Extracellular vesicles (EVs) are a family of particles/vesicles present in blood and body fluids, composed of phospholipid bilayers that carry a variety of molecules that can mediate cell communication, modulating crucial cell processes such as homeostasis, induction/dampening of inflammation, and promotion of repair. Their existence, initially suspected in 1946 and confirmed in 1967, spurred a sharp increase in the number of scientific publications. Paradoxically, the increasing interest for EV content and function progressively reduced the relevance for a precise nomenclature in classifying EVs, therefore leading to a confusing scientific production. The aim of this review was to analyze the evolution of the progress in the knowledge and definition of EVs over the years, with an overview of the methodologies used for the identification of the vesicles, their cell of origin, and the detection of their cargo. The MISEV 2018 guidelines for the proper recognition nomenclature and ways to study EVs are summarized. The review finishes with a “more questions than answers” chapter, in which some of the problems we still face to fully understand the EV function and potential as a diagnostic and therapeutic tool are analyzed.  相似文献

We used stable water suspensions of copper oxide particles with mean diameter 20 nm and of particles containing copper oxide and element copper with mean diameter 340 nm to assess the pulmonary phagocytosis response of rats to a single intratracheal instillation of these suspensions using optical, transmission electron, and semi-contact atomic force microscopy and biochemical indices measured in the bronchoalveolar lavage fluid. Although both nano and submicron ultrafine particles were adversely bioactive, the former were found to be more toxic for lungs as compared with the latter while evoking more pronounced defense recruitment of alveolar macrophages and especially of neutrophil leukocytes and more active phagocytosis. Based on our results and literature data, we consider both copper solubilization and direct contact with cellular organelles (mainly, mitochondria) of persistent particles internalized by phagocytes as probable mechanisms of their cytotoxicity.  相似文献

Efficient yellow–green electroluminescence emission at λmax = 530 nm with CIE coordinates x = 0.3913, y = 0.4947 was obtained with organic light-emitting devices based on tris(2-methyl-8-quinolinolate) scandium (1). The device with the configuration of indium tin oxide/N,N′-bis(3-methylphenyl)-N,N′-diphenylbenzidine/1/Yb exhibits current efficiency of 3.1 cd/A and power efficiency of 1.8 lm/W at a luminance of 100 cd/m2. The DFT calculations demonstrate that structural changes of the scandium complex 1 influence the electroluminescence spectrum, the better agreement with experimental data being achieved when monodentate ligands are taken into consideration.  相似文献

A progestin receptor (PR) has been detected in the olfactory organ of the trout Salmo trutta fario. The specificity of this receptor was high for 17α,20β‐dihydroxy‐4‐pregnen‐3‐one (17α,20β‐DP), but it also bound 17α‐hydroxy‐progesterone (17α‐OHP) and 21‐hydroxyprogesterone (21‐OHP), even when present at low concentrations (10‐fold in relative binding affinity assay). Progesterone (P) competed effectively at much higher concentrations (1,000‐fold in relative binding affinity assay). Immunohistochemical studies carried out with three different monoclonal antibodies against human progesterone receptor (hPR), chicken progesterone receptor hinge region (cPR), and chicken progesterone receptor A/B domain (PR22), revealed that immunoreactivity was present in the epithelium of the olfactory organ of females and males of the trout Salmo trutta fario only against hPR. Western blotting showed two hPR immunoreactive bands of about 62 and 66 kDa. Finally, a portion of the cDNA of about 300 nucleotides extending over the DNA binding domain and the ligand binding domain was cloned and sequenced, revealing a high degree of sequence homology of the PR in Salmo trutta fario with the PR in other teleosts. The determination of additions of soybean proteins in commercial bakery products containing soybean–wheat and soybean–rice binary mixtures has been achieved in this work by high-performance liquid chromatography using a perfusive column. Soybean proteins were solubilized in a 25:75 acetonitrile–water mixture containing 0.3% (v/v) acetic acid by ultrasonication for 10 min and centrifugation for 5 min. Soybean proteins were separated from rice and wheat proteins in less than 4 min using a linear binary gradient of acetonitrile–water containing 0.3% (v/v) acetic acid as ion-pairing agent. The proposed method was proved to be specific and sensitive making possible the detection and the quantitation of additions of about 0.10% (w/w) and 0.33% (w/w), respectively, of soybean proteins in soybean–wheat and soybean–rice products (related to 1 g of initial product). Precision and recoveries observed were also acceptable. The method was applied to the determination of soybean proteins in different commercial bakery products.  相似文献

Anionic polymerization of ε‐caprolactam was performed in the presence of only 5 wt % of aromatic polyimides having five‐ and six‐membered imide cycles and bearing hexafluoroisopropylidene, ether, fluorene, siloxane, phthalide, or SO3H? groups, both in the polymer backbone and as side moieties. The synthesized new copolymers were fully characterized by IR spectroscopy, thermomechanical analysis, dilatometry, and X‐ray diffraction. Tough and frictional properties of the obtained copolymers were estimated as well. It was found that on the selection of polyimide activator it is possible to gain the desirable control over the polymer properties, namely the gel‐fraction content, phase composition, compression modulus, notched Izod impact strength, temperature of frictional contact, friction coefficient, etc. These investigations will facilitate the choice of the optimum macromolecular activator for the modification of both the synthetic method and the properties of the commercial polycaproamide. © 2009 Wiley Periodicals, Inc. Coastal populations with high seafood consumption in the Mediterranean have a significant exposure to dietary methylmercury, and areas where environmental mercury pollution is an issue due to industrial activities are of special concern. The study was undertaken with the aim of assessing methylmercury exposure through fish consumption in a community of north Morocco and characterizing the relevant health risk. Concentrations of total mercury were determined in human hair, a biomarker of methylmercury exposure, and in locally consumed fish by cold vapour atomic absorption spectrophotometry. Based on consumption frequencies reported by the 108 subjects included in the study the weekly intake of methylmercury was estimated and biomarker data were evaluated in relation to the estimated intake and the sociodemographic characteristics of the population. Multiple regression analysis was employed for the interpretation of hair mercury data in relation to fish consumption frequency, gender and the age of individuals. Mercury concentrations in hair ranged from 0.22 to 9.56 µg g-1 (geometric mean = 1.79 µg g-1) and were closely related to fish intake. Fisherman and their families consumed fish three to five times per week and were the most exposed population subgroup. A high proportion of women of child-bearing age (50%) had relatively high levels of mercury in their hair (3.08-7.88 µg g-1).  相似文献

The addition of foreign proteins (mainly soybean proteins and milk proteins) to heat-processed meat products is a common practice. This work approaches the determination of additions of soybean proteins in heat-processed meat products prepared with chicken meat, beef meat, and complex mixtures of meats from different species (chicken, pork, beef, and turkey) by perfusion reversed-phase high-performance liquid chromatography. The applied method was previously developed for the determination of soybean proteins in pork and turkey meat products but it has never been tested for the determination of soybean proteins in other heat-processed meat products containing other kinds of meats. This paper demonstrates the validity of this method for the detection of soybean proteins in heat-processed meat products containing different varieties of meats and even in the presence of other foreign proteins such as milk proteins. The specificity and existence of matrix interferences have been checked for these samples and accuracy has been evaluated by the comparison of the soybean protein contents determined by the proposed method and the official ELISA method.  相似文献
