the intracellular Ca2+ concentration with a threshold gravity of around 3 g. This is above hypergravity levels achieved during parabolic flights (1.8 g). The use of mutants with different sub-cellular targets of cameleon expression (nucleus, tonoplast, plasma membrane) gave the same results, i.e. Ca2+ is obviously exported from several intracellular compartments.  相似文献

The rate of appearance (entry) of 22Na into the posterior chamber of the eye was determined in eight dogs under control conditions and again after pretreatment with acetazolamide, 50 mg/kg i.v. In comparison with the paired controls, pretreatment with acetazolamide decreased the average rate of entry of 22Na by 29% during the first 3 minutes after the intravenous administration of a tracer dose of the isotope. In separate experiments, the volumes of the chambers of the dog eye and the rate of aqueous humor formation were determined for the purpose of calculating nascent fluid ion concentrations. Comparisons of sodium data with the previously reported anion data are made. Results support the suggestion that reduction of intraocular pressure caused by acetazolamide is partially, if not wholly, the result of the action of the drug to reduce the secretion of aqueous humor.  相似文献

Cathepsin K-mediated thyroglobulin proteolysis contributes to thyroid hormone (TH) liberation, while TH transporters like Mct8 and Mct10 ensure TH release from thyroid follicles into the blood circulation. Thus, thyroid stimulating hormone (TSH) released upon TH demand binds to TSH receptors of thyrocytes, where it triggers Gαq-mediated short-term effects like cathepsin-mediated thyroglobulin utilization, and Gαs-mediated long-term signaling responses like thyroglobulin biosynthesis and thyrocyte proliferation. As reported recently, mice lacking Mct8 and Mct10 on a cathepsin K-deficient background exhibit excessive thyroglobulin proteolysis hinting towards altered TSH receptor signaling. Indeed, a combination of canonical basolateral and non-canonical vesicular TSH receptor localization was observed in Ctsk−/−/Mct8−/y/Mct10−/− mice, which implies prolonged Gαs-mediated signaling since endo-lysosomal down-regulation of the TSH receptor was not detected. Inspection of single knockout genotypes revealed that the TSH receptor localizes basolaterally in Ctsk−/− and Mct8−/y mice, whereas its localization is restricted to vesicles in Mct10−/− thyrocytes. The additional lack of cathepsin K reverses this effect, because Ctsk−/−/Mct10−/− mice display TSH receptors basolaterally, thereby indicating that cathepsin K and Mct10 contribute to TSH receptor homeostasis by maintaining its canonical localization in thyrocytes. Moreover, Mct10−/− mice displayed reduced numbers of dead thyrocytes, while their thyroid gland morphology was comparable to wild-type controls. In contrast, Mct8−/y, Mct8−/y/Mct10−/−, and Ctsk−/−/Mct8−/y/Mct10−/− mice showed enlarged thyroid follicles and increased cell death, indicating that Mct8 deficiency results in altered thyroid morphology. We conclude that vesicular TSH receptor localization does not result in different thyroid tissue architecture; however, Mct10 deficiency possibly modulates TSH receptor signaling for regulating thyrocyte survival.  相似文献

BACKGROUND: Carnosic acid and carnosol, the two major phenolic diterpenes present in rosemary and sage extracts, have received attention in food science and biomedicine because of their potent antioxidant properties. In plants, these compounds have been identified as being present in some species of the family Lamiaceae, but there is still little information about their distribution within the plant kingdom. The aim of the present study was to gain a better understanding of the occurrence of these compounds within leaf extracts of the genus Salvia and also to examine to what extent leaf senescence may influence their accumulation in relation to that of α‐tocopherol (vitamin E). RESULTS: Methanolic leaf extracts of 60 species of the genus Salvia were tested for the presence of carnosic acid and carnosol. These two diterpenes were detected in 48 and 27 species respectively. In contrast, α‐tocopherol was present in all species examined. Leaf senescence in Salvia officinalis resulted in α‐tocopherol increases by up to 5.5‐fold, while carnosic acid and carnosol increased by up to 18 and 290% respectively. Isorosmanol, an oxidation product of carnosic acid, increased by up to 2.1‐fold in senescing leaves. CONCLUSION: It was found that, while α‐tocopherol is ubiquitous in the genus Salvia, phenolic diterpenes are present in several but not all species. It was also shown that leaf senescence may increase the yield of both phenolic diterpenes and α‐tocopherol in sage extracts. Copyright © 2008 Society of Chemical Industry  相似文献

Background

The purpose of this study was to examine the efficacy of L-alanyl-L-glutamine (AG) ingestion on basketball performance, including jump power, reaction time, shooting accuracy and fatigue.

Methods

Ten women (21.2 ± 1.6 years; height: 177.8 ± 8.7 cm; body mass: 73.5 ± 8.0 kg), all scholarship NCAA Division I basketball players, volunteered for this study. Subjects participated in four trials, each consisting of a 40-min basketball game with controlled time-outs for rehydration. During the first trial (DHY) subjects were not allowed to rehydrate, and the total weight lost during the contest was used to determine fluid replenishment during the subsequent three trials. During one trial subjects consumed only water (W), while during the other two trials subjects consumed the AG supplement mixed in water using either a low dose (1 g per 500 ml) (AG1) or high dose (2 g per 500 ml) (AG2) concentration. All data assessed prior to and following each game were converted into a ?? score (Post results - Pre results). All performance data were then analyzed using a one-way repeated measures analysis of variance.

Results

During DHY subjects lost 1.72 ± 0.42 kg (2.3%) of their body mass. No differences in fluid intake (1.55 ± 0.43 L) were seen between rehydration trials. A 12.5% (p = 0.016) difference in basketball shooting performance was noted between DHY and AG1 and an 11.1% (p = 0.029) difference was seen between AG1 and W. Visual reaction time was significantly greater following AG1 (p = 0.014) compared to DHY. Differences (p = 0.045) in fatigue, as determined by player loads, were seen only between AG2 and DHY. No differences were seen in peak or mean vertical jump power during any trial.

Conclusion

Rehydration with AG appears to maintain basketball skill performance and visual reaction time to a greater extent than water only.  相似文献

Objective: The calibration of item banks provides the basis for computerized adaptive testing that ensures high diagnostic precision and minimizes participants’ test burden. The present study aimed at developing a new item bank that allows for assessing depression in persons with mental and persons with somatic diseases. Method: The sample consisted of 161 participants treated for a depressive syndrome, and 206 participants with somatic illnesses (103 cardiologic, 103 otorhinolaryngologic; overall mean age = 44.1 years, SD =14.0; 44.7% women) to allow for validation of the item bank in both groups. Persons answered a pool of 182 depression items on a 5-point Likert scale. Results: Evaluation of Rasch model fit (infit 2.0), and reliability (>.80) resulted in a bank of 79 items with good psychometric properties. Conclusions: The bank provides items with a wide range of content coverage and may serve as a sound basis for computerized adaptive testing applications. It might also be useful for researchers who wish to develop new fixed-length scales for the assessment of depression in specific rehabilitation settings.
